Nowadays, healthcare informatics is mainly focussing on the classification of breast cancer images, due to the lethal nature of this cancer. There are chances of inter- and intra-observer variability that may lead to misdiagnosis in the detection of cancer. This study proposed an automatic breast cancer classification system that uses support vector machine (SVM) classifier based on integrated features (texture, geometrical, and color). The University of California Santa Barbara (UCSB) dataset and BreakHis dataset, which are available in public domain, were used. A classification comparison module which involves SVM, k-nearest neighbor (k-NN), random forest (RF), and artificial neural network (ANN) was also proposed to determine the classifier that best suits for the application of breast cancer detection from histopathology images. The performance of these classifiers was analyzed against metrics like accuracy, specificity, sensitivity, balanced accuracy, and F-score. Results showed that among the classifiers, the SVM classifier performed better with a test accuracy of approximately 90% on both the datasets. Additionally, the significance of the proposed integrated SVM model was statistically analyzed against other classifier models.

To evaluate the effect of capsular tension rings with multifunctional lens position and optical outcomes.

We defined multifunctional lens as more than improving vision, but also to restore visual quality. PubMed, EMBASE, Cochrane Library and Scopus were searched for English-language articles published up to November 11, 2020. Randomized controlled trials and comparative prospective clinical trials were selected. Data extraction was completed by independent pairs of reviewers. The risk of bias was evaluated using the Cochrane Collaboration's risk-of-bias tool for RCTs and select items from the Newcastle-Ottawa Scale for comparative prospective clinical trials.

A total of 5 randomized controlled trials and 6 prospective comparative clinical trials were included. One thousand nine hundred and ninety-nine eyes of implantation intraocular lens were evaluated. Capsular tension ring was helpful in un-corrected distance visual acuity (SMD 0.54, 95% CI = 0.15 to 0.94, p = 0.829) in 1st month. Contrary to 1st mon5% CI = - 1.19 to 0.51, p = 0.038) and tilt (SMD - 1.00, 95% CI = - 2.19 to 0.19, p = 0.007), but capsular tension ring is helpful to prevent lens tilt in 1st month (SMD - 0.67, 95%CI = - 1.08 to 0.27, p = 0.323). In 3rd month, there was no significant difference between two groups in lens rotation (SMD - 0.51, 95%CI = - 1.71 to 0.69, p < 0.001).

The correlation is low between capsular tension ring and postoperative optical outcomes and lens position, based on small numbers of studies in a short range of follow-up.

Plant-derived phytochemicals such as flavonoids have been explored to be powerful antioxidants that protect against oxidative stress-related diseases. In the present study, Morin, a flavonoid compound was studied for its antioxidant and antidiabetic properties in relation to oxidative stress in insulin resistant models conducted in rat skeletal muscle L6 cell line model.

Evaluation of antioxidant property of morin was assayed using in vitro methods such as cell viability by MTT assay, estimation of SOD and CAT activity and NO scavenging activity. The anti-oxidative nature of morin on L6 cell line was conducted by the DCF-DA fluorescent activity. Glucose uptake in morin treated L6 myotubes are accessed by 2-NBDG assay in the presence or absence of IRTK and PI3K inhibitors. Further glycogen content estimation due to the morin treatment in L6 myotubes was performed. Antioxidant and insulin signaling pathway gene expression was examined over RT-PCR analysis.

Morin has a negligible cytotoxic effect at doses ty to act as an anti-oxidant by lowering ROS levels and demonstrating insulin mimetic activity by reversing insulin resistance associated with oxidative stress.

Renin-angiotensin system (RAS) influences the central nervous system not only through its peripheral impact-the brain possesses its own local RAS. Studies showed altered RAS components in Parkinson's disease (PD) and their association with oxidative stress which may be linked to neurodegeneration and dementia. Moreover, the protective functions of RAS blockade antagonists against cognitive decline and dementia have been suggested. This study aimed to examine whether genetic variability in RAS genes correlates with cognitive decline in PD.

We genotyped single nucleotide polymorphisms (SNPs) in angiotensinogen (AGT rs699, rs4762), angiotensin II receptors (AGTR1 rs5186 and AGTR2 rs5194, rs1403543) genes, as well as insertion/deletion polymorphism in the angiotensin-converting enzyme (ACE I/D) gene in 256 PD patients, divided into three groups without cognitive decline, with mild cognitive impairment and with PD dementia. We did not find any significant differences in the frequencies of the analysed polymorphisms in any of the groups.

Despite no direct correlation between the investigated polymorphisms in RAS genes and cognitive decline in PD, we believe the impact of those genotypes may be indirect, affecting RAS blockade treatment.

Human endogenous retroviruses (HERVs), remnants of ancestral infections, represent 8% of the human genome. HERVs are co-opted for important physiological functions during embryogenesis; however, little is known about their expression in human gametes. We evaluated the transcriptional levels of several retroviral sequences in human spermatozoa.

We assessed, through a Real-Time PCR assay, the transcription levels of the pol genes of HERV-H, -K and -W families and of env genes of syncytin (Syn)1 and Syn2 in the spermatozoa from 8 normospermic subjects. The entity and distribution of their expressions were compared to values found in white blood cells (WBCs) from 16 healthy volunteers. The level of HERV transcripts was significantly lower in spermatozoa than in WBCs for HERV-H-pol, HERV-K-pol, HERV-W-pol, and Syn2.In contrast, the level of expression of Syn1 in the sperm was similar to that found in WBCs and it was significantly higher than the mRNA concentrations of other HERV genes in spermatozoa.

Our fin manipulation procedures, such as cryopreservation, may potentially influence HERV transcription in the human male gamete.

In adolescent idiopathic scoliosis (AIS), non-invasive surgical techniques such as anterior vertebral body tethering (AVBT) enable to treat patients with mild and severe degrees of deformity while maintaining lower lumbar motion by avoiding spinal fusion. However, multiple features and characteristics affect the overall patient outcome, notably the 3D spine geometry and bone maturity, but also from decisions taken intra-operatively such as the selected tethered vertebral levels, which makes it difficult to anticipate the patient response.

We propose here a forecasting method which can be used during AVBT surgery, exploiting the spatio-temporal features extracted from a dynamic networks. The model learns the corrective effect from the spine's different segments while taking under account the time differences in the initial diagnosis and between the serial acquisitions taken before and during surgery. Clinical parameters are integrated through an attention-based decoder, allowing to associate geometrical features to patient status. Long-term relationships allow to ensure regularity in geometrical curve prediction, using a manifold-based smoothness term to regularize geometrical outputs, capturing the temporal variations of spine correction.

A dataset of 695 3D spine reconstructions was used to train the network, which was evaluated on a hold-out dataset of 72 scoliosis patients using the baseline 3D reconstruction obtained prior to surgery, yielding an overall reconstruction error of [Formula see text]mm based on pre-identified landmarks on vertebral bodies. The model was also tested prospectively on a separate cohort of 15 AIS patients, demonstrating the integration within the OR theatre.

The proposed predictive network allows to intra-operatively anticipate the geometrical response of the spine to AVBT procedures using the dynamic features.

Several DDE solvers have been implemented in NONMEM 7.5 for the first time. Two of them are based on algorithms already applied elsewhere, while others are extensions of existing ordinary differential equations (ODEs) solvers. The purpose of this tutorial is to introduce basic concepts underlying DDE based models and to show how they can be developed using NONMEM. The examples include previously published DDE models such as logistic growth, tumor growth inhibition, indirect response with precursor pool, rheumatoid arthritis, and erythropoiesis-stimulating agents. We evaluated the accuracy of NONMEM DDE solvers, their ability to handle stiff problems, and their performance in parameter estimation using both first-order conditional estimation (FOCE) and the expectation-maximization (EM) method. GSK-4362676 chemical structure NONMEM control streams and excerpts from datasets are provided for all discussed examples. All DDE solvers provide accurate and precise solutions with the number of significant digits controlled by the error tolerance parameters. For estimation of population parameters, the EM method is more stable than FOCE regardless of the DDE solver.Intravitreal (IVT) injection remains the preferred administration route of pharmacological agents intended for the treatment of back of the eye diseases such as diabetic macular edema (DME) and neovascular age-related macular degeneration (nvAMD). The procedure enables drugs to be delivered locally at high concentrations whilst limiting whole body exposure and associated risk of systemic adverse events. Nevertheless, intravitreally-delivered drugs do enter the general circulation and achieving an accurate understanding of systemic exposure is pivotal for the evaluation and development of drugs administered in the eye. We report here the full pharmacokinetic properties of THR-687, a pan RGD integrin antagonist currently in clinical development for the treatment of DME, in both rabbit and minipig. Pharmacokinetic characterization included description of vitreal elimination, of systemic pharmacokinetics, and of systemic exposure following IVT administration. For the latter, we present a novel pharmacokinetic model that assumes clear partition between the vitreous humor compartment itself where the drug is administered and the central systemic compartment.
