To date, the diagnosis of these disorders mainly relied on the recognition of clinical symptoms when neurodegeneration was already at an advanced phase. In recent years, several efforts have been made to develop new diagnostic tools for the early diagnosis of prion diseases. The real-time quaking-induced conversion (RT-QuIC) assay, an in vitro assay that can indirectly detect very low amounts of PrPSc aggregates, has provided a very promising tool to improve the early diagnosis of human prion diseases. Over the decade since RT-QuIC was introduced, the diagnosis of not only prion diseases but also synucleinopathies and tauopathies has greatly improved. One-step nucleic acid amplification (OSNA) analyzes the entire LN by detecting cytokeratin 19 (CK19) mRNA as a surrogate for LN metastases requiring less effort than conventional biomolecular techniques. We aimed to evaluate performance of OSNA in detecting sentinel LN (SLN) metastases in PCa. Methods SLNs (n = 534) of 64 intermediate- or high-risk PCa patients undergoing radical prostatectomy with extended and sentinel-guided lymphadenectomy were cut into slices and alternatingly assigned to OSNA and histopathology (hematoxylin-eosin staining, CK19, and CK AE1/AE3 immunohistochemistry). Sensitivity and specificity of OSNA and concordance and measure of agreement (Cohen's kappa (κ)) between OSNA and histopathology were assessed. Results Histopathology revealed metastases in 76 SLNs. Sensitivity and specificity of OSNA were 84.2% and 96.1%, respectively. Discordant results were recorded for 30 of 534 SLNs, revealing high concordance (94.4%). Twenty-four discordant cases were classified as micrometastases, indicating a possible allocation bias. In 18 cases, positive results were conferred only by OSNA resulting in seven LN-positive patients who were missed by histopathology. Overall, the level of agreement was high (κ = 0.78). Conclusions OSNA provided a diagnosis that was as least as accurate as detailed histological examination and might improve LN staging in PCa.In late 2019, a global pandemic occurred. In this study, we present an analysis on the substances identified in the human metabolome capable of binding the active site of the SARS-CoV-2 main protease (Mpro). The substances present in the human metabolome have both endogenous and exogenous origins. The aim of this research was to find molecules whose biochemical and toxicological profile was known that could be the starting point for the development of antiviral therapies. Our analysis revealed numerous metabolites-including xenobiotics-that bind this protease, which are essential to the lifecycle of the virus. Among these substances, silybin, a flavolignan compound and the main active component of silymarin, is particularly noteworthy. Silymarin is a standardized extract of milk thistle, Silybum marianum, and has been shown to exhibit antioxidant, hepatoprotective, antineoplastic, and antiviral activities. Our results-obtained in silico and in vitro-prove that silybin and silymarin, respectively, are able to inhibit Mpro, representing a possible food-derived natural compound that is useful as a therapeutic strategy against COVID-19.This study aimed to identify the relationships between gender role stereotypes (GRS), patriarchal family environment, and major satisfaction (MS) and their associated factors in nursing students. A total of 195 nursing students (154 women, 41 men) were surveyed online in South Korea from May to June 2020. The Gender Role Stereotype Inventory was used to assess gender role stereotypes, while patriarchal family environment and MS were evaluated using a 11-item instrument for testing patriarchal family environment and the Major Satisfaction Inventory, respectively. Men demonstrated stronger gender role stereotypes and patriarchal family culture than women. Patriarchal family culture was significantly correlated with gender role stereotypes and MS. A multiple regression analysis was performed to identify the effects of age, academic performance, motive for MS, intellectual GRS, social GRS, and patriarchal family environment on MS. The explanatory power of this model was 12.2%. Younger age, higher grade point average, voluntary selection of major, lower intellectual gender stereotypes, and higher social gender stereotypes predicted higher MS. Further studies on cultures in Asia and the world are needed to understand the impact of the patriarchal family culture and gender role stereotypes of nursing students on school adaptation.The present work describes a facile and convenient procedure for synthesizing zinc oxide nanoparticles using luteolin isolated from Eclipta alba plant (L-ZnONPs) at room temperature. The formation of as-grown L-ZnONPs was confirmed by X-ray diffraction analysis (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), high-resolution transmission electron microscopy (HR-TEM), and selected area electron diffraction (SAED). The Wurtzite structure of ZnO was observed by its hexagonal phases in diffraction patterns. The SEM images revealed the different sizes and morphologies of L-ZnONPs, with diameters between 12 and 25 nm. The HR-TEM result showed that the inter-planar distance between two lattice fringes was 0.262 nm, which coincides with the d-spacing of (002) and (101) lattice planes of the as-obtained material. The anticancer activity of L-ZnONPs against the breast cancer cell line MCF-7 was greater as compared to that of luteolin or ZnO alone. The mechanistic evaluation of such an activity carried out using in silico methods suggested that the anti-breast cancer activity of L-ZnONPs was mediated by polo-like kinase 1 (PLK1) proteins.Shallow depth-of-field (DoF), focusing on the region of interest by blurring out the rest of the image, is challenging in computer vision and computational photography. It can be achieved either by adjusting the parameters (e.g., aperture and focal length) of a single-lens reflex camera or computational techniques. In this paper, we investigate the latter one, i.e., explore a computational method to render shallow DoF. The previous methods either rely on portrait segmentation or stereo sensing, which can only be applied to portrait photos and require stereo inputs. To address these issues, we study the problem of rendering shallow DoF from an arbitrary image. In particular, we propose a method that consists of a salient object detection (SOD) module, a monocular depth prediction (MDP) module, and a DoF rendering module. The SOD module determines the focal plane, while the MDP module controls the blur degree. Specifically, we introduce a label-guided ranking loss for both salient object detection and depth prediction. For salient object detection, the label-guided ranking loss comprises two terms (i) heterogeneous ranking loss that encourages the sampled salient pixels to be different from background pixels; (ii) homogeneous ranking loss penalizes the inconsistency of salient pixels or background pixels. For depth prediction, the label-guided ranking loss mainly relies on multilevel structural information, i.e., from low-level edge maps to high-level object instance masks. In addition, we introduce a SOD and depth-aware blur rendering method to generate shallow DoF images. Comprehensive experiments demonstrate the effectiveness of our proposed method.Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the causative agent of novel coronavirus disease 2019 (COVID-19), has become a severe threat to global public health. There are currently no antiviral therapies approved for the treatment or prevention of mild to moderate COVID-19 as remdesivir is only approved for severe COVID-19 cases. Here, we evaluated the antiviral potential of a Propylamylatin formula, which is a mixture of propionic acid and isoamyl hexanoates. The Propylamylatin formula was investigated in gaseous and liquid phases against 1 mL viral suspensions containing 105 PFU of SARS-CoV-2. Viral suspensions were sampled at various times post-exposure and infectious virus was quantified by plaque assay on Vero E6 cells. Propylamylatin formula vapors were effective at inactivating infectious SARS-CoV-2 to undetectable levels at room temperature and body temperature, but the decline in virus was substantially faster at the higher temperature (15 min versus 24 h). The direct injection of liquid Propylamylatin formula into viral suspensions also completely inactivated SARS-CoV-2 and the rapidity of inactivation occurred in an exposure dependent manner. The overall volume that resulted in 90% viral inactivation over the course of the direct injection experiment (EC90) was 4.28 µls. Further investigation revealed that the majority of the antiviral effect was attributed to the propionic acid which yielded an overall EC90 value of 11.50 µls whereas the isoamyl hexanoates provided at most a 10-fold reduction in infectious virus. The combination of propionic acid and isoamyl hexanoates was much more potent than the individual components alone, suggesting synergy between these components. These findings illustrate the therapeutic promise of the Propylamylatin formula as a potential treatment strategy for COVID-19 and future studies are warranted.Increasingly, raw materials of natural origin are used as fillers in polymer composites. Then, they were subjected to a series of tests to assess the impact of the hybrids used on the final product. It has been shown that the addition of optimal amounts of biofillers can, inter alia, increase the tensile strength of the composite, improve damping properties, extend the burning time of the material and affect the course of vulcanization or cross-linking density.
