Other strains belonged to the genotype 1A (3%). We sequenced 39 strains from nine other countries, which, together with previously published genomes, amounted to 242 genomes that were used for molecular dating. Specific SNP markers for the new 1D-Malagasy genotype were used to screen samples from 11 countries and revealed this genotype to be restricted to Madagascar, with the sole exception being a strain from Malawi. The overall analysis thus ruled out a possible introduction of leprosy by the Austronesian settlers and suggests a later origin from East Africa, the Middle East, or South Asia. The fate of future food productivity depends primarily upon the health of soil used for cultivation. For Atlantic Europe, increased precipitation is predicted during both winter and summer months. Interactions between climate change and the fertilization of land used for agriculture are therefore vital to understand. This is particularly relevant for inorganic phosphorus (P) fertilization, which already suffers from resource and sustainability issues. The soil microbiota are a key indicator of soil health and their functioning is critical to plant productivity, playing an important role in nutrient acquisition, particularly when plant available nutrients are limited. A multifactorial, mesocosm study was established to assess the effects of increased soil water availability and inorganic P fertilization, on spring wheat biomass, soil enzymatic activity (dehydrogenase and acid phosphomonoesterase) and soil bacterial community assemblages. buy RBPJ Inhibitor-1 Our results highlight the significance of the spring wheat rhizosphere in shaping soil bacterial community assemblages and specific taxa under a moderate soil water content (60%), which was diminished under a higher level of soil water availability (80%). In addition, an interaction between soil water availability and plant presence overrode a long-term bacterial sensitivity to inorganic P fertilization. Together this may have implications for developing sustainable P mobilization through the use of the soil microbiota in future. Spring wheat biomass grown under the higher soil water regime (80%) was reduced compared to the constant water regime (60%) and a reduction in yield could be exacerbated in the future when grown in cultivated soil that have been fertilized with inorganic P. The potential feedback mechanisms for this need now need exploration to understand how future management of crop productivity may be impacted.Colorants contained in palm oil mill effluent (POME) are recalcitrant and carcinogenic in nature. The commonly applied ponding treatment methods have been reported inefficient for remediating the concentration of the colorants before discharge. The need for sustainable and efficient treatment technique is crucial in order to preserve the environment. In this view, this study reported the first attempt to decolorize POME using a proliferate Klebsiella Pneumonia ABZ11 at varied inoculum sizes of 5-25% (v/v), initial color concentration (650-2,600 ADMI) and treatment time of 5-40 h. The treatment conditions were optimized using Response Surface Methodology. At optimal conditions of 20% (v/v) inoculum size, initial-color concentration of 2,600 ADMI, initial pH of 7 and 35 h treatment retention time, over 80.40% color removal was achieved with insignificant disparity compared with the model predicted value of 81.538%. Also, the Monod model excellently described the decolorization kinetic process with 0.9214 coefficient of correlation (R 2), and the calculated maximum growth μ max ) and half-saturation constant (K s ) were 7.023 d-1 and 340.569 ADMI d-1, respectively. This study revealed that the Klebsiella Pneumonia ABZ11 was highly prolific and such feature may favor a synergistic biodegradation process.Percutaneous devices are a key technology in clinical practice, used to connect internal organs to external medical devices. Examples include prosthesis, catheters and electrical drivelines. Percutaneous devices breach the skin's natural barrier and create an entry point for pathogens, making device infections a widespread problem. Modification of the percutaneous implant surface to increase skin integration with the aim to reduce subsequent infection is attracting a great deal of attention. While novel surfaces have been tested in various in vitro models used to study skin integration around percutaneous devices, no skin model has been reported, for the study of bacterial infection around percutaneous devices. Here, we report the establishment of an in vitro human skin equivalent model for driveline infections caused by Staphylococcus aureus, the most common cause of driveline-related infections. Three types of mock drivelines manufactured using melt electrowriting (smooth or porous un-seeded and porous pre-seeded with human fibroblasts) were implanted in human skin constructs and challenged with S. aureus. Our results show a high and stable load of S. aureus in association with the skin surface and no signs of S. aureus-induced tissue damage. Furthermore, our results demonstrate that bacterial migration along the driveline surface occurs in micro-gaps caused by insufficient skin integration between the driveline and the surrounding skin consistent with clinical reports from explanted patient drivelines. Thus, the human skin-driveline infection model presented here provides a clinically-relevant and versatile experimental platform for testing novel device surfaces and infection therapeutics. This study explored the effects of four sugar source treatments, including no sugar (CON), fructose (FRU), pectin (PEC), and starch (STA), on the microbiota and metabolome of alfalfa (Medicago sativa) silage. The bacterial community was determined via 16S rRNA gene sequencing. The metabolome was analyzed using ultra high-performance liquid chromatography tandem time-of-flight mass spectrometry (UHPLC/TOF-MS). After 60 days of fermentation, the pH values in FRU and PEC were lower than those in STA and CON. FRU had a greater lactic acid concentration compared to STA and CON. Weissella (47.44%) and Lactobacillus (42.13%) were the dominant species in all four groups. The abundance of Pediococcus was lower, and the abundance of Leuconostoc, Pantoea, and Microbacterium was higher, in FRU compared to CON. The abundance of norank_f__Bacteroidales_S24-7_group was higher, and the abundance of Turicibacter was lower, in both FRU and PEC than in CON. Leuconostoc was negatively correlated with the pH value, and Pediococcus was positively correlated with the pH value.
