To investigate the characteristics and risk factors for neurologic adverse events (AEs) induced by immune checkpoint inhibitors (ICIs).
An observational, retrospective, and pharmacovigilance study based on the FAERS database collected between January 2014 and December 2019 was conducted. ICI-related AEs were defined as adverse reactions in patients using anti-PD-1 (nivolumab and pembrolizumab), anti-PD-L1 (atezolizumab, avelumab, and durvalumab), and anti-CTLA-4 (ipilimumab and tremelimumab). Neurologic AEs previously reported to be associated with ICI were evaluated in the disproportionality analysis using the reporting odds ratio (ROR).
Among 50,406 ICI-related reports, 3619 (7.2%) neurological case was found 1985 with anti-PD-1, 372 with anti-PD-L1, 366 with anti-CTLA-4, and 896 with the combination of ICIs. Here, we investigated the decontamination of citrus fruits to test the antifungal potential of a novel non-thermal gas plasma apparatus, termed a roller conveyer plasma instrument. This instrument generates an atmospheric pressure dielectric barrier discharge (APDBP) plasma on a set of rollers. Penicillium venetum was spotted onto the surface of the fruit or pericarps, as well as an aluminium plate to act as a control, before performing the plasma treatment. The results showed that viable cell number of P. venetum decreased with a decimal reduction time (D value or estimated treatment time required to reduce viable cell number by 90%) of 0.967 min on the aluminium plate, 2.90 min and 1.88 min on the pericarps of 'Kiyomi' (Citrus unshiu × C. sinensis) and 'Kawano-natsudaidai' (C. natsudaidai) respectively, and 2.42 min on the surface of 'Unshu-mikan' (C. unshiu). These findings confirmed a fungicidal effect of the plasma not only on an abiotic surface (aluminium plate) but also on a biotic surface (citrus fruit). The expression of miR-1225 was investigated in breast cancer tissues and cells by quantitative real-time PCR. The role of miR-1225 in the cell process of OS was analyzed by CCK-8 assay and Transwell assay. The prognostic value of miR-1225 was evaluated by Kaplan-Meier survival curves and Cox regression analysis. miR-1225 was significantly upregulated in breast cancer tissues, which was associated with the TNM stage of breast cancer patients. The prognosis of patients with high miR-1225 expression was worse than that of patients with low miR-1225 expression, which indicated that miR-1225 acted as an independent factor for the prognosis of breast cancer. Additionally, the upregulation of miR-1225 promoted cell proliferation, migration, and invasion of breast cancer, which suggested miR-1225 might be involved in the progression of breast cancer. JAK1 was identified as the direct target of miR-1225, which was also involved in cell proliferation, migration, and invasion of breast cancer. click here The overexpression of miR-1225 in breast cancer indicates a poor prognosis of patients and promotes the progression of breast cancer by targeting JAK1. miR-1225 may be a biomarker and therapeutic target for the treatment of breast cancer.
Adjuvant chemotherapy after curative resection is established as a standard therapy for colon and rectal cancer. Although the efficacy of adjuvant chemotherapy has been shown by pooled analyses from randomized controlled trials, elderly patients still receive adjuvant chemotherapy less frequently than younger patients. In this systematic review and meta-analysis, we aimed to assess the survival benefit of adjuvant chemotherapy in elderly patients based on observational studies in which the elderly patients would likely be representative of those encountered in real-world clinical settings.
A comprehensive literature search was conducted using PubMed, Scopus, and the Cochrane Central Register of Controlled Trials. Observational studies that investigated the survival benefit of adjuvant chemotherapy after curative resection in elderly patients (age ≥ 70years) with colon or rectal cancer were included. The 5-year overall survival (OS) rate and OS were assessed. Risk of bias was assessed using the ROBINS-I tool.
Eleven studies in elderly patients with colon cancer were included. No relevant study was identified for rectal cancer. Elderly patients who received adjuvant chemotherapy had a significantly higher 5-year OS rate than those who did not (risk ratio 1.51, 95% confidence interval 1.29-1.76, P < 0.001). There was also a significant improvement in OS in elderly patients who received adjuvant chemotherapy (hazard ratio 0.59, 95% confidence interval 0.53-0.66, P < 0.001). The overall risk of bias was judged to be critical for both outcomes.
