Herein, core-sheath nanofibrous mats (NFs) consisting of biocompatible poly(ethylene oxide) (PEO, core) and poly(l-lactic acid) (PLLA, sheath) for drug delivery were fabricated via coaxial electrospinning strategy. The nontoxic layered silicate rectorite (REC) with 0.5-1 wt % amount was introduced in the sheath for sustained drug delivery. Layered REC could be intercalated with PLLA macromolecule chains, leading to the densified structure for loading and keeping doxorubicin hydrochloride (DOX) while reversibly capturing and releasing DOX to delay the drug migration due to its high cation activity. The addition of REC in NFs could delay the initial burst release of DOX and prolong the residence time from 12 to 96 h. Moreover, DOX-loaded core-sheath NFs had in vitro culture with strong antitumor activity, which was confirmed by cytotoxicity results and live and dead assay. HepG2 tumor-bearing xenograft further demonstrated the tumor-suppression effect and the excellent safety of the DOX-loaded core-sheath NFs in vivo. Our group has developed optogenetic inverter circuits that activate transcription using darkness, as well as amplifier circuits that reach high expression levels under limited light. However, because both types of circuits harness Gal4p and Gal80p from the galactose (GAL) regulon they cannot be used simultaneously. Here, we apply the Q System, a transcriptional activator/inhibitor system from Neurospora crassa, to build circuits in Saccharomyces cerevisiae that are inducible using quinic acid, darkness, or blue light. We develop light-repressed OptoQ-INVRT circuits that initiate darkness-triggered transcription within an hour of induction, as well as light-activated OptoQ-AMP circuits that achieve up to 39-fold induction. The Q System does not exhibit crosstalk with the GAL regulon, allowing coutilization of OptoQ-AMP circuits with previously developed OptoINVRT circuits. As a demonstration of practical applications in metabolic engineering, we show how simultaneous use of these circuits can be used to dynamically control both growth and production to improve acetoin production, as well as enable light-tunable co-production of geraniol and linalool, two terpenoids implicated in the hoppy flavor of beer. We report here that PLTs preserved under three-dimensional (3D) conditions using novel biomimetic nanofiber peptides showed reduced apoptosis compared with classical PLTs stored at 22 °C and facilitated the storage and transportation of PLTs. The mechanism of PLT 3D preservation involves the formation of cross-links and a 3D nanofibrous network by a self-assembled peptide scaffold material at physiological conditions after initiation by triggers in plasma. PLTs adhere to the surface of the nanofibrous network to facilitate the 3D distribution of PLTs. The 3D microstructure, rheological properties, and effect on the inflammatory response and hemolysis were evaluated. Compared to traditional PLTs stored at 22 °C, PLTs subjected to 3D preservation showed similar morphology, number, aggregation activity, and reduced apoptosis. The detection of the reactive oxygen species (ROS) levels demonstrated that both reduced intracellular and mitochondrial ROS levels were correlated with reduced apoptosis. Combining metal single atoms with CsPbX3 NCs may be a practical way in exploring perovskite-based catalysts. However, such hybrids have not been achieved experimentally yet, mainly due to the weak interaction between the metal atom and the CsPbX3 surface. Here, we demonstrate that Pt single atoms can be deposited on CsPbBr3 NCs through a photoassisted approach, in which the surface was partially oxidized first, followed by the anchoring of Pt single atoms through the formation of Pt-O and Pt-Br bonds. The deposition of Pt single atoms can significantly change the photophysical properties of CsPbBr3 NCs by inducing the generation of deep trap states in the band gap. The as-prepared Pt-SA/CsPbBr3 can be used as efficient and durable catalysts for photocatalytic semi-hydrogenation of propyne. PFASs often require an extraction pretreatment for enrichment before detection and analysis, which is still challenged by the relatively low efficiency because of the limited specific interactions involved. Here, we deliberately introduced multiple interactions into the solid-phase microextraction (SPME) process via a dual-functional modification of MIL-101(Cr), i.e., amination and subsequent fluorination, which is then used as an adsorbent for the efficient enrichment of PFASs. In combination with ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS), ultrasensitive quantitative analysis is available for nine selected PFASs with high linearities above 0.9941 in the ranges of 0.5-1500 ng/L, low limits of detection of 0.004-0.12 ng/L, satisfactory repeatability and reproducibility with a relative standard deviation (RSD) less then 11.6%, as well as excellent performance in complicated real water samples (recovery ratio of 76.2-108.6%). Abnormally high activity in the lateral habenula causes anxiety- or depression-like behaviours in animal experimental models. It has also been reported in humans that excessive stress in early life is correlated with the onset of psychiatric disorders in adults. These findings raise the question of whether maturation of the lateral habenula is affected under the influence of early-life experiences, which could govern behaviours throughout life.
We examined the maturation of the lateral habenula in mice based on neuronal activity markers and plastic components Zif268/Egr1, parvalbumin and perineuronal nets. We examined the effect of early-life stress using repeated maternal deprivation.
First, we found a transient highly sensitive period of the lateral habenula under stress. The lateral habenula matured through 4 stages postnatal days 1-9 (P1-9), P10-20, around P35 and after P35. At P10-20, the lateral habenula was highly sensitive to stress. We also observed experience-dependent maturation of the lateraicity between stages of lateral habenula maturation.
An increased neural response to making errors has emerged as a biomarker of anxiety. Error negativity (Ne) or errorrelated negativity (ERN) is an event-related potential generated when people commit errors; the Ne/ERN is greater among people with anxiety and predicts increases in anxiety. However, no previous study has examined whether the Ne/ERN can be used as a prognostic indicator among people with current anxiety. The present study addressed this gap by examining whether the Ne/ERN prospectively predicts increases in anxiety symptoms in clinically anxious children and adolescents.
The sample included 34 female participants between the ages of 8 and 14 years who met the criteria for a clinical anxiety disorder based on clinical interview. The Ne/ERN was measured using a flanker task.
Increased Ne/ERN at baseline predicted increases in total anxiety symptoms 2 years later, even when accounting for baseline symptoms. The Ne/ERN predicted increases in the symptom domains of generalized anxiety, social anxiety and harm avoidance/perfectionism, but not panic, separation anxiety, school avoidance or physical symptoms.
