Buccal tissues are considered one of the potential alternative delivery route because of fast drug absorption and onset of action due to high vascularization and a non-keratinized epithelial membrane. In this study, the effect of Penetratin on the permeation of salmon calcitonin (sCT), a model macromolecular peptide drug, through TR146 buccal cells and porcine buccal tissues has been evaluated. OSS_128167 To observe permeation profile of sCT, TR146 buccal cells were treated with Alexa 647 conjugated sCT (Alexa 647-sCT) with different concentrations of fluorescein isothiocyanate -labeled Penetratin (FITC-Penetratin) ranging from 0 to 40 μM, and analyzed using flow cytometry and confocal laser scanning microscopy. Intracellular penetration of FITC-Penetratin rapidly increased at low concentrations from 0 to 15 μM and it gradually increased at concentrations above 15 μM. Intracellular penetration of Alexa 647-sCT enhanced with the increase of FITC-Penetratin concentration. When TR146 cell layers and buccal tissues were co-treated with sCT and Penetratin as permeation enhancer, the flux of sCT increased as per Penetratin concentration. Compared to the control, 12.2 μM of Penetratin enhanced the flux of sCT in TR146 cell layers and buccal tissues by 5.5-fold and 93.7-fold, respectively. These results strongly suggest that Penetratin may successfully act as a non-invasive permeation enhancer for macromolecular peptide drug delivery through buccal routes.This study attempted to analyze and optimize the effect of an external magnetic field (EMF) on the aspect ratio and heat input for AZ31B weld joints that were welded using the gas metal arc welding (GMAW) process. The response surface methodology (RSM) was adopted for the critical analysis, and subsequently, mathematical models were developed based on the experimental results. It was observed that the EMF and its interaction with the wire feed rate significantly affected the aspect ratio and heat input, respectively. At 119 G (magnetic field), 700 mm/min (welding speed), 5.8 m/min feed rate, and 11.5 L/min (gas flow rate), the aspect ratio was 2.26, and the corresponding heat input factor (HIf) was 0.8 with almost full weld penetration.Gender-based violence (GBV) is a serious global public health problem that becomes aggravated during public health emergencies that involve quarantine measures. It is important to train nursing students on GBV, especially in vulnerable situations, such as the current COVID-19 pandemic. The purpose of our study is to explore the perceptions of third-year nursing students about simulated nursing video consultations for providing assistance to potential cases of GBV victims using a high-fidelity clinical simulation methodology. After all of the simulated scenarios were completed, 48 scripted interviews were carried out following a guide composed of four open-ended questions to facilitate in-depth discussion. A descriptive qualitative study based on the interpretative paradigm was conducted. The nursing students indicated that they improved their knowledge on GBV victim management (mainly their awareness of the problem, recognition of the role of nursing professionals, and performance of non-technical skills), although they also mentioned the need for continuous training (particularly in socio-emotional skills, interview techniques, a holistic nursing care approach, and not presupposing). This innovative high-fidelity simulation methodology allows nursing students to improve their awareness of the GBV problem, acquire a realistic view about their role in addressing GBV, and build their non-technical skills (such as active listening, communication skills, empathy, and generating confidence) required to adequately care for victims of GBV.Van-der Waals heterostructures assembled from one or few atomic layer thickness crystals are becoming increasingly more popular in condensed matter physics. These structures are assembled using transfer machines, those are based on mask aligners, probe stations or are home-made. For many laboratories it is vital to build a simple, convenient and universal transfer machine. In this paper we discuss the guiding principles for the design of such a machine, review the existing machines and demonstrate our own construction, that is powerful and fast-in-operation. Our results thus show that technology of the two-dimensional materials and heterostructures becomes accessible to anyone.The emerging, often multidrug-resistant Candida auris is increasingly being associated with outbreaks in healthcare facilities. Here we describe the molecular epidemiology of a C. auris outbreak during 18 months, which started in 2018 in the high dependency unit (HDU) of a secondary-care hospital in Kuwait. Demographic and clinical data for candidemia and colonized patients were prospectively recorded. Clinical and environmental isolates were subjected to phenotypic and molecular identification; antifungal susceptibility testing by broth microdilution method; PCR-sequencing of ERG11 and FKS1 for resistance mechanisms to triazoles and echinocandins, respectively; and molecular fingerprinting by short tandem repeat (STR) analyses. Seventy-one (17 candidemic and 54 colonized) patients including 26 with candiduria and seven environmental samples yielded C. auris. All isolates were identified as C. auris by Vitek2, MALDI-TOF MS, PCR amplification and/or PCR-sequencing of rDNA. Twelve candidemia and 26 colonized patients were admitted or exposed to HDU. Following outbreak recognition, an intensive screening program was instituted for new patients. Despite treatment of all candidemia and 36 colonized patients, 9 of 17 candidemia and 27 of 54 colonized patients died with an overall crude mortality rate of ~50%. Nearly all isolates were resistant to fluconazole and contained the Y132F mutation in ERG11 except one patient's isolates, which were also distinct by STR typing. Only urine isolates from two patients developed echinocandin resistance with concomitant FKS1 mutations. The transmission of C. auris in this outbreak was linked to infected/colonized patients and the hospital environment. However, despite continuous surveillance and enforcement of infection control measures, sporadic new cases continued to occur, challenging the containment efforts.On-site manual inspection of metro tunnel leakages has been faced with the problems of low efficiency and poor accuracy. An automated, high-precision, and robust water leakage inspection method is vital to improve the manual approach. Existing approaches cannot provide the leakage location due to the lack of spatial information. Therefore, an integrated deep learning method of water leakage inspection using tunnel lining point cloud data from mobile laser scanning is presented in this paper. It is composed of three parts as follows (1) establishment of the water leakage dataset using the acquired point clouds of tunnel linings; (2) automated leakage detection via a mask-region-based convolutional neural network; and (3) visualization and quantitative evaluation of the water leakage in 3D space via a novel triangle mesh method. The testing result reveals that the proposed method achieves automated detection and evaluation of tunnel lining water leakages in 3D space, which provides the inspectors with an intuitive overall 3D view of the detected water leakages and the leakage information (area, location, lining segments, etc.).Lactoferrin (Lf) and other whey proteins have been isolated from red deer milk for the first time using a three-step anion and cation exchange chromatography protocol. The separated deer Lf was subject to in vitro gastric and duodenal digestions to generate peptides. The purity of the deer Lf and its hydrolysis products were analyzed by SDS-PAGE. The antibacterial activity of the deer Lf and its hydrolysates were investigated and was compared to cow counterpart. Gastric and duodenal digested deer Lf had strong bactericidal activity against E. coli ATCC 25922 with minimum inhibition concentration (MIC) of 280 µM and 402 µM, respectively. These results suggest that deer milk contains bioactive whey proteins and can generate bioactive peptides, which can benefit human health by inhibiting food-borne pathogenic bacteria.Purpose Fewer studies are about the influence of psychopath traits on moral judgment and the underlying psychological mechanism in Chinese cultural background. In this paper, we use the creative CNI (Consequences, Norms, Inaction versus action) model to quantify the subject's reaction to moral dilemmas. Method In this research, the Chinese version of the Levenson Psychopathic Scale, CNI model materials, and a multinomial model to further analyze the associations among the psychopathy characteristics and utilitarian moral judgment are applied. The CNI model is proposed by Gawronski et al., which can quantify the subjects' sensitivity to moral consequence, sensitivity to moral norms, and the general preference for inaction or action in moral dilemmas. Result This study finds that there were significant differences in the utilitarian moral judgment between the groups, t (360) = 3.24, p = 0.001, and Cohen's d = 0.36. The analysis results of the CNI model show that the high psychopathy group on the N parameter was significantly lower than the group of low psychopathy, ΔG2 (2) = 79.70, p = 0.001. In terms of the C parameter, we found no significant distinctions between the two groups, ΔG2 (2) = 1.356, p = 0.244. For the I parameter, the two groups also have no significant differences, ΔG2 (2) = 0.093, p = 0.76. Conclusion Persons with high psychopathy traits prefer to make more utilitarian moral judgments and have a weak sensitivity to moral norms (N). The sensitivity to consequences (C) of the two groups is no significant difference. The general preference for inaction versus action (I) also has no significant differences between those two groups. Moreover, the CNI model fits well in Chinese subjects.Through considering both nonlocality and surface energy effects, this paper suggests suitable mathematical-continuum-based models for free vibration of nanorods with multiple defects acted upon by a bidirectional-transverse magnetic field. By employing both theories of elasticity of Eringen and Gurtin-Murdoch, the equations of motion for the magnetically affected-damaged rod-like nanostructures are derived using the nonlocal-differential-based and the nonlocal-integral-based models. The local defects are modeled by a set of linearly appropriate axial springs at the interface of appropriately divided nanorods. Through constructing the nonlocal-differential equations of motion for sub-divided portions and by imposing the appropriate interface conditions, the natural frequencies as well as the vibrational modes are explicitly obtained for fixed-free and fixed-fixed nanorods with low numbers of defects. The extracted nonlocal-integral governing equations are also solved for natural frequencies using the finite-element technique.
