Despite being a major cause of blindness in individuals of 65 years of age and older, some forms of AMD, including GA, still lack targeted treatment. Our previous study demonstrated that mini-αA peptide, which contains the functional site of αA-crystallin, protected RPE cells from NaIO3 -induced apoptosis. METHODS To further investigate the underlying mechanism, we applied next-generation sequencing analysis to identify miR-1246 as a putative mediator of mini-αA protective function. To investigate the role of miR-1246 in RPE cell apoptosis, a stable miR-1246-low-expression cell line was established by using miR-1246 inhibitor. A 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay was used to investigate the proliferation of RPE cells, mRNA and miR-1246 expression were detected by the quantitative reverse transcriptase-polymerase chain reaction. RESULTS We have further identified caspase-3 and caspase-14 as molecular targets of miR-1246 involved in regulation of apoptosis in NaIO3 -incubated cells. Interestingly, disruption of miR-1246 expression enhanced anti-apoptotic effect of mini-αA on RPE cells during oxidative stress. CONCLUSIONS Our results provide a mechanistic basis for evaluation of miR-1246 as a new candidate target for the clinical treatment of AMD. © 2020 Royal Australian and New Zealand College of Ophthalmologists.In this paper, a convenient reverse-phase microemulsion method for the synthesis of SiO2 nanoparticles (NPs) by simply introducing the chitosan and fluorescent dye of lucigenin during the formation reaction of SiO2 NPs was proposed. Addition of chitosan can make the SiO2 NPs porous, and increases lucigenin molecule incorporation into chitosan/SiO2 NPs nanopores based on electrostatic interaction and supermolecular forces. Therefore, fluorescence quantum yield of the lucigenin/chitosan/SiO2 composite nanoparticles was increased by introduction of chitosan and compared with lucigenin/SiO2 NPs without chitosan. Because the number of negative charges carried when using single-stranded DNA (ssDNA) was different from that of double-stranded DNA (dsDNA), the numbers of lucigenin/chitosan/SiO2 composite nanoparticles with positive charge adsorbed using ssDNA or dsDNA were different. Consequently, fluorescence intensity caused using ssDNA or dsDNA/miRNA was clearly discriminative. With increase in target DNA/miRNA concentration, the difference in fluorescence intensity also increased, resulting in a good linear relationship between fluorescence intensity sensitizing value and target miRNA concentrations. Therefore, a new fluorescence analysis method for direct detection of let-7a in human gastric cancer cell samples without enzyme, label free and no immobilization was established using lucigenin/chitosan/SiO2 composite nanoparticles as a DNA hybrid indicator. The proposed method had high sensitivity and selectivity, low cost and the detection limit was 10 fM (S/N = 3). © 2020 John Wiley & Sons, Ltd.OBJECTIVE To assess the influence of the 2012 USPSTF recommendation against PSA-based screening on oncological and functional outcomes following RALP. MATERIALS AND METHODS We retrospectively analyzed patients that underwent RALP between 2008-2018 with a minimum of 12 months follow up from a prospectively collected IRB approved database. The impact USPSTF statement against PSA screening on our surgical outcomes was assessed using a logistic regression model using two groups indicating patients treated before/after the USPSTF statement and time trends for each successive year. RESULTS The mean preoperative PSA increased from 6.0 to 7.4 ng/ml after the USPSTF recommendations. We detected statistically significant time-trend changes after 2012 including an increase in the positive slope of Gleason ≥3+4 or ≥pT3. We detected a fall in bilateral FNS and increase in PNS. Total PSM rate after the USPSTF recommendation increased. However, PSM rates pertinent to each pathological stage did not change significantly after 2012. There was a significant negative trend change in the postoperative 12-month continence and potency indicating a breakpoint in functional outcomes after 2012. We detected a 1.7 fold increase in 12-month BCR rates. The 12-month BCR, potency and continence rates were maintained in young (22 and low volume disease. CONCLUSION Since the USPSTF's recommendation in 2012, we have seen a significant increase in the incidence of the high-risk disease that has forced us to modify our approach to the procedure and the grade of NS leading to a wider resection to maintain the PSM has led to a decrease in postoperative functional recovery. Patients with good characteristics have performed well before and after the USPSTF's recommendation, implying that the quality of surgery did not change over time. Here, we develop a facile, general, and high throughput solvent-free self-assembly strategy to synthesize a series of MMOs including single-component MMOs and multi-component MMOs (e.g., doped MMOs, composite MMOs, and polymetallic oxide) with high crystallinity and remarkable porous properties by grinding and heating raw materials. Compared with the traditional solution self-assembly process, the avoidance of solvents in this method not only greatly increases the yield of target products and synthesis efficiency, but also reduces the environmental pollution and the consumption of cost and energy.
