Regenerative medicine has greatly progressed, but tendon regeneration mechanisms and robust in vitro tendon differentiation protocols remain to be elucidated. Recently, tendon explant co-culture (CO) has been proposed as an in vitro model to recapitulate the microenvironment driving tendon development and regeneration. Here, we explored standardized protocols for production and storage of bioactive tendon-derived secretomes with an evaluation of their teno-inductive effects on ovine amniotic epithelial cells (AECs). Teno-inductive soluble factors were released in culture-conditioned media (CM) only in response to active communication between tendon explants and stem cells (CMCO). Unsuccessful tenogenic differentiation in AECs was noted when exposed to CM collected from tendon explants (CMFT) only, whereas CMCO upregulated SCXB, COL I and TNMD transcripts, in AECs, alongside stimulation of the development of mature 3D tendon-like structures enriched in TNMD and COL I extracellular matrix proteins. Furthermore, although the tenogenic effect on AECs was partially inhibited by freezing CMCO, this effect could be recovered by application of an in vivo-like physiological oxygen (2% O2) environment during AECs tenogenesis. Therefore, CMCO can be considered as a waste tissue product with the potential to be used for the development of regenerative bio-inspired devices to innovate tissue engineering application to tendon differentiation and healing.Gene editing platforms have revolutionized the field of genetics with a direct impact on the public health system. In the Malaysian context, modern biotechnology has raised questions about how to best govern gene editing in regulations, biosafety, and biosecurity. Even though standards and guidelines on stem cell and cell-based therapies have been developed, there are no appropriate legal frameworks available for gene editing yet. Nevertheless, biosafety regulations were established to balance promoting biotechnology and protecting against their potential environmental and human health risks. There is also a need to address the potential of genetically modified organisms (GMOs) as bioweapons. Numerous frameworks from several international organizations may provide valuable input in formulating documents on gene editing. By establishing comprehensive guidelines, legal policies, and standards to tackle the challenges and risks associated with gene editing, Malaysia can successfully apply this modern technology in this country.The sugar alcohols and functional sugars have wide applications in food, pharmaceutical, and chemical industries. However, the smaller quantities of natural occurring sugar alcohols and functional sugars restricted their applications. As there exist reviews about the chemical structures, physicochemical properties, biological functions, applications, and biosynthesis of sugar alcohols and/or functional sugars in Y. lipolytica, this mini review will not only update the recent advances in enzymatic and microbial production of sugar alcohols (erythritol, D-threitol, and xylitol) and functional sugars (isomaltulose, trehalose, fructo-oligosaccharides, and galacto-oligosaccharides) by using recombinant Y. lipolytica but also focus on the studies of gene discovery, pathway engineering, expanding substrate scope, bioprocess engineering, and novel breeding methods to resolve the aforementioned issues.We have employed fullerenes as versatile agents to "cork" the open tips of multiwalled carbon nanotubes (MWCNTs), and as promoting species for the release of the inorganic material filled within the nanotubes' cavities. High Z element compounds, namely, PbI2, ZnI2, and CeI3, were chosen to easily determine the presence of the filler inside the hosting nanotubes by transmission electron microscopy (TEM). Fullerenes can isolate inorganic nanostructures confined within the hollow cavities of MWCNTs, which allows the removal of the external material remnant after the filling. Otherwise, taking advantage of the affinity of fullerenes with selected solvents, we have confirmed the ability of the C60 molecules to promote the displacement of the inorganic guest from the host. We propose two different strategies to trigger the release, employing vapor and liquid phase treatments. The first protocol involves annealing filled MWCNTs in presence of fullerenes (to obtain C60PbI2@MWCNTs) and the subsequent washing of the sample in ethanol under mild conditions. On the other hand, the simultaneous introduction of the C60 molecules and the liberation of the guest are produced by a single step wet procedure; the latter being potentially useful when materials that are not stable at high temperatures are employed for filling.The aim of this study was to compare the aerodynamics of able-bodied and amputee cyclists by computational fluid dynamics. The cyclists' geometry was obtained by a 3D scanner. Three CAD models were created as able-bodied, transtibial (Tt), and transradial (Tr) amputees. Numerical simulations were conducted up to 13 m/s with increments of 1 m/s to assess drag force. The drag ranged between 0.36 and 39.25 N for the able-bodied model, 0.36-43.78 for the Tr model and 0.37-41.39 N for the Tt model. The pressure drag ranged between 0.20 and 22.94 N for the normal model, 0.21-28.61 for the Tr model and 0.23-28.02 N for the Tt model. The viscous drag ranged between 0.16 and 15.31 N for the normal model, 0.15-15.17 for the Tr model and 0.14-13.38 N for the Tt model. The rolling resistance (RR) was higher on the able-bodied (2.23 N), followed by the Tr (2.20 N) and Tt (2.17 N) models. As a conclusion, the able-bodied cyclist showed less drag, followed by the Tt and Tr models, respectively. The RR presented higher values in the able-bodied, followed by the Tr and Tt models.
