Male and female Plasmodium gametocytes ingested by the Anopheles mosquitoes during a blood meal egress from the red blood cells by rupturing the two surrounding membranes, the parasitophorous vacuole and the red blood cell membranes. Proteins of the so-called osmiophilic bodies, (OBs), secretory organelles resident in the cytoplasm, are important players in this process. Once gametes emerge, the female is ready to be fertilized while the male develops into motile flagellar gametes. Here, we describe the function(s) of PBANKA_1115200, which we named Gamete Egress Protein (GEP), a protein specific to malaria parasites. GEP is restricted to gametocytes, expressed in gametocytes of both genders and partly localizes to the OBs. A mutant lacking the protein shows aberrant rupture of the two surrounding membranes, while OBs discharge is delayed but not aborted. Moreover, we identified a second function of GEP during exflagellation since the axonemes of the male flagellar gametes were not motile. Genetic crossing experiments reveal that both genders are unable to establish infections in mosquitoes and thus the lack of GEP leads to a complete block in Plasmodium transmission from mice to mosquitoes. The combination of our results reveals essential and pleiotropic functions of GEP in Plasmodium gametogenesis. © 2020 The Authors. MicrobiologyOpen published by John Wiley & Sons Ltd.The present study analyses the role of weight distortion and weight self-criticism in explaining the relationship between actual weight and perceived health, well-being, body satisfaction and emotional eating. Eight hundred and six individuals were selected from a national study on nutrition. They were divided into two groups according to self-reported dieting. A questionnaire measuring the outcome variables was implemented. Weight distortion and self-criticism were calculated using actual, estimated and desired BMI. A multiple mediation model was estimated using structural equation modelling. A valid model was obtained in which weight self-criticism is a partial mediator of the relationship between weight and body satisfaction and a full mediator of the relationship between weight and emotional eating for the non-dieter group. For the dieter group, full mediation of weight self-criticism is observed in the relationships between weight and health, well-being and body satisfaction. For weight distortion, no mediation effect is found in either group. The results highlight the importance of weight perceptions in understanding the influence of body weight on several aspects of health and well-being. The type of weight perception is also relevant. Here, a new method for real-time monitoring and sorting of single cells based on their inherent levitation profiles driven by their lipid content is reported. As a model system, human-induced pluripotent stem cell (hiPSC)-derived cardiomyocytes (CMs) from a patient with neutral lipid storage disease (NLSD) due to loss of function of adipose triglyceride lipase (ATGL) resulting in abnormal lipid storage in cardiac muscle are used. This levitation-based strategy detects subpopulations within ATGL-deficient hiPSC-CMs with heterogenous lipid content, equilibrating at different levitation heights due to small density differences. In addition, sorting of these differentially levitating subpopulations are monitored in real time. Using this approach, sorted healthy and diseased hiPSC-CMs maintain viability and function. KGaA, Weinheim.BACKGROUND AND OBJECTIVES Expanded hemodialysis therapy (HDx) provides increased clearance of conventional and large middle molecules through innovative medium cut-off (MCO) membranes. However, there is a paucity of real-world data regarding the benefits and safety of HDx. This large observational study evaluated outcomes among patients in Colombia undergoing HDx at a large dialysis clinical services provider. DESIGN, SETTING, PARTICIPANTS, & MEASUREMENTS This was a prospective single cohort study of prevalent patients who were treated with HDx; baseline information was collected from the most recent data before patients were started on HDx. Patients were followed prospectively for 1 year for changes in serum albumin and other laboratory parameters compared with the baseline. Survival, hospitalization and safety were assessed from the start of HDx. RESULTS A total of 1000 patients were invited to enroll; 992 patients met the inclusion criteria for data analysis and 638 patients completed the year of follow-up. Seventy-four (8%) patients died during 866 patient-years (PY) of follow-up; the mortality rate was 8.54 deaths/100 PY (95% confidence interval [CI], 6.8 to 10.7). There were 673 hospitalization events with a rate of 0.79 events/PY (95% CI, 0.73 to 0.85) with 6.91 hospital days/PY (95% CI, 6.74 to 7.09). The observed variability from baseline and maximum average change in mean serum albumin levels were - 1,8% and - 3,5% respectively. No adverse events were related to the MCO membrane.
