An essential requirement for the success of in vitro maturation (IVM) of the oocyte is to provide an optimal microenvironment similar to in vivo conditions. Recently, somatic cell-based coculture or supplementation of a conditioned medium during IVM has been performed to obtain better quality of oocytes, because they mimic the in vivo reproductive tract by secreting paracrine factors. In this study, human adipose-derived stem cells (ASC) and their conditioned medium (ASC-CM) were applied to IVM of porcine oocytes to evaluate the effectiveness of ASC on oocyte development and subsequent embryo development. In results, both ASC and ASC-CM positively influence on oocyte maturation and embryo development by regulating growth factor receptors (VEGF, FGFR, and IGFR), apoptosis (BCL2), cumulus expansion (PTGS2, HAS2, and TNFAIP6), and oocyte maturation-related genes (GDF9 and BMP15). In particular, the fluorescence intensity of GDF9 and BMP15 was markedly upregulated in the oocytes from the ASC-CM group. Furthermore, significantly high levels of growth factors/cytokine including VEGF, bFGF, IGF-1, IL-10, and EGF were observed in ASC-CM. Additionally, the ASC-CM showed active scavenging activity by reducing the ROS production in a culture medium. Consequently, for the first time, this study demonstrated the effect of human ASC-CM on porcine oocyte development and the alteration of mRNA transcript levels in cumulus-oocyte complexes.Extracellular vesicles (EVs) have been isolated from follicular (FF) and ampullary oviduct fluid (AOF), using different isolation methods. However, it is not clear whether different purification methods can affect the functionality of resulting EVs. Here, we compared two methods (OptiPrep™ density gradient ultracentrifugation (ODG UC) and single-step size exclusion chromatography (SEC) (qEV IZON™ single column)) for the isolation of EVs from bovine FF and AOF. Additionally, we evaluated whether the addition of EVs derived either by ODG UC or SEC from FF or AOF during oocyte maturation would yield extra benefits for embryo developmental competence. The characterization of EVs isolated using ODG UC or SEC from FF and AOF did not show any differences in terms of EV sizes (40-400 nm) and concentrations (2.4 ± 0.2 × 1012-1.8 ± 0.2 × 1013 particles/mL). Temozolomide Blastocyst yield and quality was higher in groups supplemented with EVs isolated from FF and AOF by ODG UC, with higher total cell numbers and a lower apoptotic cell ratio compared with the other groups (p less then 0.05). Supplementing in vitro maturation media with EVs derived by ODG UC from AOF was beneficial for bovine embryo development and quality.Wide-bandgap (WBG) material-based switching devices such as gallium nitride (GaN) high electron mobility transistors (HEMTs) and silicon carbide (SiC) metal-oxide-semiconductor field-effect transistors (MOSFETs) are considered very promising candidates for replacing conventional silicon (Si) MOSFETs for various advanced power conversion applications, mainly because of their capabilities of higher switching frequencies with less switching and conduction losses. However, to make the most of their advantages, it is crucial to understand the intrinsic differences between WBG- and Si-based switching devices and investigate effective means to safely, efficiently, and reliably utilize the WBG devices. This paper aims to provide engineers in the power engineering field a comprehensive understanding of WBG switching devices' driving requirements, especially for mid- to high-power applications. First, the characteristics and operating principles of WBG switching devices and their commercial products within specific voltage ranges are explored. Next, considerations regarding the design of driving circuits for WBG switching devices are addressed, and commercial drivers designed for WBG switching devices are explored. Lastly, a review on typical papers concerning driving technologies for WBG switching devices in mid- to high-power applications is presented.Intense selection for milk yield has increased environmental sensitivity in animals, and currently, heat stress is an expensive problem in dairy farming. The objectives were to identify the best model for characterizing environmental sensitivity in Holstein cattle, using the test-day milk yield (TDMY) combined with the temperature-humidity index (THI), and identify sires genetically superior for heat-stress (HS) tolerance and milk yield, through random regression. The data comprised 94,549 TDMYs of 11,294 first-parity Holstein cows in Brazil, collected from 1997 to 2013. The yield data were fitted to Legendre orthogonal polynomials, linear splines and the Wilmink function. The THI (the average of two days before the dairy control) was used as an environmental gradient. An animal model that fitted production using a Legendre polynomials of quartic order for the days in milk and quadratic equations for the THI presented a better quality of fit (Akaike's information criterion (AIC) and Bayesian information criterion (BIC)). The Spearman correlation coefficient of greatest impact was 0.54, between the top 1% for TDMY and top 1% for HS. Only 9% of the sires showed plasticity and an aptitude for joint selection. Thus, despite the small population fraction allowed for joint selection, sufficient genetic variability for selecting more resilient sires was found, which promoted concomitant genetic gains in milk yield and thermotolerance.With the development of Internet technology, the speed of information dissemination and accelerated updates result in frequent discussion of topics and expressions of public opinion. In general, multi-dimensional discussion topics related to the same event are often generated in the network, and the phenomenon of multi-dimensional public opinion polarization is formed under the mutual influence of groups. This paper targets the phenomenon of multi-dimensional public opinion polarization under topic-derived situations as the research object. Firstly, this paper identifies the factors influencing multi-dimensional public opinion polarization, including the mutual influence of different topic dimensions and the interaction of viewpoints within the same topic. Secondly, the topic correlation coefficient is introduced to describe the correlation among topics in different dimensions, and the individual topic support degree is used to measure the influence of topics in different dimensions and that of information from external intervention on individual attitudes.
