To elucidate the relationship between antofloxacin (AT) plasma concentration and QT interval prolongation, compare the effects of different correction and analytical methods on conclusions, and estimate the possible false-positive rate in thorough QT (TQT) studies.

Twenty-four healthy Chinese volunteers from a four-period crossover TQT study orally received 200 mg/d AT, 400 mg/d AT, 400 mg/d moxifloxacin, and a placebo in a random order for 5 d for each. QT interval samples were collected on d 1 and d 5. Population models were established describing the relationship between QT and AT concentration. The yardstick from ICH E14 guidelines was used to measure the effect of drugs on QT prolongation both in biostatistical and modeling analyses. A possible false-positive rate was estimated by constructing a 1000-time bootstrap to obtain the rate-of-difference values between d 1 and d 5 over 5 ms in the placebo period.

In the modeling analysis, the QT prolongation estimate at the mean maximal concentration of AT (4.51 μg/mL) was 3.84 ms, and its upper bound of the one-sided 95 % CI was 7.04 ms, which showed a negative effect on QT interval prolongation. The estimation for the false-positive rate was 31 % in this study.

Several essential oils (EOs) have displayed some antimicrobial activity and could potentially act as antibiotic adjuvants. Research in this area aims to develop new therapeutic alternatives to treat infections caused by these pathogens. MICs of different EOs were determined against A. baumannii and Klebsiella pneumoniae. The fractional inhibitory concentration index (FICindex) was calculated in order to categorize the interaction. Time-kill assays were also performed. The EOs that displayed the highest levels of antimicrobial activity were clove (Syzygium aromaticum L.) and thyme (Thymus zygis L.). Combined disk diffusion tests and checkerboard assays revealed synergy between these EOs and colistin. Addition of either clove or thyme EO decreased the MIC of colistin by 8- to 64-fold and 8- to 128-fold in the colistin-resistant A. baumannii and K. pneumoniae strains, respectively (FICindex ≤ 0.5, synergy). MICs were also reduced in the colistin-susceptible strains. Time-kill assays also indicated the strong activity of the combined therapy. In summary, the use of clove or thyme EO in combination with colistin could improve the efficacy of the antibiotic and significantly reduce the concentrations needed to inhibit growth of A. baumannii and K. pneumoniae.

Swertiamarin (Sw) and quercetin (Qu) have been isolated from different plants and are reported for their antidiabetic activities. The plants from which swertiamarin and quercetin were isolated are also traditionally used in the treatment of diabetes mellitus.

The present study is aimed to evaluate the synergistic effect of a combination of swertiamarin and quercetin (CSQ) on α-amylase in vitro and on streptozotocin (STZ) induced diabetes mellitus in vivo.

Swertiamarin was isolated from the plant Enicostemma axillare and quercetin was procured in its pure form. Sw, Qu and CSQ were evaluated for in vitro α-amylase inhibitory activity. Based on the in vitro study results, CSQ was assessed for in vivo streptozotocin induced diabetes mellitus in Wistar rats. The effect of CSQ on blood glucose levels, body weight, serum biochemical parameters and antioxidant enzymes such as glutathione (GSH), superoxide dismutase (SOD), catalase, glutathione peroxidase (GPx) and lipid peroxide levels were estimated. The histopathological observations of pancreatic tissues were also made.
