We also found that 6PPD-Q was leached from dust particles within a few hours, with a log partitioning coefficient between organic carbon and water (KOC) of 3.2-3.5. The present results will help to understand the environmental occurrence, fate, and behavior of 6PPD-Q.Beaches are an integral part of coastal tourism, but they are deteriorated by the beachgoers and recreational activities due to lack of adequate beach environmental awareness and management. Litter is widely distributed in marine and coastal environment and has been considered a severe concern. In China investigations to determine the beach litter abundance and pollution level are limited. The aim of this study is to estimate spatio-temporal distribution and composition of litter on 10 well-known Qingdao tourist beaches, involving pollution level by beach quality indexes. Beach litter was collected within an area of 25 × 25 m2 in both summer (May, June and July) and winter (Nov, Dec and Jan) seasons, and was classified into eight categories. The abundance of beach litter was found higher in summer (0.13 ± 0.04 items/m2) than in winter (0.04 ± 0.01 items/m2). Overall, the percentage of plastics were higher in both summer (23.48%) and winter (24.04%) than that of other litter categories. Despite regular cleaning operation along most of Qingdao beaches, suggested management practices involve mitigation measures, source reduction, change in littering behavior to improve further quality of beaches.Lead concentrations in hard antlers of adult European roebucks (Capreolus capreolus) were analyzed to assess lead exposure of roe deer roaming the floodplain of the Innerste River, a river system contaminated due to historical metal ore mining, processing, and smelting in its upper reaches. Antler lead concentrations of roebucks culled in the period 1939-2018 within or close to the Innerste floodplain ranged between less then 0.17 mg Pb/kg (limit of detection) and 51.5 mg Pb/kg (air-dry weight). Median lead concentration in antlers of roebucks culled within the floodplain was 11.1 mg Pb/kg, compared to 2.3 mg Pb/kg in antlers of bucks culled in the floodplain vicinity (P less then 0.01). Sampling year had no significant effect on antler lead concentrations (P = 0.748). Lead isotope ratios of antlers from the Innerste downstream area (206Pb/207Pb 1.179-1.181; 208Pb/206Pb 2.083-2.085) fell within the range of those reported for hydrothermal vein deposits from the upper catchment area of the Innerste River in the Harz Mountains. Our study demonstrates the long-lasting impact of the historical metal ore mining, processing, and smelting in the Harz Mountains on lead pollution in floodplains of rivers draining this area and the lead exposure of wild herbivores inhabiting the floodplains. Furthermore, it highlights the suitability of roe deer antlers for monitoring environmental lead levels and the usefulness of lead isotope signatures in antlers for source apportionment of lead pollution.The importance of per-, poly-fluoroalkyl substances (PFASs) effects on riverine microbiomes is receiving increased recognition in the environmental sciences. However, few studies have explored how PFASs affect microbiomes across trophic levels, specifically through predator-prey interactions. This study examined the community profiles of planktonic archaea, bacteria, fungi, algae, protozoa, and metazoa in a semi-industrial and agricultural river alongside their interactions with 15 detected PFASs. As abiotic factors, PFASs affected community coalescence more than biogenic substances (p less then 0.05). For biotic regulations, sub-communities in rare biospheres (including always rare taxa-ART and critically rare taxa-CRT) contributed to spatial community coalescence more than sub-communities in abundant biospheres (always abundant taxa-AAT and critically abundant taxa-CAT) (p less then 0.05). Metazoa-bacteria (Modularity = 1.971) and protozoa-fungi (1.723) were determined to be the most stable predator-prey networks. Based on pathway models, short-chain PFBA (C4) was shown to weaken the trophic transfer efficiencies from heterotrophic bacteria (HB) to heterotrophic flagellates (HF) (p less then 0.05). Increased di-ethylhexyl phthalate (DEHP) exposure is correlated with chronic inflammation in vivo and increased cytokine release in exposed monocytes in vitro. To determine if acute phthalate exposure enhanced inflammation in a model of cardiac damage, we measured immune cell infiltration, inflammasome expression and cardiac function in male C57bl/6 N mice exposed to phthalates during recovery from a surgically-induced myocardial infarction (MI). Phthalate exposed mice had greater neutrophil and pro-inflammatory macrophage infiltration, greater cardiac dilation and reduced cardiac function when compared with control mice. The greater expression of NLRP3 and NLRP6, but not AIM2 or P2xR7, in the infarcts of phthalate exposed versus control mice suggests a selectivity in pattern recognition receptor activation. Treatment of human THP-1 macrophages with phthalates revealed increased NLRP3 and NLRP6 expression and induction of a pro-inflammatory macrophage population. Pre-treatment with the PPARγ antagonist GW9662 reduced these increases. An increase in expression of IL-1R, MyD88 and IRAK4 in infarcts of phthalate exposed mice and THP-1 cells argues for greater priming downstream of IL-1R signaling and increased susceptibility for inflammasome activation. However, after a protracted drug administration, many patients develop a form of androgen insensitivity since R-bicalutamide starts to exhibit some agonistic properties lead by the W741L AR mutation in the ligand-binding pocket even if the mechanism of the antagonist-agonist switch is still not clear. To study the drug-resistant mechanism, we explored the structural effects of the antagonist R-bicalutamide on the homodimer stability considering both the AR wild-type and W741L employing molecular dynamic (MD) simulations. The results obtained indicate that the binding of R-bicalutamide in the two AR monomers induces a great instability in the homodimer, which may determine the monomer's dissociation preventing AR migration into the nucleus and avoiding the transcriptional activity. If the W741L mutation occurs, the homodimer tends to have a behaviour close to the agonistic system where the two monomers are tightly bound, which may explain the effect of the W741L in drug insensitivity from a structural point of view.Parkinson's disease (PD) is the second most common degenerative disease of the central nervous system (CNS) after Alzheimer's disease. In addition to the typical motor symptoms, the clinical manifestations of patients with PD include gastrointestinal symptoms, which even precede the motor symptoms. Recent research has found that the gut microbiota regulates the brain-gut axial interaction through immune, endocrine, and direct neural mechanisms, supporting the hypothesis that the pathological process of PD spreads from the gut to the brain. In this review article, we highlight the landmark findings in the field of PD, with particular attention to the brain-gut-microbiota axis. We summarize the changes and their clinical effects on the gut microbiota and metabolites observed in PD. The intestinal microbiota may contain appropriate targets for the prevention and treatment of PD. Clinical cohort studies suggest that certain intestinal microbes have protective or pathogenic effects on the progression of PD. A better understanding of the interaction between the gut-brain axis, the gut microbiota, and PD has the potential to lead to new diagnostic and therapeutic approaches. Animal experiments suggest that fecal microbiota transplantation (FMT) is helpful for treating PD, and FMT is expected to be an effective treatment for PD in the future.

We compared the prognostic abilities of neurofilament light (NfL) and neuron-specific enolase (NSE) in patients resuscitated from out-of-hospital cardiac arrest (OHCA) of various aetiologies.

We analysed frozen blood samples obtained at 24 and 48 hours from OHCA patients treated in 21 Finnish intensive care units in 2010 and 2011. We defined unfavourable outcome as Cerebral Performance Category (CPC) 3-5 at 12months after OHCA. We evaluated the prognostic ability of the biomarkers by calculating the area under the receiver operating characteristic curves (AUROCs [95% confidence intervals]) and compared these with a bootstrap method.

Out of 248 adult patients, 12-month outcome was unfavourable in 120 (48.4%). The median (interquartile range) NfL concentrations for patients with unfavourable and those with favourable outcome, respectively, were 689 (146-1804) pg/mL vs. 31 (17-61) pg/mL at 24h and 1162 (147-4360) pg/mL vs. 36 (21-87) pg/mL at 48h, p<0.001 for both. The corresponding NSE concentrations were 13.3 (7.2-27.3) µg/L vs. 8.5 (5.8-13.2) µg/L at 24h and 20.4 (8.1-56.6) µg/L vs. 8.2 (5.9-12.1) µg/L at 48h, p<0.001 for both. The AUROCs to predict an unfavourable outcome were 0.90 (0.86-0.94) for NfL vs. 0.65 (0.58-0.72) for NSE at 24h, p<0.001 and 0.88 (0.83-0.93) for NfL and 0.73 (0.66-0.81) for NSE at 48h, p<0.001.

Compared to NSE, NfL demonstrated superior accuracy in predicting long-term unfavourable outcome after OHCA.Many sarcomas with DICER1 pathogenic variants (PVs) exhibit a characteristic morphology, including a subepithelial layer of malignant mesenchymal cells, areas of rhabdomyoblastic differentiation and cartilaginous and/or osseous elements. We report 5 DICER1-associated neoplasms (1 moderately to poorly differentiated Sertoli Leydig cell tumour and 4 sarcomas) containing variable amounts of neuroectodermal elements. The neoplasms predominantly involved or were in close proximity to the female genital tract (ovary, uterine corpus, abdominal and pelvic cavity) and occurred in females aged 14 months to 54 years. The neuroectodermal elements were characterised by solid and tubular/rosette-like patterns and variable immunoreactivity with SALL4 and neuroendocrine markers. In some cases, the neuroectodermal component was focal while in others it was exclusive. In one case, the focal neuroectodermal component within an ovarian Sertoli Leydig cell tumour resulted in extraovarian metastasis. In reporting these cases, we suggest that neuroectodermal elements, including pure neuroectodermal tumours, are part of the morphological spectrum of DICER1-associated neoplasms.
