The subsequent steps involve desaturation, isomerisation and cyclisation reactions to form α- and β-carotene stereoisomers, via all-trans lycopene. The marine microalga Dunaliella salina is the richest source of β-carotene, but it can accumulate phytoene and phytofluene as well. In the present study, different analytical tools including High-Performance Liquid Chromatography (HPLC), Ultra-Performance Convergence Chromatography (UPC2-MS) and Nuclear Magnetic Resonance (NMR) were used to characterize and quantify the phytoene isomeric configurations in D. salina in order to explore both the feasibility of D. salina as a cell factory for phytoene production and to gain new insight into the carotenoid synthesis pathway in D. salina. D. salina, similar to tomato, produced predominantly 15-cis phytoene isomer (>98%) and a trace amount of all-trans phytoene ( less then 2%). High light stress, red light stress, or use of a phytoene desaturase inhibitor or a mitotic disrupter herbicide led to the accumulation of 15-cis phytoene but not all-trans phytoene. 9-cis phytoene was not detected in any of the extracts of D. salina biomass. Our main findings suggest that 15-cis phytoene is the most abundant isomer in D. salina and that it is subject to a series of isomerisation and desaturation reactions to form all-trans and 9-cis β-carotene.Berberine is a quaternary isoquinoline alkaloid that has been isolated from numerous plants which are still in use today as medicine and herbal supplements. Despite the longstanding international and national legislation regulating transnational trade and domestic recycling, informal e-waste economies are still clustering in many Global South countries. This study offers historically and geographically specific explanations of this conundrum, by interrogating the multi-scalar regulatory frameworks in which the informal e-waste economies and their pollutions are embedded, by drawing on China, particularly the former global e-waste hub-Guiyu town, as the case study. We argue that the contested and problematic application of current international and national legislation in regulating e-waste is in part pertaining to the slippery definition of what counts as "e-waste" and its paradoxical nature as both resources and pollutants. At the global scale, trajectories of global e-waste flows are shaped by the multitude of loopholes, contradictions and ambiguous articles left by the Basel Convention and by different countries' disparate attitudes towards the e-waste trade. At the national scale, the ambiguities and contradictions in the Basel Convention have been passed on to and shaped China's national e-waste regulatory frameworks. China's equivocal legislation, paradoxical attitude, and formal enterprises' weak competence contribute to the rise of informal e-waste recycling in Guiyu. Yet, China's e-waste regime has been greatly restructured within the past decade, with formal recycling enterprises playing an increasingly significant role.The anxiolytic and antidepressant-like activities of the naturally occurring monoterpene 1,8-cineole and its structural isomer 1,4-cineole were evaluated in mice via inhalation administration at doses ranging from 4 × 10-6 to 4 × 10-1 mg per 400 μL of triethyl citrate. Mice were tested for anxiety-like behaviours by using the light-dark box test (LDB) and marble-burying test (MBT) and for depression-like symptoms by using the forced swimming test (FST) and tail suspension test (TST). Diazepam and fluoxetine were used as standard drugs for anxiolytic and antidepressant tests, respectively. The results showed that 1,8-cineole at 4 × 10-4 mg, and 1,4-cineole at 4 × 10-4 and 4 × 10-3 mg significantly increased the amount of time spent in the light box and the number of entries in the light box in the LDB as well as reduced the number of marbles buried in the MBT relative to those in the control, suggesting an anxiolytic effect. Similarly, 1,8-cineole at 4 × 10-4 and 4 × 10-2 mg and 1,4-cineole at doses of 4 × 10-4 to 4 × 10-2 mg significantly reduced immobility times in the FST and TST relative to those of the control, suggesting an antidepressant activity. The role of the GABAA/benzodiazepine receptor system in the anxiolytic effects of 1,8- and 1,4-cineole was investigated through co-administration of flumazenil, a GABAergic system antagonist. Flumazenil reversed the effects of diazepam and 1,8-cineole, suggesting that 1,8-cineole affects the GABAA/benzodiazepine receptors. We investigated the impacts of culture media, coconut water (CW), and plant hormones (gibberellic acid (GA3), indole-3-acetic acid (IAA), indole-3-butyric acid (IBA), α-naphthaleneacetic acid (NAA), and thidiazuron (TDZ)) on asymbiotic germination, multiplication and conversion of protocorms, and plantlet development. Maximal seed germination (93.3%) was achieved on ½ MS medium without vitamins plus 5% CW, 1 µM NAA, and 1.5 µM GA3. The results showed that increasing the length of the RNA duplex in such a conjugate increases its biological activity when delivered using a transfection agent. However, the efficiency of accumulation in human drug-resistant KB-8-5 cells during delivery in vitro in a carrier-free mode was reduced as well as efficiency of target gene silencing. TsiRNAs demonstrated a similar biodistribution in KB-8-5 xenograft tumor-bearing SCID mice with more efficient accumulation in organs and tumors than cholesterol-conjugated canonical siRNAs; however, this accumulation did not provide a silencing effect. The lack of correlation between the accumulation in the organ and the silencing activity of cholesterol conjugates of siRNAs of different lengths can be attributed to the fact that trimeric Ch-TsiRNA lags mainly in the intercellular space and does not penetrate sufficiently into the cytoplasm of the cell. Super Skills for Life (SSL) is a CBT-based intervention to reduce anxiety and comorbid problems that, apart from social skills training and behavioural activation, integrates video-feedback with cognitive preparation. This study aimed to evaluate SSL effects, implemented in a school setting, on social performance and to test self-concept and social skills as potential mediators of pre- and post-test changes in social anxiety and generalized anxiety. Sample comprised 57 children aged 8-11 years with emotional symptoms. Children were video recorded in the first and last session to assess social performance. Anxiety and self-concept measures were completed by children pre-test and post-test. Participants reduced anxiety behaviours and improved social and communication skills after treatment. In general, girls showed better social performance than boys, but SSL impact was greater in males. Social self-concept was the only mediator of change in pre- to post-treatment social anxiety. This study provides evidence of SSL to improve children's social performance and reduce anxiety through video-feedback with cognitive preparation. Improving social concept seems essential to reduce social anxiety. An SSL programme is an ideal prevention protocol for anxious children.Drug target prediction is an important method for drug discovery and design, can disclose the potential inhibitory effect of active compounds, and is particularly relevant to many diseases that have the potential to kill, such as dengue, but lack any healing agent. An antiviral drug is urgently required for dengue treatment. Some potential antiviral agents are still in the process of drug discovery, but the development of more effective active molecules is in critical demand. Herein, we aimed to provide an efficient technique for target prediction using homopharma and network-based methods, which is reliable and expeditious to hunt for the possible human targets of three phenolic lipids (anarcardic acid, cardol, and cardanol) related to dengue viral (DENV) infection as a case study. Using several databases, the similarity search and network-based analyses were applied on the three phenolic lipids resulting in the identification of seven possible targets as follows. Based on protein annotation, three phenolic lipids may interrupt or disturb the human proteins, namely KAT5, GAPDH, ACTB, and HSP90AA1, whose biological functions have been previously reported to be involved with viruses in the family Flaviviridae.
