Aim Scientists have tried to culture and transplant the ovarian tissues (OT), to preserve fertility in cancer patients. Methods Fresh sheep OT were grafted onto the CAM for 5 days, and divided into four groups based on the addition of increasing doses of GDF9β (0, 150, 200 and 250 ng/mL). Following culture, histological (hematoxylin and eosin [H&E] staining) and immunohistological studies (Ki-67) were done. Fibrotic and necrotic regions were measured using MICROVISIBLE software. For comparing the follicle development rates between the groups as well as differences in the Ki-67-positive follicles, analysis of variance was applied. Results In both 200 and 250 ng/mL GDF9β groups, significantly higher rates of intermediary and primary follicles were observed, also the numbers of good quality follicles increased in the aforementioned groups and the rates of fibrotic and necrotic areas decreased. Moreover, in the 200 and 250 ng/mL GDF9β groups, the number of capillaries and the proliferative activity increased. The lower dose of GDF9β (150 ng/mL) neither activated the primordial follicles nor lead to an increase in the number of growing follicles. Conclusion Addition of high dosages of GDF9β to the OT, grafted onto the CAM resulted in higher folliculogenesis and better transplantation features due to improvement in angiogenesis.How to solve a problem like man-made plastics? This study aimed to propose a population pharmacokinetic (PK) model of dexmedetomidine in a heterogeneous group of intensive care unit patients, incorporating 29 covariates potentially linked with dexmedetomidine PK. Data were collected from 70 patients aged between 0.25 and 88 years and treated with dexmedetomidine infusion for various durations at 1 of 4 medical centers. Statistical analysis was performed using a nonlinear mixed-effect model. Categorical and continuous covariates including demographic data, hemodynamic parameters, biochemical markers, and 11 single-nucleotide polymorphisms were tested. A 2-compartment model was used to describe dexmedetomidine PK. An allometric/isometric scaling was used to account for body weight difference in PK parameters, and the Hill equation was used to describe the maturation of clearance. Typical values of the central and peripheral volume of distribution and the systemic and distribution clearance for a theoretical adult patient were central volume of distribution = 22.50 L, peripheral volume of distribution = 86.1 L, systemic clearance = 34.7 L/h, and distribution clearance = 40.8 L/h. The CYP1A2 genetic polymorphism and noradrenaline administration were identified as significant covariates for clearance. A population PK model of dexmedetomidine was successfully developed. The proposed model is well calibrated to the observed data. The identified covariates account for less then 5% of interindividual variability and consequently are of low clinical significance for the purpose of dose adjustment.Hereditary Hemochromatosis is a condition caused by defects in iron-sensing genes leading to parenchymal iron loading. Here we provide a systematic review of iron reduction in HH, including 24 studies and 6000 patients. While strong deductions are prohibited by an absence of robust clinical trial data, the purported benefits of venesection are reviewed and we report an improvement in fatigue, liver function tests and fibrosis, and overall survival. In conclusion, these findings, coupled with the absence of an alternative, low-cost, effective and tolerable therapy, suggests venesection will remain the mainstay of treatment in HH for decades to come.Organic materials with long-lived, color-tunable phosphorescence are potentially used for optical recording, anti-counterfeiting, and bioimaging. Herein, we develop a series of novel host-guest organic phosphors allowing dynamic color tuning from the cyan (502 nm) to orange red (608 nm). Guest materials are employed to tune the phosphorescent color, while the host materials show a synergistic effect to the guest to activate the phosphorescence emission. These organic phosphors have an ultra-long lifetime of 0.7 s and a maximum phosphorescence efficiency of 18.2%. Although color tunable inks have already been developed using visible dyes, solution-processed security inks that are temperature dependent and display time-resolved printed images are unprecedented. This strategy can provide a crucial step towards the next-generation of security technologies for information handling.
