Results Nine hundred and six patients were included for analysis. Higher preoperative NLR was significantly associated with postoperative atrial fibrillation. Day 1 and day 2 NLR were associated with postoperative atrial fibrillation in analyses including all patients. Older age, male gender, preexisting atrial arrhythmias, and higher EuroSCORE II also had a significant association. Diabetes mellitus was protective for postoperative arrhythmias. Preoperative NLR was not significantly associated with acute kidney injury. Day 2 NLR, older age, higher EuroSCORE II, and longer CPB time were independently associated with acute kidney injury post cardiac surgery. This study aims to investigate the effect of miR-1245a on the process of CA cells and its target gene BRCA2. Methods The expression of CA tissues and cells were evaluated by q RT-PCR. Then we explore the association between expression of miR-1245a and prognosis in the CA patients from the TCGA database. CCK8 assays, colony formation assays were performed to explore the effect of miR-1245a in CA cell proliferation. The invasion ability of CA cells was evaluated by Transwell assays. Western blot was performed to assess the BRCA2 expression. Luciferase reporter assay was employed to scrutinize the relationship between miR-1245a and BRCA2. Finally, rescue experiments were performed through BRCA2 downregulation and miR-1245a inhibitors by using colony formation assay and Transwell invasion assay. Results miR-1245a is upregulated in CA cells and tissues. Additionally, the high expression of miR-1245a was related to poor survival. CCK8 assays, colony formation assays and Transwell assays showed that miR-1245a promotes the proliferation and invasion of CA cells. The luciferase reporter assay indicated that miR-1245a targeted BRCA2 and inhibited its expression. The rescue experiment further showed that miR-1245a could restore the effect of BRCA2 on CA. Conclusions miR-1245a promotes the proliferation and invasion of CA by targeting BRCA2.Our results suggested that miR-1245a could be a potential biomarker for CA progression. 2019 Annals of Translational Medicine. All rights reserved.Background We aimed to characterize the relationships of lymphocyte activation gene-3 (LAG-3) expression, cytotoxic T-lymphocyte-associated antigen-4 (CTLA-4) expression, and CD8+ tumor-infiltrating lymphocyte (TIL) density, and to investigate the joint prognostic impact of these three markers in patients with surgically resected esophageal squamous cell carcinoma (ESCC). Methods Expression of LAG-3, CTLA-4 and the density of CD8+ TILs were evaluated by immunohistochemistry in resected ESCC. The associations between LAG-3 expression and clinicopathologic characteristics, as well as patient prognoses, were analyzed. Results A total of 183 patients were included. LAG-3 expression was observed in 69 (37.7%) patients. Positive LAG-3 expression was significantly associated with CTLA-4 expression (P=0.004). LAG-3 positivity, CTLA-4 positivity, and low CD8+ TIL densities were significantly associated with worsening recurrence-free survival (RFS) [LAG-3 hazard ratio (HR), 1.72; 95% confidence interval (CI), 1.10-2.89; P=0.019; CTLA-4 HR, 1.69; 95% CI, 1.04-2.73; P=0.033; CD8+ HR, 0.60; 95% CI, 0.38-0.94; P=0.025] and overall survival (OS) (LAG-3 HR, 2.09; 95% CI, 1.24-3.53; P=0.006; CTLA-4 HR, 1.47; 95% CI, 0.86-2.53; P=0.161; CD8+ HR, 0.56; 95% CI, 0.33-0.95; P=0.032). Subgroup analysis revealed that the LAG-3 CTLA-4 CD8+ group had the best RFS (P less then 0.001) and OS (P less then 0.001). Conclusions LAG-3 expression was correlated with CTLA-4 expression on TILs. Positive LAG-3 expression was associated with poor prognoses in ESCC. All rights reserved.Background We aimed to assess the prognostic value of the skeletal muscle index (SMI) and monocyte-to-lymphocyte ratio (MLR) in lymph node-positive breast cancer patients after mastectomy. Methods We enrolled female lymph node-positive breast cancer patients who had undergone mastectomy between January 2011 and December 2013 with lymph node metastasis. Skeletal muscle tissue was measured using computed tomography (CT), and the patients were grouped based on the receiver operating characteristic curves to obtain the cut-off point for SMI; similarly, the optimal cutoff point for the MLR was obtained. Survival analysis was chiefly performed to determine overall survival (OS) among the patients. Results The median age of the 97 included patients was 46 years (range, 27-73 years), whereas the median follow-up duration was 62.5 months. Of these patients, 71 exhibited low SMI and 66 exhibited high MLR. Kaplan-Meier curves indicated that low SMI (5-year OS, 97.2% vs. 84.6%; log-rank P=0.021) and low MLR (5-year OS, 98.5% vs. 83.9%; log-rank P=0.004) were associated with better OS. Moreover, patients with both high SMI and MLR (high SMLR) had significantly worse OS (5-year OS, 66.7% vs. 96.6%; log-rank P less then 0.001), relative to the low SMLR group. Multivariate analysis indicated that patients with low SMI had a lower overall dying risk, relative to those with high SMI [hazard ration (HR), 0.188; P=0.038], whereas patients with high MLR had a higher risk of death as compared to those with low MLR (HR, 7.152; P=0.021). Furthermore, SMLR was an independent prognostic factor of poor OS (HR, 13.272; P=0.001). Conclusions Low SMI and low MLR are both associated with better OS in lymph node-positive breast cancer patients after mastectomy. Methods The study used a total of 12 domestic pigs, 6 animals underwent resection with inhaled hydrogen during general anesthesia, and 6 animals underwent the same procedure using conventional, unsupplemented, general anesthesia. Intraoperative preparation of the left branch of the hepatic portal vein and the left hepatic artery was performed, and a tourniquet was applied. Warm ischemia was induced for 120 minutes and then followed by liver reperfusion for another 120 minutes. Samples from the ischemic and non-ischemic halves of the liver were then removed for histological and biochemical examinations.
