In psychiatry, mental dysfunction is typically framed in relation to models that seek to be continuous with physiology or evolutionary biology and that compare individual fitness to a broader functional norm. Proponents of the neurodiversity movement, however, challenge the pathologization of minority cognitive styles and argue that we should reframe neurocognitive diversity as a normal and healthy manifestation of biodiversity. Neurodiversity proponents have thus far drawn on social-relational models of disability to challenge the medical model of disability, but they have not developed an alternative functional analysis to replace conceptions of neurological dysfunction or impairment. Here I clarify and defend the neurodiversity perspective by drawing on ecological functional models that take relational contributions to collectives, and group functioning, into account alongside individual functionality. Using the example of autism as well as recent developments in the study of cognitive diversity, I apply these models to human mental functioning and argue that what I call the ecological model has greater utility for research and practice than the leading psychiatric functional analyses of mental functioning.Weight loss and exercise reduce colorectal cancer (CRC) risk in persons with obesity. To address this gap, male C57Bl/6J mice were fed a high-fat diet (HFD) to induce obesity or a control (CON) diet prior to azoxymethane injection to induce preneoplastic colorectal lesions. The HFD group was then randomized to weight loss conditions that included (1) switching to the CON diet only (HFD-SED) or switching to the CON diet with treadmill exercise training (HFD-EX). Average myofibre cross-sectional area was not different between groups. There were more smaller-sized fibers in HFD-EX (p less then 0.05 vs. CON), and more fibrosis in HFD-SED (p less then 0.05 vs. HFD-EX and CON). There was a trend for more committed (Pax7+MyoD+) myoblasts (p=0.059) and more fibro-adipogenic progenitors (FAPs) in HFD-EX (p less then 0.05 vs. CON). Additionally, the canonical pro-inflammatory marker p-NF-κB, was markedly reduced in the insterstitium of HFD-EX (p less then 0.05 vs. CON and HFD-SED). Our findings suggest that in mice with preneoplastic colorectal lesions, HFD followed by weight loss with exercise, reduces muscle fibrosis and results in a higher content of muscle stem/progenitor cells. MAVS (mitochondrial antiviral signaling protein) represents such an example that functions as a platform molecule to mediate mitochondrial innate immune signaling. Recently, multimeric aggregation of MAVS has been identified as a key molecular process for its signaling. The underlying mechanisms to regulate this, however, are still incompletely understood. We hypothesized that PINK1 (PTEN-induced kinase 1) plays an important role in the regulation of multimeric MAVS aggregation and its consequent pathobiology. To test whether PINK1 interacts with MAVS, bimolecular fluorescence complementation analysis and IP were performed. In vivo functional significance of PINK1 in the regulation of MAVS signaling was evaluated from both murine modeling of influenza viral infection and bleomycin-induced experimental pulmonary fibrosis, wherein MAVS plays important roles. Multimeric MAVS aggregation was induced by mitochondria dysfunction, and, during this event, the stabilized PINK1 interacted physically with MAVS and antagonized multimeric MAVS aggregation. Accordingly, the MAVS-mediated antiviral innate immune and NLRP3 inflammasome signaling were enhanced in PINK1 deficiency. In addition, in vivo studies revealed that MAVS-mediated pulmonary antiviral innate immune responses and fibrotic responses after bleomycin injury were enhanced in PINK1 deficiency. In conclusion, these results establish a new role of PINK1 in the regulation of MAVS signaling and the consequent pulmonary pathobiology.The influenza vaccine is not included in China's national immunization program. Here, we assessed influenza vaccine coverage among Shanghai residents and estimated the impacts of the coronavirus disease 2019 (COVID-19) epidemic and a hypothetical free vaccine strategy on the willingness of participants to receive influenza vaccines. The coverage of influenza vaccines among Shanghai residents from 2015 to 2019 was 1.4% (range 1%-2.4%). A total of 792 adult participants (aged 19-59 years) along with 821 children and 445 older individuals were included in the analysis. The willingness of participants to receive influenza vaccines reached 68.4% following the COVID-19 epidemic. Furthermore, if the vaccine was made available at no cost, 85.9% of participants were willing to be vaccinated. Our data indicated that influenza vaccine coverage is extremely low in Shanghai but that more than two-thirds of participants were willing to receive influenza vaccines following the COVID-19 epidemic. Making influenza vaccines available for free could further increase coverage.A previous isolated Gordonia sp. (Lff) was used to degrade di-n-octyl phthalate (DOP) contamination in both aqueous solution and soil. The influence of temperature, pH, inoculum size, salt content and initial concentration of DOP on DOP degradation by Lff were analysed. The response of soil bacterial community to DOP and Lff was also analysed by Illumina MiSeq sequence method. Results showed that the optimal temperature, pH, inoculum size and salt content were 35oC, 8.0, 5% and 0.917) with the half-life (t1/2) changing irregularly between 0.58 and 0.83 d. In addition, Lff enhanced the removal of DOP in soil and alleviated the toxicity of DOP on soil microorganisms. Furthermore, its influence on soil bacterial community is not obvious. These results suggested that Lff was effective in remediating DOP contamination in different environments.
