One of the characteristic features of aging is the progressive loss of muscle mass, a nosological syndrome called sarcopenia. It is also a pathologic risk factor for many clinically adverse outcomes in older adults. Therefore, delaying the loss of muscle mass, through either boosting muscle protein synthesis or slowing down muscle protein degradation using nutritional supplements could be a compelling strategy to address the needs of the world's aging population. Here, we review the recently identified properties of docosahexaenoic acid (DHA). It was shown to delay muscle wasting by stimulating intermediate oxidative stress and inhibiting proteasomal degradation of muscle proteins. Both the ubiquitin-proteasome and the autophagy-lysosome systems are modulated by DHA. Collectively, growing evidence indicates that DHA is a potent pharmacological agent that could improve muscle homeostasis. Better understanding of cellular proteolytic systems associated with sarcopenia will allow us to identify novel therapeutic interventions, such as omega-3 polyunsaturated fatty acids, to treat this disease.Copper nanoclusters (Cu NCs) are generally formed by several to dozens of atoms. Because of wide range of raw materials and cheap prices, Cu NCs have attracted scientists' special attention. However, Cu NCs tend to undergo oxidation easily. Thus, there is a dire need to develop a synthetic protocol for preparing fluorescent Cu NCs with high QY and better stability. Herein, we report a one-step method for preparing stable blue-green fluorescent copper nanoclusters using glutathione (GSH) as both a reducing agent and a stabilizing agent. High-resolution transmission electron microscopy (HRTEM), X-ray photoelectron spectroscopy (XPS) and electrospray ionization mass spectrometer (ESI-MS) were used to characterize the resulting Cu NCs. The as-prepared Cu NCs@GSH possess an ultrasmall size (2.3 ± 0.4 nm), blue-green fluorescence with decent quantum yield (6.2%) and good stability. MTT results clearly suggest that the Cu NCs@GSH are biocompatible. After incubated with EB-labeled HEK293T cells, the Cu NCs mainly accumulated in nuclei of the cells, suggesting that the as-prepared Cu NCs could potentially be used as the fluorescent probe for applications in cellular imaging.Chronic hepatitis B virus (HBV) infection is a major risk factor for the development of hepatocellular carcinoma (HCC), the leading cause of cancer-related death worldwide. Despite progress in the prevention and therapy of HCC, high incidence and recurrence rates of HCC remain big threats, resulting in poor patient survival. Effective biomarkers and targets of HCC are therefore urgently needed for better management and to improve patient outcomes. Pre-S mutants have been well demonstrated as HBV oncoproteins that play important roles in HCC development through activation of multiple oncogenic signal pathways in hepatocytes, in vitro and in vivo. The presence of pre-S mutants in patients with chronic HBV infection and HBV-related HCC has been associated with a significantly higher risk of HCC development and recurrence after curative surgical resection, respectively. For this purpose, measurements of nitric oxide (NO), peroxynitrite (ONOO-), protein S-nitrosylation and nitration, and nitrosoglutathione reductase (GSNOR) in A. thaliana roots from 1 day to 15 days post-inoculation (dpi) were performed. The cyst nematode infection caused generation of NO and ONOO- in the infected roots. These changes were accompanied by an expansion of S-nitrosylated and nitrated proteins. The enzyme activity of GSNOR was decreased at 3 and 15 dpi and increased at 7 dpi in infected roots, whereas the GSNOR1 transcript level was enhanced over the entire examination period. The protein content of GSNOR was increased in infected roots at 3 dpi and 7 dpi, but at 15 dpi, did not differ between uninfected and infected roots. The protein of GSNOR was detected in plastids, mitochondria, cytoplasm, as well as endoplasmic reticulum and cytoplasmic membranes. We postulate that RNS metabolism plays an important role in plant defense against the beet cyst nematode and helps the fine-tuning of the infected plants to stress sparked by phytoparasitic nematodes.The idea of what is natural has particular relevance in the thoroughbred racing and breeding discourse. It guides breeding regulations; influences how the thoroughbreds' behaviour is perceived and has implications for husbandry, handling, training and racing practices. This study investigates how key industry and animal advocacy informants based in the US, Australia and the UK conceptualise naturalness within the context of common racing practices that potentially impact the horses' welfare. The informants were interviewed using semi-structured interviewing and photo-elicitation. Four common images of thoroughbreds on race day were presented to elicit the informants' responses. Differences emerged between how the two groups tended to describe the images and the role naturalness played in their conceptualisations. The findings were analysed using an updated version of the Layers of Engagement with Animal Protection developed by Bergmann to situate the informants' conceptualisations of naturalness within the wider thoroughbred protection discourse. In conclusion, the industry informants tended to defend the status quo of common racing practices. They tended to naturalise and normalise these practices and downplay their welfare impact. This poses risks for thoroughbred welfare, which are amplified by misrepresentations of what is natural. With the public's understanding of welfare and racing practices growing, racing's legitimacy may be further questioned. Opportunities to leverage the potential of the notion of naturalness for thoroughbred protection are discussed.
