Glycogen synthase kinase 3 (GSK3) is a serine/threonine kinase which in the presence of ATP in its ATP-binding pocket transfers a phosphate to a primed substrate. GSK3β is an isoform of GSK3 which has been projected as a potent therapeutic target in human diseases including cancers and metabolic syndrome. Incidentally, cardiovascular disease is a common cause of non-cancer related deaths in prostate cancer (PCa) patients, mainly due to the effects of androgen-deprivation therapy (ADT), a mainstay for PCa treatment. Several small molecular inhibitors of GSK3 are either ATP-competitive (bind to the ATP-binding pocket), or non-ATP-competitive inhibitors (binding to the substrate-binding site of the enzyme). In this study, 2-β-D-glucopyranosyl-1,3,6,7-tetrahydroxy-9H-xanthen-9-one (βDGT), a natural xanthonoid present in many plant species, is reported to bind to the ATP-binding pocket of GSK3β and inhibit its activity, as demonstrated by the molecular docking and molecular dynamics simulation analysis and experim natural ATP-competitive inhibitor of GSK3β and implicates its use in prostate cancer patients on androgen-deprivation therapy, a cardiovascular disease risk, and other pathological conditions.Communicated by Ramaswamy H. Sarma.Breast cancer is a severe problem for women around the world especially in developing countries, according to recent reports from the World Health Organization (WHO). High accuracy and early detection of breast cancer reduces the mortality rate, in the other hand, recognition of breast cancer is a complicated issue. Various studies and methods have been carried out to overcome this problem and to obtain accurate screening of breast cancer. One of the most recent methods with high performance is deep learning; it has been used to classify breast cancer using mammograms or histopathological images. This paper proposes a new using the concept of sliding window, and using the ensemble of four pre-trained convolutional neural networks (CNN) in order to classify breast cancer into eight classes. Breast Cancer Histopathological Database (BreakHis) database has been employed in this paper with eight classes (Adenosis, Ductal Carcinoma, Fibroadenoma, Lobular Carcinoma, Mucinous Carcinoma Papillary Carcinoma, Phyllodes Tumour, Tubular Adenoma). The proposed method is applied to four magnification cases 40x, 100x, 200x, and 400x images. The proposed ensemble technique achieved an accuracy of 99.3325%. The results of the proposed system are comparable to recent studies results.
Depression and cardiovascular disease (CVD) are major causes of global disease burden that are interrelated through mostly unknown mechanisms. We studied the relationship of melancholic and non-melancholic depressive symptoms with arterial stiffness, an important underlying mechanism of CVD.
The Helsinki Birth Cohort Study recruited 683 previously extensively phenotyped subjects for this sub-study. Cross-sectional data along with responses regarding depressive symptoms were obtained for each participant. For evaluation of depressive symptoms, the Beck Depression Inventory (BDI)and subscales were used to measure melancholic and non-melancholic depressive symptoms. Arterial stiffness was assessed as pulse wave velocity (PWV) that was measured between the carotid and radial artery, and carotid and femoral artery.
Of the participants, 532 scored <10 on the BDI and were classified as not having depressive symptoms. Of the 151 participants that scored ≥10 on the BDI, 122 were classified as having non-melandial pulse wave velocity in men.Non-melancholic depressive symptoms are associated with higher carotid-radial pulse wave velocity in men.
Arterial stiffness has a relationship with depressive symptoms and subtypes of depressive symptoms, at least in men. There is a significant relationship between higher PWV and non-melancholic depressive symptoms in men. Due to the intricate nature of the disease causality or directionality is impossible to infer solely based on this study. Further studies into the subtypes of depressive symptoms may be of benefit to understanding depression.KEY MESSAGESIt is known that arterial stiffness contributes to cardiovascular disease, and is associated with depression.Higher Beck Depression Inventory scores are associated with higher carotid-radial pulse wave velocity in men.Non-melancholic depressive symptoms are associated with higher carotid-radial pulse wave velocity in men.

There are many lateral bending fracture cases presented in the literature that mostly involve facet dislocations or corpus collapse. In this report, we aim to describe a novel asymmetric lateral bending, flexion and distraction fracture, propose a mechanism, and delineate its clinical importance.
A 13-year-old girl arrived at our trauma center 12 hours after a head-on truck collision. She had paraplegia, and her imaging revealed a spinal cord avulsion at the T10 level and a horizontal fracture at the L4 spinous process, left lamina, left pedicle, and left posterior-upper corner of the corpus, extending through the right lamina. Her posterior ligamentous complex, right facet joint, pedicle, and right side of the corpus were spared from the injury. The patient stated that she had been sitting on the right side of the back seat, turned toward her cousin in the middle. Her left leg was externally rotated and flexed on the seat while her right foot was on the floor at the time of the accident. The patient was managed conservatively with an orthosis.
