TC release was significantly enhanced in acidic pH media (pH 3.6 and 5.6) indicating pH-dependent release from nanohybrid microspheres; releasing 80-100 % of the loaded drug in 4-14 days. Drug/polymer interactions and molecular structures were investigated by FT-IR spectroscopy and DSC analysis. According to the results obtained, enzymatically synthesized nanohybrids have potential for pH-dependent release of the model drug, trans-chalcone.In this study, a novel method of coupling phytohormones with saline wastewater was proposed to drive efficient microalgal lipid production. All the six phytohormones effectively promoted microalgae growth in saline wastewater, and further increased the microalgal lipid content based on salt stress, so as to achieve a large increase in microalgal lipid productivity. Among the phytohormones used, abscisic acid had the most significant promoting effect. Under the synergistic effect of 20 g/L salt and 20 mg/L abscisic acid, the microalgal lipid productivity reached 3.7 times that of the control. Transcriptome analysis showed that differentially expressed genes (DEGs) of microalgae in saline wastewater were mainly up-regulated under the effects of phytohormones except brassinolide. Common DEGs analysis showed that phytohormones all regulated the expression of genes related to DNA repair and substance synthesis. In conclusion, synergistic effect of salt stress and phytohormones can greatly improve the microalgal lipid production efficiency.In this study, organic acids were demonstrated as a promising carbon source for bisabolene production by the non-conventional yeast, Rhodosporidium toruloides, at microscale with a maximum titre of 1055 ± 7 mg/L. A 125-fold scale-up of the optimal process, enhanced bisabolene titres 2.5-fold to 2606 mg/L. Implementation of a pH controlled organic acid feeding strategy at this scale lead to a further threefold improvement in bisabolene titre to 7758 mg/L, the highest reported microbial titre. Finally, a proof-of-concept sequential bioreactor approach was investigated. Firstly, the cellulolytic bacterium Ruminococcus flavefaciens was employed to ferment cellulose, yielding 4.2 g/L of organic acids. R. toruloides was subsequently cultivated in the resulting supernatant, producing 318 ± 22 mg/L of bisabolene. This highlights the feasibility of a sequential bioprocess for the bioconversion of cellulose, into biojet fuel candidates. Future work will focus on enhancing organic acid yields and the use of real lignocellulosic feedstocks to further enhance bisabolene production.The aerobic denitrification performance of actinomycetes was investigated. Two strains of actinomycetes were isolated and identified as Streptomyces sp. LJH-12-1 and Streptomyces diastatochromogenes LJH-12-2. Strain LJH-12-1 could remove 94% of organic carbon and 91% of total nitrogen. Meanwhile, strain LJH-12-2 could reduce 96% of organic carbon and 93% of total nitrogen. Two strains of actinomycetes revealed excellent carbon source metabolism activity. Moreover, the total nitrogen removal efficiencies were 69%, and 54%, respectively for strains LJH-12-1, and LJH-12-2 during the micro-polluted landscape raw water treatment. Futhermore, strains LJH-12-1 and LJH-12-2 could utilize aromatic proteins, soluble microbial products, and humic acid to drive aerobic denitrification processes in the landscape water bodies. These results will provide a new insight into applying aerobic denitrification actinomycetes to treat micro-polluted water bodies.Thraustochytrids are the most prominent source of polyunsaturated fatty acids, specifically docosahexaenoic acid (DHA). Downstream processing constitutes a significant fraction of total production cost and thus needs judicious optimization. Currently, hazardous solvent-based extraction methods are used to extract oil from the dry or wet thraustochytrids cell mass. The process is also highly energy-intensive due to involvement of dewatering and drying as unit operations. Current work devised an energy-efficient acid-assisted extraction (AAE) methodology to overcome dry and wet biomass-based extraction limitations. AAE recovered 91 % of total oil with 35-40 % PUFA from the direct fermentation broth, eliminating the need for dewatering and drying of fermentation broth/cell biomass. The current work also presents an all-inclusive comparison of the energy assessment of oil extraction from dry and AAE method. AAE produced PUFA enriched oil with a total energy consumption of 210 MJ/kg, which was four times lower than that of conventional dry cell extraction methodology.Naturally occurring, microbial contaminants were found in plant biomasses from common bioenergy crops and agricultural wastes. Unexpectedly, indigenous thermophilic microbes were abundant, raising the question of whether they impact thermophilic consolidated bioprocessing fermentations that convert biomass directly into useful bioproducts. Candidate microbial platforms for biomass conversion, Acetivibrio thermocellus (basionym Clostridium thermocellum; Topt 60 °C) and Caldicellulosiruptor bescii (Topt 78 °C), each degraded a wide variety of plant biomasses, but only A. thermocellus was significantly affected by the presence of indigenous microbial populations harbored by the biomass. Indigenous microbial growth was eliminated at ≥75 °C, conditions where C. bescii thrives, but where A. thermocellus cannot survive. Therefore, 75 °C is the thermophilic threshold to avoid sterilizing pre-treatments on the biomass that prevents native microbes from competing with engineered microbes and forming undesirable by-products. Thermophiles that naturally grow at and above 75 °C offer specific advantages as platform microorganisms for biomass conversion into fuels and chemicals.Pine needles (PNs) are one of the largest bio-polymer produced worldwide. Its waste, i.e., fallen PNs, is mostly responsible for forest fires and is a major challenge. In present article, we have reviewed differenteffortsmadeto tackle this situation. PNs have been used in various fields such asin composite, water purification industries,electronic devices, etc. Gasification is one of the appealing processes for turning PNs into bio-energy; pyrolysis technique has been employed to create various carbon-based water purification materials; saccharification combined with fermentation produced good yields of bio-ethanol; Pd or Ni/PNs biocatalyst showed good catalytic properties in variousreactionsand pyrolysis with or without catalyst is an alluring technique to prepare bio-fuel. Pargyline in vitro Nano cellulose extracted from PNs showed appealing thermal and mechanical strength. The air quality of nearbyenvironment was examinedby studying the magnetic properties of PNs. Packing materials made of PNs showed exceptional ethylene scavenging abilities.Short-chain fatty acids (SCFAs) recovery through anaerobic fermentation is a promising technology to achieve carbon-neutral in waste activated sludge (WAS) management. After 0.15 g CaO2/g volatile suspended solids (VSS) addition and three-cycle freezing co-pretreatments, the maximal SCFAs production of 438.5 mg COD/g VSS was achieved within 4 days fermentation, and more than 70 % of SCFAs was composed of acetate and propionate, which achieved a higher level than reported in previous studies. Mechanism explorations elucidated that co-pretreatment triggered sludge solubilization, promoting the release of biodegradable organics, providing more biodegradable substrates for SCFAs generation. Further microbial community analysis indicated that the abundances of hydrolytic microorganisms and acidogens were enriched, whereas methanogens were inhibited. Besides, environmental analysis suggested that co-pretreatment could achieve carbon reduction benefits of 0.116-0.291 ton CO2/ton WAS, demonstrating its huge carbon-neutral potential benefits.The pre-adsorption of non-catalytic/blocking proteins onto the lignin component of pretreated biomass has been shown to significantly increase the effectiveness of subsequent enzyme-mediated hydrolysis of the cellulose by limiting non-productive enzyme adsorption. Layer-by-layer adsorption of non-catalytic proteins and enzymes onto lignin was monitored using Quartz Crystal Micro balancing combined with Dissipation monitoring (QCM-D) and conventional protein adsorption. These methods were used to assess the interaction between soft/hardwood lignins, cellulases and the three non-catalytic proteins BSA, lysozyme and ovalbumin. The QCM-D analysis showed higher adsorption rates for all of the non-catalytic proteins onto the lignin films as compared to cellulases. This suggested that the "blocking" proteins would preferentially adsorb to the lignin rather than the enzymes. Pre-incubation of the lignin films with blocking proteins resulted in reduced adsorption of cellulases onto the lignin, significantly enhancing cellulose hydrolysis.One-carbon (C1) compounds have emerged as a key research focus due to the growth of metabolic engineering and synthetic biology as affordable and sustainable nonfood sugar feedstocks for energy-efficient and environmentally friendly biomanufacturing. This paper summarizes and discusses current developments in C1 compounds for biomanufacturing. First, two primary groups of microbes that use C1 compounds (native and synthetic) are introduced, and the traits, categorization, and functions of C1 microbes are summarized. Second, engineering strategies for C1 utilization are compiled and reviewed, including reconstruction of C1-utilization pathway, enzyme engineering, cofactor engineering, genome-scale modeling, and adaptive laboratory evolution. Third, a review of C1 compounds' uses in the synthesis of biofuels and high-value compounds is presented. Finally, potential obstacles to C1-based biomanufacturing are highlighted along with future research initiatives.Herein, a mild fractionation method by employing polyol-based Brønsted acidic DESs (BDESs) was proposed to extract lignin with well-preserved β-O-4 substructures and to enhance fermentable sugar yields simultaneously. For ethylene glycol-oxalic acid (EG-OA), more than 53 % of lignin was obtained and superb carbohydrate digestibility (i.e., glucose and xylose yields were reached to 94.6 % and 87.7 %, respectively) was achieved after pretreatment. Remarkably, detailed structural studies revealed that the polyol was incorporated into lignin, which stabilized reactive carbocation intermediates formed during BDESs treatment and prevented undesired recondensation reactions. This lignin protection mechanism was shown to play a key role in enzymatic hydrolysis enhancement and lignin valorization. The resultant β-O-4 linkage-rich lignin fractions were attractive for natural sunscreen applications due to their lighter color and excellent UV-blocking performance. Overall, this work proposed a sustainable and economically practical lignin-first biorefinery approach that is beneficial for achieving comprehensive valorization of lignocellulose.Several types of molecular therapy have become a novel opportunity in the precision treatment of hereditary neuromuscular disorders. This cursive review of gene therapy in hereditary myopathies will focus on selected current phase 1 to 3 trials of common adult hereditary myopathies such as Becker muscular dystrophy, facioscapulohumeral muscular dystrophy, calpainopathy, and dysferlinopathy. The treatment options for Pompe disease serve as an example for hereditary metabolic myopathies.
