Improving plant water-use efficiency (WUE) is important to plant survival and crop yield in the context of water limitation. In this study, SlTLFP8 (Tubby-like F-box protein 8) was identified as an osmotic-induced gene in tomato. Transgenic tomato with up-regulated expression of SlTLFP8 showed enhanced water-deficient resistance, whereas knockout mutants generated by CRISPR/Cas9 were more sensitive to water deficit. SlTLFP8 overexpression significantly enhanced WUE by suppressing transpiration under both water-sufficient and water-deficient conditions. Further study showed that overexpressing SlTLFP8 significantly increased leaf epidermal cell size and thereby decreased stomatal density 10-20%, conversely SlTLFP8 knockout resulted in decreased cell size and thereby increased stomatal density 20-50%. SlTLFP8 overexpression and knockout modulated ploidy levels in leaf cells. Changes in expression of cell cycle related genes also indicated that SlTLFP8 affected cell size and stomatal density through endocycle tritionally, overexpressing SlTLFP8 significantly decreased stomatal density, while SlTLFP8 knockout resulted in increased stomatal density, and SlTLFP8 affected stomatal density through endoreduplication and altered epidermal cell size. Despite changes in stomata density, altering the expression of SlTLFP8 did not result in distinct changes in photosynthesis, biomass and yield of tomato.Understanding the role of species interactions within communities is a central focus of ecology. A key challenge is to understand variation in species interactions along environmental gradients. The stress gradient hypothesis posits that positive interactions increase and competitive interactions decrease with increasing consumer pressure or environmental stress. This hypothesis has received extensive attention in plant community ecology, but only a handful of tests in animals. Furthermore, few empirical studies have examined multiple co-occurring stressors. Here we test predictions of the stress gradient hypothesis using the occurrence of mixed-species groups in six common grazing ungulate species within the Serengeti-Mara ecosystem. We use mixed-species groups as a proxy for potential positive interactions because they may enhance protection from predators or increase access to high-quality forage. Alternatively, competition for resources may limit the formation of mixed-species groups. Using more than 115,000 camera trap observations collected over 5 yr, we found that mixed-species groups were more likely to occur in risky areas (i.e., areas closer to lion vantage points and in woodland habitat where lions hunt preferentially) and during time periods when resource levels were high. These results are consistent with the interpretation that stress from high predation risk may contribute to the formation of mixed-species groups, but that competition for resources may prevent their formation when food availability is low. Here, we provide evidence for the existence of a LONG HYPOCOTYL 5 (HY5)-MYB15-CBFs transcriptional cascade that plays a crucial role in the cold response in tomato. The exposure of tomato plants to cold (4°C) increased the levels of HY5, MYB15 and CBFs transcripts. Moreover, mutations in HY5 or MYB15 decreased the levels of CBF transcripts. In contrast, overexpression of HY5 or MYB15 increased CBF transcript abundance. Crucially, the HY5 transcription factor activated the expression of MYB15 by directly binding to the promoter region, while both HY5 and MYB15 activated the expression of CBF1, CBF2 and CBF3. Taken together, these data show that HY5 can directly regulate CBF transcript levels, and also influence CBF expression indirectly via MYB15. The coordinated action of HY5 and MYB15 allows precise regulation of CBF expression and subsequent cold tolerance. These findings provide an improved understanding of the molecular mechanisms affording transcriptional regulation of CBFs, which can be exploited in the future to enhance cold tolerance in crops.
