Long-chain polyunsaturated fatty acids (LCPUFAs) and their metabolites are considered essential factors to support bone and joint health. The n-6 PUFAs suppress the osteoblasts differentiation via increasing peroxisome proliferator-activated receptor gamma (PPARγ) expression and promoting adipogenesis while n-3 PUFAs promote osteoblastogenesis by down-regulating PPARγ and enhancing osteoblastic activity. Arachidonic acid (AA) and its metabolite prostaglandin E2 (PGE2) are key regulators of osteoclast differentiation via induction of the receptor activator of nuclear factor kappa-Β ligand (RANKL) pathway. Marine-derived n-3 LCPUFAs have been shown to inhibit osteoclastogenesis by decreasing the osteoprotegerin (OPG)/RANKL signalling pathway mediated by a reduction of pro-inflammatory PGE2 derived from AA. Omega-3 PUFAs reduce the expression of cartilage degrading enzyme matrix metalloproteinase-13 (MMP-13) and a disintegrin and metalloprotease with thrombospondin motifs-5 (ADAMTS-5) protein, oxidative stress and thereby apoptosis via nuclear factor kappa-betta (NF-kβ) and inducible nitric oxide synthase (iNOS) pathways. In this review, a diverse range of important effects of LCPUFAs on bone cells and chondrocyte was highlighted through different mechanisms of action established by cell cultures and animal studies. This review allows a better understanding of the possible role of LCPUFAs in bone and chondrocyte metabolism as potential therapeutics in combating the pathological complications such as osteoporosis and osteoarthritis.Freezing of gait (FOG) appears to be associated with increased risk of forward falls in patients with Parkinson's disease (PD). This study aimed to experimentally validate forward gait instability in PD patients with FOG (PD + FOG). Eleven PD + FOG patients, 9 PD patients without FOG (PD - FOG), and 13 healthy controls participated. Self-selected paced gait was analyzed by a three-dimensional motion-capture analysis system. We assessed the distance between the center of mass and the base of support (COM-BOS distance) and the margin of stability (MOS), considering the position and velocity of the COM as gait stability parameters, spatiotemporal gait parameters and kinematic parameters. The anteroposterior COM-BOS distance was smaller in PD + FOG patients than in PD-FOG patients and controls. Anteroposterior MOS was larger in PD + FOG and PD - FOG patients than controls (p less then 0.05). PD + FOG patients showed smaller anteroposterior MOS than PD - FOG patients, when adjusting for disease severity (p less then 0.05). Only in the PD + FOG group, when adjusting for disease severity, step length was positively correlated with the anteroposterior COM-BOS distance (p less then 0.05), and cadence was negatively correlated with the anteroposterior MOS (p less then 0.05). These results indicated that PD patients with FOG have forward gait instability and suggested that such instability may be associated with reduced step length and increased cadence.Mound structures that soil termites build have diverse morphologies. Previous observational studies documented that mounds are built to provide regulated environments for the termites that live within them and their structures are formed in ways to support this purpose under the influence of the mounds' immediate environment. The objective of this study is to provide a methodology and a predictive computational model to investigate the reason behind the different but systematic shapes of termite mounds, considering all the relevant forces imposed on them and their thermoregulatory and gas-exchange functions. The gas-exchange function accounts for the capacity of the mound to diffuse metabolic gases generated in the mound's underground nest, while the thermoregulatory function satisfies the connection between the underground nest and deep ground temperatures. The proposed predictive model is based on the principles of heat transfer and thermodynamics and allows optimized mechanically stable structures to freelr external walls, and well-defined cone- (as opposed to the dome-) shaped structures. Overall, the form of termite mounds is strongly correlated to both regulatory functions and local environments, and the resulting mound shape arises as a combination of these factors.

To analyze fatigue after mild traumatic brain injury (TBI) with latent class growth analysis (LCGA) to determine distinct recovery trajectories and investigate influencing factors, including emotional distress and coping styles.

An observational cohort study design with validated questionnaires assessing fatigue, anxiety, depression, posttraumatic stress, and coping at 2 weeks and 3 and 6 months postinjury.

Fatigue was measured with the fatigue severity subscale of the Checklist Individual Strength, including 8 items (sum score, 8-56). Subsequently, 3 clinical categories were created high (score, 40-56), moderate (score, 26-38), and low (score, 8-25).

From the entire mild TBI group, 4 patient clusters with distinct patterns for fatigue, emotional distress, and coping styles were found with LCGA. Clusters 1 and 2 showed favorable recovery from fatigue over time, with low emotional distress and the predominant use of actcute phase postinjury based on the presence of high fatigue, high passive coping, and, for a subgroup of patients, high emotional distress. LCGA proved to be a highly valuable and multipurpose statistical method to map distinct courses of disease-related processes over time.The olfactory bulbectomized (OBX) rat model is a well-established model of depression in which antidepressant drugs reverse deficits in the passive avoidance test 14 days after administration. Recently, the olfactory bulbectomized rat model has been proposed to be a model of Alzheimer's disease (AD), and the available data indicate similarities between the changes that typically occur in AD and those observed in OBX animals. In the present study, the occurrence of neurochemical impairments related to AD were investigated 8 months after OB ablation. The expression of the nitric oxide synthases eNOS and nNOS, receptor for advanced glycation endproducts (RAGEs) and dimethylarginine dimethylaminohydrolase (DDAH1) in the prefrontal cortices (PFCs), hippocampi and striata of olfactory bulbectomized and sham-operated rats was evaluated. Subsequently, the impact of the administration of a positive allosteric modulator of the mGlu5 receptor, CDPPB (14 days, 2.5 or 5 mg/kg), on OBX-related changes was assessed. OB ablation induced typical deficits in passive avoidance. Significant aberrations in the expression of both isoforms of NOS were observed in the hippocampus and striatum, and the expression of DDAH1 was increased in the PFCs of OBX animals. CDPPB at a dose of 5 mg/kg ameliorated cognitive impairment in the passive avoidance test and partially reversed the changes in eNOS and nNOS expression induced by the lesion. The results of this study confirm that some of the neurochemical changes observed in OBX animals may resemble those associated with AD pathology and that activation of the mGlu5 receptor may partially counteract these pathological alterations.A new species of black fly from rock fields of southeaster Brazil, Simulium maiaherzogae sp. nov., is described based on morphological features of larval, pupal and adult specimens. Morphological features of this new species were compared with members of Simulium (Trichodagmia) Enderlein 1934, showing a high similarity with S. scutistriatum Lutz 1909 and S. itajara Nascimento, et al. 2020. However, features of the male and female scutum and genitalia, pupal tubercles disposition, pupal gills, larval cephalic appendages, larval gill histoblast, numbers of posterior proleg hooks and rectal papillae lobules, allow to differentiate S. maiaherzogae sp. nov. Moreover, a more detailed analysis using Scanning Electron Microscopy (SEM) showed significant differences and numerical variations in meristic data between the new species and S. ML324 chemical structure scutistriatum. To date, no drugs or vaccines effectively treat or prevent Der p 1 sensitization. We applied in silico immunoinformatics to design T-cell and B-cell epitopes that were specified and developed from the allergen Der p 1 of D. pteronyssinus. We identified the conserved epitope areas by predicting the accessibility and flexibility of B-cell epitopes, and the percentage of human leukocyte antigen representing T cells. Molecular docking using HADDOCK software indicated three optimal clusters cluster 6 (z-score -2.1), cluster 1 (z-score -1.2), and cluster 3 (z-score -0.6). The most negative Z-score was found in cluster 6, which represented three epitopes. The interaction between A chain proteins (IgE protein residues) and B chains (Der p 1 protein residues) exhibited a knowledge-based FADE and contact value >1, suggesting the best protein interactions occurred in the conserved area. Molecular dynamic simulation further predicted the stable nature of Der p 1 protein. The IQRDNGYQP region is the best candidate to be utilized as a D. pteronyssinus epitope vaccine, which could be used in the development of allergen-specific immunotherapy.

States have passed legislation to expand the scope of pharmacists to directly prescribe contraception. It is thought that pharmacist prescription of contraception may promote correct and consistent use of contraception by reducing barriers to access. However, it is not known how this may impact ongoing contraceptive use.

This study aimed to determine whether 12-month rates of continuation of an effective form of contraception or perfect use of contraception differ by prescribing provider (pharmacist or clinician).

We conducted a 1-year prospective cohort study of 388 women seeking contraception in 139 pharmacies across 4 states (California, Colorado, Hawaii, and Oregon). Our study was powered to detect a 10% difference in 12-month continuation of an effective form of contraception. We clarified women's pregnancy intention at baseline and subsequent follow-ups. Women received a prescription directly from a pharmacist (n=149) or were filling a prescription from a clinician, our comparison group (n=239). Wted odds ratio, 0.70; confidence interval, 0.28-1.71) or with perfect use of contraception (adjusted odds ratio, 0.87; confidence interval, 0.51-1.48), controlling for other woman-level characteristics.

We found no difference in use of any effective contraception, perfect use, or switching at 12 months among those who received their baseline prescription from a pharmacist vs a clinician. This study is limited by not examining information on safety outcomes.
