In the present study, un-anthropized natural environments and spontaneous processed foods were exploited for wild yeast isolation with the goal of amplifying the knowledge of probiotic aptitudes of different yeast species. For this purpose, 179 yeast species were isolated, identified as belonging to twelve different genera, and characterized for the most important probiotic features. Findings showed interesting probiotic characteristics for some yeast strains belonging to Lachancea thermotolerans, Metschnikowia ziziphicola, Saccharomyces cerevisiae, and Torulaspora delbrueckii species, although these probiotic aptitudes were strictly strain-dependent. These yeast strains could be proposed for different probiotic applications, such as a valid alternative to, or in combination with, the probiotic yeast S. cerevisiae var. boulardii.Cephalopods are very relevant food resources. The common cuttlefish (Sepia officinalis) is highly appreciated by consumers and there is a lack of rapid methods for its authentication in food products. We introduce a new minor groove binding (MGB) TaqMan real-time PCR (Polymerase Chain Reaction) method for the authentication of S. officinalis in food products to amplify a 122 base pairs (bp) fragment of the mitochondrial COI (Cytochrome Oxidase I) region. Reference and commercial samples of S. officinalis showed a threshold cycle (Ct) mean of 14.40, while the rest of the species examined did not amplify, or showed a significantly different Ct (p less then 0.001). The calculated efficiency of the system was 101%, and the minimum DNA quantity detected was 10-4 ng. No cross-reactivity was detected with any other species, thus, the designed method differentiates S. officinalis from other species of the genus Sepia and other cephalopod species and works for fresh, frozen, grilled, cooked and canned samples of Sepia spp. CDK2-IN-4 ic50 The method has proved to be reliable and rapid, and it may prove to be a useful tool for the control of fraud in cuttlefish products.This study aimed to describe metabolic, oxidative, and mineral blood profiles of Saanen does through lactation compared with Mediterranean breed clusters (Maltese and Rossa Mediterranea, and Jonica, Garganica, and Girgentana). Milk and blood samples of 57 dairy goats (9-10 goats per breed) were collected from the 2nd to the 30th week of lactation every 2-3 weeks. Saanen showed greater milk yield and somatic cell score, and lower fat and protein percentage through lactation (p less then 0.05) than the Mediterranean breed clusters. Blood analysis revealed that stage of lactation had a greater impact than breed cluster, except for uric acid, alkaline phosphatase, and aspartate aminotransferase. Plasmatic non-esterified fatty acids indicated a greater negative energy balance in Saanen than the other breed clusters during early and medium lactation stages (p less then 0.05). Serum Cl, Mg, and Ca increased in all the breed clusters from early to the following stages of lactation (p less then 0.05). No significant prooxidant/antioxidant imbalances were detected in any of the three clusters during the entire lactation. In conclusion, Mediterranean breeds tended to recover earlier from negative energy balance than Saanen, but effects of breed or stage of lactation on long-term oxidative stress indicators were not evident.MicroRNA-offset RNAs (moRNAs) are microRNA-like small RNAs generated by microRNA precursors. To date, little is known about moRNAs and bioinformatics tools to inspect their expression are still missing. We developed miR&moRe2, the first bioinformatics method to consistently characterize microRNAs, moRNAs, and their isoforms from small RNA sequencing data. To illustrate miR&moRe2 discovery power, we applied it to several published datasets. MoRNAs identified by miR&moRe2 were in agreement with previous research findings. Moreover, we observed that moRNAs and new microRNAs predicted by miR&moRe2 were downregulated upon the silencing of the microRNA-biogenesis pathway. Further, in a sizeable dataset of human blood cell populations, tens of novel miRNAs and moRNAs were discovered, some of them with significantly varied expression levels among the cell types. Results demonstrate that miR&moRe2 is a valid tool for a comprehensive study of small RNAs generated from microRNA precursors and could help to investigate their biogenesis and function.Italian ryegrass (Lolium multiflorum L.) is an annual grass widely distributed in cultivated crops around the world. The aim of this study was to develop a cohort-based stochastic population dynamics model that integrates both emergence (thermal time) and dynamic population models as a tool to simulate the population dynamics of susceptible and resistant populations of L. multiflorum under the effects of climate change. The current climate scenario and the increase in the average air temperature by 2.5 °C were considered. Chemical and cultural management strategies commonly used in the South Region of Brazil during the winter and summer seasons were incorporated into the model. In the absence of control and under the current climate conditions, the seed bank population grew until reaching an equilibrium density of 19,121 ± 371 seeds m-2 for the susceptible and 20463 ± 363 seeds m-2 for the resistant populations.
