MicroRNAs (miRNAs) are small, endogenous, non-coding, single stranded RNAs which play a role in the regulation of gene expression and function. Therefore, the analysis of differentially expressed miRNAs are of great importance in disease diagnosis. This study is focussed on the differential expression of miRNAs in serum of PCOS subjects compared to control and their correlation with metabolic and endocrine parameters. Anthropometry, hormone concentrations and biochemical characteristics were measured in healthy (n = 20) and PCOS (n = 20) subjects. MiR-24, miR-29a and miR-502-3p were determined in serum by quantitative RT-PCR. The levels of miR-24 was significantly decreased in PCOS subjects (P = 0.00) compared to control. No significant difference was observed in the levels of miR-29a and miR-502-3p in PCOS and control subjects. MiR-24 showed significant inverse correlation with BMI, glucose, insulin, FIRI, HOMA, LH, testosterone, TG, and LHFSH ratio whereas HDL levels showed significant positive association with miR-24 and miR-29a. LH showed significant negative association with miR-29a. No correlation was observed between the expression of miR-502-3p with any of the studied parameters. The receiver operating characteristic curve for miR-24 alone showed a significant discriminative capacity. The study suggests that serum miR-24 analysis in PCOS patients could be of diagnostic value that can be used as a biomarker for PCOS. © Association of Clinical Biochemists of India 2019.Survivin can affect the progression of infection and is considered as a marker of various malignancies. Selleckchem Panobinostat The aim of the study was to investigate the possible association of gene polymorphisms of survivin (-1547A/G, -644C/T, -625 C/G, -241C/T, -31G/C, -141G/C) and chronic hepatitis B infection in Iranian patients. The genotypes of survivin SNPs were investigated by polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) method using 100 chronic HBV infected patients (HBV), 40 spontaneously recovered HBV subjects and 100 healthy controls (C). Serum level of survivin was determined using ELISA method. The -1547G, -625C, -241T and -31C alleles were associated with increased susceptibility to chronic hepatitis B infection (P = 0.001, P  less then  0.001, P = 0.003 and P  less then  0.001 respectively). Chronic HBV patients with -625CC, -241TT and -31CC genotypes had higher levels of survvin. Survivin -1547A/G, -625 C/G, -241C/T and -31G/C gene polymorphisms may be associated with chronic HBV susceptibility in Iranian HBV patients. © Association of Clinical Biochemists of India 2019.The present work has been designed to investigate the hepatoprotective and renoprotective efficiency of alcoholic extract of Allium porrum and Bauhinia variegata leaves in thioacetamide-induced toxicity in adult Wistar rats. Allium porrum leaf extract, Bauhinia variegata leaf extract and their combinations were orally administered for 14 days then TAA (300 mg/kg) i.p. was injected once and the rats were sacrificed 2 days later. Plasma AST, ALT, GGT, total bilirubin, creatinine, urea, uric acid, triglyceride, cholesterol, HDL and LDL were measured. Liver MDA, GSH, CAT, SOD and TNF-α were evaluated. Histological examination was performed. The rats treated with TAA showed a significant increase in AST, ALT, GGT, total bilirubin, creatinine, urea, uric acid, total, triglyceride, cholesterol and HDL while it led to a significant decrease in protein and HDL. The treatment of rats with TAA resulted in a significant decrease of the hepatic GSH, SOD and CAT and a significant elevation of MDA and TNF-α. Allium porrum and Bauhinia variegata extracts alleviated the toxic effects of TAA on the liver and the kidney. In conclusion, treatment with Allium porrum and Bauhinia variegata extracts and their combination reduced deleterious effects of TAA on liver through antioxidant and anti-inflammatory properties. © Association of Clinical Biochemists of India 2019.Telomeres play an important role to conserve genomic integrity by protecting the ends of chromosomes in normal cells. Notably, telomere length is perpetuated by telomerase in large majority of cancers, thereby ensure indefinite cell proliferation-a hallmark of cancer-and this unique feature has provided telomerase as the preferred target for drug development in cancer therapeutics. Cancer cells have acquired the potential to have telomere length maintenance by telomerase activation- up-regulation of hTERT gene expression in tumor cells is synchronized by multiple genetic and epigenetic modification mechanisms viz hTERT structural variants, hTERT promoter mutation and epigenetic modifications through hTERT promoter methylation which have been implicated in various cancers initiation and progression. In view of these facts, strategies have been made to target the underlining molecular mechanisms involved in telomerase reactivation as well as of telomere structure with special reference to distortion of sheltrin proteins. This review is focussed on extensive understanding of telomere and telomerase biology. which will provide indispensable informations for enhancing the efficiency of rational anticancer drug design. However, there is also an urgent need for better understanding of cell signalling pathways for alternative lengthening of telomere which is present in telomerase negative cancer for therapeutic targets. © Association of Clinical Biochemists of India 2020.Low consistency (LC) refining of (chemi-)thermomechanical pulp (TMP) provides an energy efficient alternative to high consistency refining for pulp property development. However, the benefit of LC refining is often limited by excessive fibre shortening, lower tear strength and a reduction of bulk caused by the refining process. In this study, microfibres produced by LC refining of TMP and kraft pulp fibres were investigated for their reinforcement potential in high freeness mechanical pulp. Primary pulp at 645 mL Canadian Standard Freeness was LC refined to different energy targets as a baseline for mechanical and optical property development. In contrast, the same primary pulp was reinforced with different microfibre types in ratios that yielded the same specific energies of the baseline LC refined pulp. The study revealed that at equivalent energies, the addition of TMP microfibres to the high freeness primary pulp displayed tensile development identical to the LC refined pulp, with significantly improved tear and bulk.
