Introduction This study aims at identifying mechanical characteristics under bi-axial loading conditions of extracted swine pia mater (PM) and dura and arachnoid complex (DAC). Rapamycin Methods 59 porcine spinal samples have been tested on a bi-axial experimental device with a pre-load of 0.01 N and a displacement rate of 0.05 mm·s-1. Post-processing analysis included an elastic modulus, as well as constitutive model identification for Ogden model, reduced Gasser Ogden Holzapfel (GOH) model, anisotropic GOH model, transverse isotropic and anisotropic Gasser models as well as a Mooney-Rivlin model including fiber strengthening for PM. Additionally, micro-structure of the tissue was investigated using a bi-photon microscopy. Results Linear elastic moduli of 108 ± 40 MPa were found for DAC longitudinal direction, 53 ± 32 MPa for DAC circumferential direction, with a significant difference between directions (p less then 0.001). PM presented significantly higher longitudinal than circumferential elastic moduli (26 ± 13 on microscopy confirmed the use of anisotropic Gasser model for pia mater and existence of fenestration. While clear identification of fibre population was not possible in DAC, results from anisotropic Gasser model depicted better fitting on experimental data as per this protocol. Bi-axial mechanical testing allows quasi-static characterization under conditions closer to the physiological context and the results presented could be used for further simulations of physiology. Indeed, the inclusion of meningeal tissue in finite element models will allow more accurate and reliable numerical simulations.The artificial intelligence (AI) revolution is well underway, including in the medical field, and has dramatically transformed our lives. An understanding of the basics of AI applications, their development, and challenges to their clinical implementation is important for clinicians to fully appreciate the possibilities of AI. Such a foundation would ensure that clinicians have a good grasp and realistic expectations for AI in medicine and prevent discrepancies between the promised and real-world impact. When quantifying the track record for AI applications in cardiology, we found that a substantial number of AI systems are never deployed in clinical practice, although there certainly are many success stories. Successful implementations shared the following they came from clinical areas where large amount of training data was available; were deployable into a single diagnostic modality; prediction models generally had high performance in external validation; and most were developed as part of collaborations with medical device manufacturers who had substantial experience with implementation of new clinical technology. When looking into the current processes used for developing AI-based systems, we suggest that expanding the analytic framework to address potential deployment and implementation issues at project outset will improve the rate of successful implementation, and will be a necessary next step for AI to achieve its full potential in cardiovascular medicine.

To measure the choroidal structural parameters in a population of myopic children and determine the factors effecting the choroidal vascularity index (CVI).

In total, 200 eyes of 200 children (100 females, 100 males) with a mean age of 11.5±1.6 years were included in the study. Macular imaging was performed using EDI mode of spectral domain-optical coherence tomography. Binarization of the choroidal area was performed with ImageJ software. Total choroidal area, luminal area (LA), stromal area (SA), and CVI were automatically calculated.

The mean choroidal, stromal and luminal areas were measured as 0.952 ± 0.127 mm

, 0.626 ± 0.103 mm

and 0.325 ± 0.076 mm

, respectively. The mean CVI was 65.81% ± 6.56. Age and the axial length (AL) of the participants were not found to be associated with the LA and the CVI. (r=-0.078, p=0.274, r=0.017, p=0.808, and r=0,051, p=0.474 and r=-0.128, p=0.071, respectively). There was a statistically significant strong association between the LA and CVI measurements and SE of the participants (r=0.736, p=0.001, and r=-0.605; p=0.001).

Photodynamic therapy with photosensitizers can reduce the microbial load. However, few studies have evaluated the effects of photosensitizers on the color stability of endodontically treated intraradicular dentin. This in vitro study investigated the effects of methylene blue and curcumin photosensitizers used in photodynamic therapy on the color stability of intraradicular dentin.

Sixty human incisors were divided into three experimental groups according to the photosensitizer solutions used and their concentrations methylene blue (25mg/L), curcumin (1000mg/L), and curcumin (1500mg/L). The color stability of endodontically treated intraradicular dentin was evaluated using a portable reflectance spectrophotometer before and after the samples been storage in 2mL of the photosensitizer solutions during 5min (n=20). Color stability data were subjected to a normality test, and statistical analysis was performed using the one-way analysis of variance and Tukey least significant difference test (α=0.05).

Sampshowed the highest level of color alteration.This study examined the effect of various root canal irrigants and medicaments on dentin fluorescence elicited by 655 nm visible red laser light. To replicate clinical use, irrigants were applied onto dentin samples for 2 min, while medicaments were applied for 2 weeks. Fluorescence values tracked from baseline across the following to 24 h, starting 5 min after exposure. Sodium hypochlorite, hydrogen peroxide, and articaine local anaesthetic (4% articaine with 11000,000 adrenaline) all significantly quenched fluorescence (p less then 0.0001), which then returned to baseline levels after 20 min. Conversely, elevated fluorescence readings were recorded after 3% mepivacaine (p less then 0.05), 0.2% chlorhexidine (p less then 0.01) and chloroform (p less then 0.05). A 2 week application of Ledermix™ paste containing 3% demeclocycline caused an irreversible increase in fluorescence (p less then 0.0001). Other tested endodontic materials (15% EDTA, eucalyptus oil, calcium hydroxide, Odontopaste™ clindamycin paste, and distilled water) had no impact on dentine fluorescence. The influences of endodontic materials on dentin fluorescence need to considered when using fluorescence endpoints to guide the progress of root canal treatment.Osteoporosis (OP) is characterized by decreased bone mineral density and impaired bone strength. Carfilzomib (CFZ) is a new-generation proteasome inhibitor and has been found to affect bone metabolism. However, the effect and mechanism of CFZ on OP has not been investigated systematically. In this study, we found that protein levels of proteasome activator subunit 1/2 (PSME1/2) increased in OP, and accumulated mostly in osteoblasts and osteoclasts. Treatment with PSME1/2 recombinant protein inhibited osteogenesis and promoted osteoclast formation in vitro. Also, PSME1/2 inhibited the expression of β-catenin protein, resulting in limitation of Wnt/β-catenin signaling. CFZ inhibited PSME1 and PSME2 proteasome activities and increased β-catenin protein level, resulting in the translocation of β-catenin to the nucleus and activation of canonical Wnt/β-catenin signaling, further promoting osteogenesis and inhibiting osteoclastic differentiation. In vivo, we conducted ovariectomy (OVX) to create a model of OVX-induced postmenopausal OP in mice. When analyzed by micro-CT scanning, enhancement of bone mineral density, bone volume, trabecular number, and thickness was seen in the CFZ-treated mice. Also, we noticed increased osteogenesis and decreased osteoclastogenesis, diminished expression of PSME1 and PSME2 and activated Wnt/β-catenin signaling in bone sections from OP mice treated with CFZ. Overall, our data indicated that PSME1/2 may serve as new targets for the treatment of OP, and targeting PSME1/2 with CFZ provides a candidate therapeutic molecule for postmenopausal OP.Traditional Chinese medicine (TCM) has been long time used in China and gains ever-increasing worldwide acceptance. Er Miao San (EMS), a TCM formula, has been extensively used to treat inflammatory diseases, while its bioactive components and therapeutic mechanisms remain unclear. In this study, we conducted an integrative approach of network pharmacology and experimental study to elucidate the underlying mechanisms of EMS in treating human rheumatoid arthritis (RA) and other inflammatory conditions. Quercetin, wogonin and rutaecarpine were probably the main active compounds of EMS in RA treatment as they affected the most RA-related targets, and TNF-α, IL-6 and IL-1β were considered to be the core target proteins. The main compounds in EMS bound to these core proteins, which was further confirmed by molecular docking and bio-layer interferometry (BLI) analysis. Moreover, the potential molecular mechanisms of EMS predicted from network pharmacology analysis, were validated in vivo and in vitro experiments. EMS was found to inhibit the production of NO, TNF-α and IL-6 in lipopolysaccharide (LPS)-stimulated RAW264.7 cells; reduce xylene-induced mouse ear edema; and decrease the incidence of carrageenan-induced rat paw edema. The carrageenan-induced up-regulation of TNF-α, IL-6 and IL-1β mRNA expression in rat paws was down-regulated by EMS, consistent with the network pharmacology results. This study provides evidence that EMS plays a critical role in anti-inflammation via suppressing inflammatory cytokines, indicating that EMS is a candidate herbal drug for further investigation in treating inflammatory and arthritic conditions.Alzheimer's disease (AD) is a common neurodegenerative disease that mainly affects elderly people. However, the translational research of AD is frustrating, suggesting that the development of new AD animal models is crucial. By gavage administration of acrolein, we constructed a simple sporadic AD animal model which showed classic pathologies of AD in 1 month. The AD-like phenotypes and pathological changes were as followed. 1) olfactory dysfunctions, cognitive impairments and psychological symptoms in C57BL/6 mice; 2) increased levels of Aβ1-42 and Tau phosphorylation (S396/T231) in cortex and hippocampus; 3) astrocytes and microglia proliferation; 4) reduced levels of postsynaptic density 95(PSD95) and Synapsin1, as well as the density of dendritic spines in the CA1 and DG neurons of the hippocampus; 5) high-frequency stimulation failed to induce the long-term potentiation (LTP) in the hippocampus after exposure to acrolein for 4 weeks; 6) decreased blood oxygen level-dependent (BOLD) signal in the olfactory bulb and induced high T2 signals in the hippocampus, which matched to the clinical observation in the brain of AD patients, and 7) activated RhoA/ROCK2/ p-cofilin-associated pathway in hippocampus of acrolein-treated mice, which may be the causes of synaptic damage and neuroinflammation in acrolein mice model.
