The sex- and age-related differences in the composition of iliofemoral atherosclerotic plaques are largely unknown. Therefore, the aim of the current study is to gain insight into plaque composition across strata of age and sex in a large cohort of vascular surgery patients.
Peripheral atherosclerotic plaques of patients who underwent iliofemoral endarterectomy (n=790) were harvested between 2002 and 2014. The plaques were semi-quantitatively analyzed for the presence of lipid cores, calcifications, plaque hemorrhages (PH), collagen, macrophage and smooth muscle cell (SMC) content, and quantitatively for microvessel density. Ageing was independently associated with rupture-prone iliofemoral plaque characteristics, such as higher prevalence of plaque calcifications (OR 1.52 (95%CI1.03-2.24) p=0.035) and PH (OR 1.46 (95%CI1.01-2.09) p=0.042), and lower prevalence of collagen (OR 0.52 (95%CI0.31-0.86) p=0.012) and SMCs (OR 0.59 (95%CI0.39-0.90) p=0.015). Post-mortem interval (PMI) is crucial for death scene investigations. However, estimating PMI is challenging in cases involving significantly decomposed or destroyed bodies, such as those involving skeletonized remains. In this study, 71 bones (patella) were collected from the bodies during autopsies (PMI ranging from 1 day to 2 years). As the let-7e and miR-16 miRNAs were used as internal controls for the bone tissue in previous studies, these miRNAs were selected as targets to estimate PMI. The miRNA Ce_miR-39_1 was used as a spike-in internal control to normalize the target miRNA levels. Real-time quantitative reverse transcription polymerase chain reaction was performed to correlate the expression levels of let-7e and miR-16 with increasing PMI. A negative correlation was observed between miRNA expression and increasing PMI. The expression of both let-7e and miR-16 was observed to be significantly different between group A and the other PMI groups (group A 6 months). In conclusion, these data suggest that the expression level of specific miRNAs (let-7e and miR-16) in the bone tissue could be used to estimate PMI. However, more studies using long-term PMI samples are required to further corroborate these findings.In this study, a rabbit endometritis model was developed to study cow endometritis. In addition, the protective effects of baicalin (a flavonoid) against endometritis were investigated. Clinical symptoms, differential leukocyte counting, uterine secretion smear microscopy and chemical examination, urine testing, and signs of necropsy showed abnormal changes and inflammatory responses in the uterus of rabbits. Histopathological results revealed visible inflammatory exudates and blood spots between intercellular spaces which confirmed that the rabbit endometritis model was successfully developed. Most importantly, these inflammatory signs were partially attenuated with baicalin treatment. The data revealed that the increased body temperature and leukocyte cells, pus, and the detachment of epithelial cells were alleviated with baicalin administration in a dose-dependent manner. Histopathological tissue changes such as inflammatory cells infiltrates, hyperemia, hemorrhages, and shedding of epithelial cells were partially attenuated with baicalin treatment. In addition, the mRNA expression of inflammation-related genes (iNOS, IL-1β, TNF-α, IL-10, IL-4, and IL-6) was significantly altered in RAW264.7 cells after LPS treatment. Further, the phosphorylated protein expression of JNK, p65, and IκBα were significantly reduced with LPS treatment. Intriguingly, baicalin pretreatment reversed the alteration in mRNA expression of inflammation-related genes and significantly reduced the phosphorylation of JNK, p65, and IκBα. In summary, our results suggest that baicalin has protective effects on bacterial-induced endometritis in rabbits that involve the suppression of NF-κB and JNK signaling pathways and pro-inflammatory cytokines.
