The most distinctive feature of halophilic from non-halophilic enzymes are the higher content of acidic residues (Asp and Glu), and a lower content of lysine, aliphatic hydrophobic (Leu, Met and Ile) and polar (Asn) residues. The amino acid composition and 3-D structure analysis suggested that the high content of acidic residues (Asp and Glu, 12.2%) and low content of lysine residues (0.7%), as well as the excess of surface-exposed acidic residues might be responsible for the haloadaptation of Est56.Nano-drugs based on nanoparticles (NP) or on nano-assemblies as carriers of the active pharmaceutical ingredient (API) are often expected to perform better compared to conventional dosage forms. Maximum realization of this potential though requires optimization of multiple physico-chemical, including structural and morphological, parameters. Meaningful distributions of these parameters derived from sufficient populations of individual NPs rather than ensemble distributions are desirable for this task, provided that relevant high-resolution data is available. In this study we demonstrate powerful capabilities of the up-to-date cryogenic transmission electron-microscopy (cryo-TEM) as well as correlations with other techniques abundant in the nano-research milieu. We explored Doxil®-like (an anticancer drug and the first FDA-approved nano-drug) (75-100 nm) PEGylated liposomes encapsulating single doxorubicin-sulfate nano-rod-crystals (PLD). These crystals induce liposome sphere-to-ellipsoid deformation. Doxil® was characterized by a multitude of physicochemical methods. We demonstrate, that accompanied by advanced image-analysis means, cryo-TEM can successfully enable the determination of multiple structural parameters of such complex liposomal nano-drugs with an added value of statistically-sound distributions. The latter could not be achieved by most other physicochemical approaches. It seems that cryo-TEM is capable of quantitative description of individual liposome morphological features, including meaningful distributions of all structural elements, with averages that correlate with other physical methods. Here it is demonstrated that such quantitative cryo-TEM analysis is a powerful tool in determining what is the optimal drug to lipid ratio in PLD, which is found to be the drug to lipid ratio existing in Doxil®.(1) Background The purpose of this meta-analysis was to investigate the effects of combined exercise and low carbohydrate ketogenic diet interventions (CELCKD) for overweight and obese individuals. (2) Methods Relevant studies were searched by using the MEDLINE and EMBASE databases up to October 2020. Study Inclusion and Exclusion Criteria Inclusion criteria were reporting effects of the CELCKD for overweight and obese individuals from randomized controlled trials. Studies that did not match the inclusion criteria were excluded. Iruplinalkib cost The methods for CELCKD and outcomes of selected studies were extracted. The effect sizes for interventions that included cardiorespiratory fitness, body composition, fasting glucose, and lipid profiles were calculated by using the standardized mean difference statistic. (3) Results A total of seven studies and 278 overweight and obese individuals were included. The average intervention of selected studies consisted of moderate to vigorous intensity, 4 times per week for 9.2 weeks. Participating in CELCKD interventions was decreased triglycerides (d = -0.34, CI; -0.68--0.01, p = 0.04) and waist circumference (d = -0.74, 95% confidence interval [CI]; -1.28--1.20, p = 0.01), while cardiovascular fitness, body composition, fasting glucose, total cholesterol, high density lipoprotein (HDL) cholesterol, and low density lipoprotein (LDL) cholesterol were not statistically different after the interventions. No adverse side effects were reported. (4) Conclusions Participation in interventions by overweight and obese individuals had beneficial effects including decreased waist circumference and triglycerides. Longer term intervention studies with homogenous control groups may be needed.The most important biological function of vitamin B12 is to accomplish DNA synthesis, which is necessary for cell division. Cobalamin deficiency may be especially acute for rapidly dividing cells, such as glioblastoma cells. Therefore, cobalamin antagonists offer a medicinal potential for developing anti-glioma agents. In the present study, we developed an in vitro model of cobalamin deficiency in glioblastoma cells. Long-term treatment of cells with the cobalamin analogue, hydroxycobalamin [c-lactam] (HCCL) was applied to induce an increase of hypocobalaminemia biomarker. Cytometric assays demonstrated that vitamin B12 promoted glioblastoma cells proliferation, whereas the treatment of cells with HCCL caused a dramatic inhibition of cell proliferation and an induction of cell cycle arrest at the G2/M phase. Vitamin B12 counteracted all the observed effects of HCCL. In the in silico study, we characterized the molecular interactions between HCCL and transcobalamin II (TCII). We have demonstrated that HCCL shares similar interactions with TCII as naturally occurring cobalamins and therefore may act as a competitive inhibitor of this key transporter protein. We assessed the impact of HCCL on the mortality or developmental malformations of zebrafish embryos. Collectively, our findings suggest that the use of cobalamin transport antagonists as potential anti-glioma agents would be worth exploring further.Recently, there has been a resurgence of formal language theory in deep learning research. However, most research focused on the more practical problems of attempting to represent symbolic knowledge by machine learning. In contrast, there has been limited research on exploring the fundamental connection between them. To obtain a better understanding of the internal structures of regular grammars and their corresponding complexity, we focus on categorizing regular grammars by using both theoretical analysis and empirical evidence. Specifically, motivated by the concentric ring representation, we relaxed the original order information and introduced an entropy metric for describing the complexity of different regular grammars. Based on the entropy metric, we categorized regular grammars into three disjoint subclasses the polynomial, exponential and proportional classes. In addition, several classification theorems are provided for different representations of regular grammars. Our analysis was validated by examining the process of learning grammars with multiple recurrent neural networks.
