We report a method for the detection and quantitation of twelve drugs and two metabolites in the same structural class as the illicit mu-opioid agonist U-47700 in human whole blood. These substances are either known or suspected to be present as potential novel opioids in illicit drug markets. The general class of these drugs was developed in pharmaceutical research programs in the 1970's but have recently become of concern for overdoses and death in opioid users in the United States and internationally. The scope of analysis included the following compounds methylenedioxy U-47700, ethylenedioxy U-47700, ethylenedioxy U-51754, U-69593, U-47931E (Bromadoline), U-47700, U-48800, U-49900, U-51754, U-50488, propyl U-47700, and isopropyl U-47700. Additionally, two metabolites, N,N-didesmethyl U-47700, desmethyl U-47700 were also included in the scope. Drugs were extracted from human whole blood using solid phase extraction (SPE) and the extracts were analyzed by liquid chromatography tandem mass spectrometry (LCMSMS). The assay was validated with respect to bias, carryover, interference, and within run and between run precision, and accuracy. Eight medicolegal death investigation cases which had screened positive for U-48800 by liquid chromatography time-of-flight mass spectrometry (LCTOF) were successfully confirmed and quantified using this method. The mean and median concentrations of U-48800 in these cases were 2.5 (±2.1) ng/mL and 1.8 ng/mL, respectively, with a range of concentrations of 0.27-6.2 ng/mL. Case history information including the presence of other drugs in combination are described and discussed.Philadelphia chromosome-negative myeloproliferative neoplasms (MPNs), polycythemia vera, essential thrombocythemia, and primary myelofibrosis, are hematopoietic stem cell disorders that are defined by activating mutations in signal transduction pathways and are characterized clinically by the overproduction of platelets, red blood cells, and neutrophils, significant burden of disease-specific symptoms, and high rates of vascular events. The focus of this review is to critically reevaluate the clinical burden of thrombosis in MPNs, to review the clinical associations among clonal hematopoiesis, JAK2V617F burden, inflammation, and thrombosis, and to provide insights into novel primary and secondary thrombosis-prevention strategies. In fuel cells, the oxygen reduction reaction (ORR) at the cathode plays a crucial role in their performance. High cost, low abundance, catalyst poisoning, and poor durability of the pioneering ORR catalyst Pt make it less desirable for commercial fuel cells. Herein, we demonstrate a greener process to synthesize CeO2 nanostructures by varying reaction parameters in a single-step solvothermal route and provide a detailed mechanism for the formation of CeO2 nanostructures with different shapes. The shape and size of the CeO2 nanostructures such as hollow/solid spheres, triangular flakes, nanotubes, and flower-like structures result in a strong effect on their ORR activity. A normalized effect of effective surface area and oxygen vacancies in CeO2 nanostructures is found to govern the ORR activity order. Among the CeO2 nanostructures, hollow spheres exhibit the best ORR activity with a four-electron reduction pathway. Moreover, they show comparable ORR activity and outstanding stability as well as methanol fuel tolerance and are a substitute for Pt/C.The present study evaluated the impact of different thermal (infrared-drying, hot air-drying and purée pasteurization) and non-thermal (freeze-drying) processing technologies on red-fleshed apple (poly)phenolic compounds. We further investigated the processing effect on the (poly)phenol bioavailability in a crossover postprandial study where three subjects consumed three apple products (freeze-dried snack, hot air-dried snack and pasteurized purée). (Poly)phenolic compounds present in the apple products and their biological metabolites in urine were analyzed using liquid chromatography coupled to mass spectrometry (UPLC-MS/MS). When comparing different processes, infrared-drying caused important losses in most of the apple (poly)phenolics, while hot air-drying and purée pasteurization maintained approximately 83% and 65% of total (poly)phenols compared with the freeze-dried snack, respectively. Anthocyanins in particular were degraded to a higher extent, and hot air-dried apple and pasteurized purée maintained respectively 26% and 9% compared with freeze-dried apple snack. The acute intake showed that pasteurized purée exhibited the highest (poly)phenol bioavailability, followed by hot air-drying and freeze-dried snack, highlighting the impact of processing on (poly)phenols absorption. In conclusion, for obtaining affordable new red-fleshed apple products with enhanced (poly)phenol bioavailability, purée pasteurization and hot air-drying represent viable techniques. However, to obtain a red-fleshed apple snack with high anthocyanin content, freeze-drying is the technique that best preserves them. This study aimed to extend previous research on the experiences and factors that impact law enforcement personnel when working with distressing materials such as child sexual abuse content. A sample of 22 law enforcement personnel working within one law enforcement organisation in England, United Kingdom participated in anonymous semi-structured interviews. Results were explored thematically and organised in the following headings "Responses to the material", "Impact of working with distressing evidence", "Personal coping strategies" and "Risks and mitigating factors". Law enforcement professionals experienced heightened affective responses to personally relevant material, depictions of violence, victims' displays of emotions, norm violations and to various mediums. These responses dampened over time due to desensitisation. The stress experienced from exposure to the material sometimes led to psychological symptoms associated with Secondary Traumatic Stress. see more Job satisfaction, self-care activities, the coping strategies used when viewing evidence, detachment from work outside working hours, social support and reducing exposure to the material were found to mediate law enforcement professionals' resilience. Exposure to distressing material and the risks associated with this exposure were also influenced by specific organisational procedures implemented as a function of the funding available and workload. Recommendations for individual and organisational practices to foster resilience emerged from this research. These recommendations are relevant to all organisations where employees are required to view distressing content.
