HO happens frequently in severe trauma patients such as in blast injury, central nervous system injury and burn injury, in which excessive endogenous glucocorticoid production has always been found. Glucocorticoids have a big impact on bone and muscle. However, few studies have investigated the impact of glucocorticoids on DC/HO formation in muscle. This study aimed to determine the role of glucocorticoids in DC/HO pathogenesis following muscular injury and the possible underlying mechanism. In this study, we administered a high dose of a synthetic glucocorticoid, dexamethasone (DEX), to animals with muscle injury induced by cardiotoxin (CTX) injection to mimic a glucocorticoid excess state following severe muscle trauma. The findings reported here showed that DEX treatment together with CTX-induced muscle injury led to a significant amount of DC in muscle. This effect was likely related to protein level alterations in the fibrinolytic system and resultant decreased circulating transforming growth factor-beta 1 (TGF-β1), given that supplementation of recombinant TGF-β1 markedly rescued this phenomenon. In summary, our results suggest that glucocorticoid excess impairs muscle regeneration and promotes DC/HO, and that TGF-β1 could be a key factor in modulating this process. Dewatering and sulfide control are the key challenges in treating chemically enhanced primary treatment (CEPT) sludge. In this study, an electrochemical pretreatment (EPT) approach with the input of 10 V/800 mA was explored for simultaneously improving the dewaterability of CEPT sludge and eliminating its sulfide production. The effects of different electrode materials (carbon and titanium) and EPT durations (from 5 to 15 min) were documented to reveal the underlying EPT mechanism. EPT with titanium electrodes (titanium-EPT) led to limited improvement in dewaterability and sulfide control. EPT with carbon electrodes (carbon-EPT) for 15 min, however, led to decreases in capillary suction time and specific resistance in filtration of over 80% and the suppression of about 99% of hydrogen sulfide (H2S(g)) production over 5 days of anaerobic storage. Analysis of the characteristics of treated CEPT sludge revealed that carbon-EPT disintegrated sludge flocs with ∼70% reduction in sludge particle sizes and release of aromatic and tyrosine protein-like substances, thus enhancing sludge dewaterability. GSK-3 signaling pathway The sulfur balance in the liquid and gaseous phases showed that most of the sulfur-containing compounds remained in the solid phase as aliphatic sulfur and sulfonic acid after carbon-EPT, thereby mitigating sulfide emission. While the pattern of sulfur distribution in sludge with titanium-EPT was dominated by sulfide, it was similar to the control sample. Reduction in bacteria associated with sulfide production (i.e., Lachnospiraceae) in CEPT sludge after carbon-EPT also contributed to sulfide elimination. This study demonstrates that EPT can be a superior option for simultaneously enhancing the dewaterability of CEPT sludge and mitigating its sulfide production. Tropical conditions favour the auto-decomposition of monochloramine (MCA) leading to disinfectant decay and free ammonia in drinking water distribution systems (DWDS); thus, they promote the growth of nitrifiers and the development of biofilms on the inner-pipe surface. Biofilms can adversely impact the provision of safe and biologically stable water. Moreover, there is a general lack of understanding of the role of microbial communities in DWDS in regions with warm temperatures and no distinct seasons. Here, we report a survey on biofilms from full-scale monochloraminated DWDS in a highly urbanised metropolis using next generation sequencing tools. The monitoring campaign consisted of sampling biofilms and bulk waters from 21 in-service pipes. We characterized the microbial community with emphasis on nitrifying bacteria and archaea using 16S rRNA gene amplicon sequencing and potential nitrification activity. Samples grouped into two clusters, characterized by their low (Cluster LD) and high (Cluster HD) α-di when designing effective disinfection strategies. OBJECTIVES Human papillomavirus (HPV) infection drives carcinogenesis in the oropharynx. No standard sampling or HPV detection methods for evaluating oropharyngeal HPV infection exist. The prevalence of oral HPV infection in Japan is unknown. MATERIALS AND METHODS We examined 435 healthy Japanese individuals to address whether adding tonsillar washing to oral gargling would improve HPV detection. We compared HPV assessment using GENOSEARCH HPV31 versus nested PCR and direct sequencing. Associations between HPV infection and demographic and behavioral characteristics were examined. RESULTS Most participants who were HPV-positive based on oral gargles were also HPV-positive based on tonsillar washings 11 (64.7%) of 17 on nested PCR and 12 (70.6%) of 17 on GENOSEARCH HPV31. Although HPV infection was more prevalent in oral gargles followed by tonsillar washings than in oral gargles alone, the difference was not statistically significant (nested PCR, 4.8% vs. 3.9%, P = 0.46; GENOSEARCH HPV31, 5.3% vs. 3.9%, P = 0.33). The overall agreement between nested PCR and GENOSEARCH HPV31 was 98.6%, with 76.0% positive agreement. The overall prevalence of oral HPV infection in Japan was 5.7% (95% confidence interval, 3.9-8.3%). Men had a significantly higher prevalence of oral HPV infection than women (8.3% vs. 2.6%, P = 0.02). Infection increased with number of lifetime sexual partners (P  less then  0.001 for trend). CONCLUSION The oropharynx is probably the major source of HPV-infected cells in oral gargles. Oral gargling could be a standard sampling method for evaluating oropharyngeal HPV infection. GENOSEARCH HPV31 could be an option for oral HPV detection. The efficacy of chitosan (CH) and whey protein (WP) films impregnated with garlic essential oil (GEO, 2% v/v) or nanoencapsulated GEO (NGEO, 2% v/v) to extend the shelf life of refrigerated vacuum-packed sausages were assessed and compared during 50 days. The primary evaluation of GEO and NGEO showed that GEO had a considerable amount of active compounds diallyl sulfide derivatives (~67%) and the mean size and zeta potential of NGEO were 101 nm and -7.27 mV, respectively. Based on the microbiological and lipid stability analysis of the sausages, all active films retarded lipid oxidation and the growth of main spoilage bacterial groups compared to the control, and CH film containing NGEO exhibited the best result with the peroxide value, thiobarbituric acid reactive substances and aerobic plate count of 0.37 (meq/kg lipid), 0.47 (mg malondialdehyde/kg) and 3.69 (log CFU/g), respectively, on day 50.
