BACKGROUND Clinical relapse in acute myeloid leukemia (AML) is associated with the reduced treatment response of leukemia stem cells (LSCs). This study aimed to investigate the effects of the ginseng derivative, ginsenoside Rg1 (Rg1), on CD34+CD38- LSCs derived from KG1a human acute myeloid leukemia cells. MATERIAL AND METHODS CD34+CD38- LSCs were isolated from KG1a human acute myeloid leukemia cells by cell sorting. CD34+CD38- KG1alpha LSCs were divided into the control group and the Rg1 group (treated with Rg1). The cell counting kit-8 (CCK-8) assay evaluated the proliferation of CD34+CD38- KG1alpha LSCs and flow cytometry studied the cell cycle. The mixed colony-forming unit (CFU-Mix) assay and staining for senescence-associated beta-galactosidase (SA-ß-Gal) evaluated cell senescence. Expression of sirtuin 1 (SIRT1) and tuberous sclerosis complex 2 (TSC2) were evaluated using Western blot and quantitative reverse transcription-polymerase chain reaction (qRT-PCR). RESULTS CD34+CD38- KG1alpha LSCs were isolated at 98.72%. Rg1 significantly reduced the proliferation of CD34+CD38- KG1alpha LSCs compared with the control group (p less then 0.05). Cells in the G0/G1 phase were significantly increased, and cells in the G2/M and S phase were significantly reduced compared with the control group (p less then 0.05). Rg1 significantly increased SA-ß-Gal and reduced CFU-Mix formation compared with the control group (p less then 0.05), significantly down-regulated SIRT1 expression in CD34+CD38- KG1alpha LSCs compared with the control group (p less then 0.05), and significantly reduced TSC2 expression in CD34+CD38- KG1alpha LSCs compared with the control group (p less then 0.05). CONCLUSIONS Rg1 inhibited cell proliferation and induced cell senescence markers in CD34+CD38- KG1alpha LSCs by activating the SIRT1/TSC2 signaling pathway.Hypophysitis, which is often accompanied by pituitary dysfunction, is classified into several subtypes based on the cause, histology, and the location of inflammation in the pituitary gland. A definitive diagnosis requires pituitary biopsy, which is invasive, and the process is limited to specialized clinical settings. In this opinion paper, we review the literature associated with hypophysitis, and provide the guidelines of the Japan Endocrine Society for the diagnosis and treatment of autoimmune and IgG4-related hypophysitis.Pneumonia associated with the 2019 novel coronavirus (2019-nCoV) is continuously and rapidly circulating at present. No effective antiviral treatment has been verified thus far. We report here the clinical characteristics and therapeutic procedure for four patients with mild or severe 2019-nCoV pneumonia admitted to Shanghai Public Health Clinical Center. All the patients were given antiviral treatment including lopinavir/ritonavir (Kaletra®), arbidol, and Shufeng Jiedu Capsule (SFJDC, a traditional Chinese medicine) and other necessary support care. After treatment, three patients gained significant improvement in pneumonia associated symptoms, two of whom were confirmed 2019-nCoV negative and discharged, and one of whom was virus negative at the first test. The remaining patient with severe pneumonia had shown signs of improvement by the cutoff date for data collection. Results obtained in the current study may provide clues for treatment of 2019-nCoV pneumonia. The efficacy of antiviral treatment including lopinavir/ritonavir, arbidol, and SFJDC warrants further verification in future study.The aim of this study was clarify the effects of reducing various functional pressures essential for the maintenance of bone homeostasis. Femoral bone mineral density (BMD) and biological apatite (BAp) crystallite alignment were measured in conventionally reared and hindlimb-unloaded mice. The femur was divided into 10 equal segments perpendicular to the longitudinal axis of the bone and measurements were performed on the cortical bone in the five segments closest to the midpoint of the femur. Significantly lower BMD and BAp alignment in the longitudinal (Z-axis) direction were observed in the hindlimb-unloaded group. The present findings suggest that unloading by tail suspension significantly decreases not only mouse femoral bone mass but also BAp crystallite alignment, although minimal uniaxial preferential alignment is retained.Additive manufacturing (AM) processes are increasingly used in dentistry. The underlying process is the joining of material layer by layer based on 3D data models. Four additive processes (laser stereolithography, polymer jetting, digital light processing, fused deposition modeling) are mainly used for processing dental polymers. The number of polymer materials that can be used for AM in dentistry is small compared to other areas. Applications in dentistry using AM are limited (e.g. study models, maxillo-facial prostheses, orthodontic appliances etc.). New and further developments of materials are currently taking place due to the increasing demand for safer and other applications. Biocompatibility and the possibility of using materials not only as temporarily but as definitive reconstructions under oral conditions, mechanically more stable materials where less or no post-processing is needed are current targets in AM technologies. TW37 Printing parameters are also open for further development where optical aspects are also important.The aim was to assess the effect of powder to liquid ratio (PLR) on setting time, fluoride release, and compressive strength of conventional glass ionomer cements (GICs) containing pre-reacted spherical glass fillers (SPG). GICs were prepared by mixing SPG with Fuji IX Universal liquid using PLR of 11, 1.51, 21, 2.51, 31. Setting time decreased from 221 to 51 s upon rising PLR. Increasing PLR decreased cumulative fluoride release (33 to 13 ppm). Diffusion coefficient of fluoride of experimental GICs (1.6-1.8×10-8 cm2/s) was comparable with that of Fuji IX Universal (1.6×10-8 cm2/s). Compressive strength of PLR 21 to 31 (93-140 MPa) were comparable with that of Fuji IX Universal (124 MPa). These results demonstrated that rising powder ratio reduced setting time, fluoride release, and compressive strength of GICs. However, the setting time and strength experimental GICs with PLR greater than 21 were in the acceptable range of the ISO standard.
