The aim of this study is to investigate the expression of the PD-1/PD-L1 pathway and CTLA-4 in the tumor microenvironment of epSCCs to assess the feasibility of an immunotherapeutic approach. Twenty equine epithelial tumors were retrospectively selected and submitted to RT-qPCR for PD-1 and PD-L1 genes. After testing antibodies cross-reactivity by western blotting, immunohistochemistry for PD-L1 and CTLA-4 was performed. Results from RT-qPCR demonstrated that 3/20 cases expressed the PD-L1 gene, whereas the PD-1 gene was not detected. Immunohistochemical positivity for PD-L1 was found only in one case. CTLA-4-positive cells were observe in all cases but were few (Mdn = 4.8; IQR = 2.3-7.1 cells/HPF). In this study group, PD-1/PD-L1 and CTLA-4 do not appear to be highly expressed and therefore the use of ICI in epSCCs may not have promising rates of response.This study aimed to assess the welfare of Tunisian sheep in extensive sheep production systems using animal-based measures of ewe welfare. This study encompasses the first national survey of sheep welfare in which animal-based outcomes were tested. Animal-based welfare measures were derived from previous welfare protocols. Fifty-two Tunisian farms were studied and a number from 20 to 100 animals by flock were examinated. The whole flock was also observed to detect clinical diseases, lameness, and coughing. The human-animal relationship was selected as welfare indicators. It was evaluated through the avoidance distance test. The average avoidance distance was 10.47 ± 1.23 and 8.12 ± 0.97 m for a novel person and farmer, respectively. The global mean of body condition score (BCS) was 2.4 with 47% of ewes having a BCS of two, which may be associated with an increased risk of nutritional stress, disease, and low productivity. Ten farms had more than 7% of lambs with a low body condition score, which may be an indication of a welfare problem. The results obtained in the present study suggest that the used animal-based measures were the most reliable indicators that can be included in welfare protocols for extensive sheep production systems.As the preference of consumers for casein products has increased, the protein content of milk from dairy cows is drawing more attention. Protein synthesis in the milk of dairy cows requires a proper supply of dietary protein. High protein supplementation may help to produce more milk protein, but residues in feces and urine cause environmental pollution and increase production costs. As such, previous studies have focused on protein supplements and amino acid (AA) supply. This review concerns AA nutrition for enhancing milk protein in dairy cows, and mainly focuses on three AAs methionine, lysine, and histidine. AA supplementation for promoting protein synthesis is related to the mammalian target of rapamycin (mTOR) complex and its downstream pathways. Each AA has different stimulating effects on the mTOR translation initiation pathway, and thus manifests different milk protein yields. This review will expand our understanding of AA nutrition and the involved pathways in relation to the synthesis of milk protein in dairy cows.The most common neoplasms in intact female dogs are CMGTs. BmKn-2, an antimicrobial peptide, is derived from scorpion venom and has published anticancer effects in oral and colon human cancer cell lines. Thus, it is highly likely that BmKn-2 could inhibit CMGT cell lines which has not been previously reported. This study investigated the proliferation and apoptotic properties of BmKn-2 via Bax and Bcl-2 relative gene expression in two CMGT cell lines, metastatic (CHMp-5b) and non-metastatic (CHMp-13a). The results showed that BmKn-2 inhibited proliferation and induced apoptosis in the CMGT cell lines. The cell morphology clearly changed and increased apoptosis in a dose dependent of manner. The half maximum inhibitory concentration (IC50) was 30 µg/mL for CHMp-5b cell line and 54 µg/mL for CHMp-13a cell line. The induction of apoptosis was mediated through Bcl-2 and Bax expression after BmKn-2 treatment. In conclusion, BmKn-2 inhibited proliferation and induced apoptosis in both CHMp-5b and CHMp-13a cell lines via down-regulation of Bcl-2 and up-regulation of Bax relative mRNA expression. Therefore, BmKn-2 could be feasible as candidate treatment for CMGTs.Pectins are a group of carbohydrates found in structural parts of terrestrial plants with wide industrial and biomedical applications. This study was designed to investigate the dietary effects of apple pomace-derived pectin (APDP) in rainbow trout (Oncorhynchus mykiss). Four formulated diets were provided with different inclusion levels of APDP for 30 days 0, 5, 10, and 20 g kg-1; referred to as control, P1, P2, and P3, respectively. In this study, 300 fish (3.56 ± 0.007 g) were randomly distributed into twelve fiberglass tanks and fed 3% of their respective body weight four times a day. At the end of the experiment, growth parameters, including weight gain, specific growth rate, and food conversion ratio (FCR) were significantly improved in P1 and P2 treatments compared to those of the other treatments. Results from proximate composition analysis showed that protein content increased, and lipid decreased in the P2 and P3 groups. Serum lysozyme, complement activity, total immunoglobulin levels, and total protein were significantly enhanced in all treatments compared to those of the control group. Gene expression results showed no significant difference in regulation of interleukin-1β (IL-1β); however, up-regulation of lysozyme, interleukin-8 (IL-8), and tumor necrosis factor-α (TNF-α) was observed in both P1 and P2. Unlike lipase, the activity of protease and amylase significantly increased in fish receiving different levels of APDP compared to the control (p less then 0.05). Twelve heat-stressed goats (20.21 ± 2.30 kg) were divided equally into four groups control group (CG, fed the basal diet, Saccharomyces cerevisiae supplemented group (SC, 0.60% Saccharomyces cerevisiae added to the basal diet), Clostridium butyricum supplemented group (CB, 0.05% Clostridium butyricum added to the basal diet), and their combination supplemented group (COM 0.60% Saccharomyces cerevisiae and 0.05% Clostridium butyricum added to the basal diet) and were assigned to a 4 × 3 incomplete Latin square design. The rumen fluid and feces were collected for fermentation parameters and feed digestibility analysis, and animal growth performance was also assessed during all the experiment periods. The results showed that rumen pH, rumen cellulolytic enzymes (avicelase, CMCaes, cellobiase, and xylanase) activities, and the concentrations of rumen total volatile fatty acid (TVFA), acetic acid, and propionic acid were significantly increased with Saccharomyces cerevisiae, Clostridium butyricum, and their combination supplementation (p less then 0.05). Besides, the dry matter intake (DMI), average daily gain (ADG), and the digestibility of dry matter (DM), neutral detergent fiber (NDF), and acidic detergent fiber (ADF) were significantly increased (p less then 0.05) with supplemented these probiotics.
