Despite interest in cardiovascular health, awareness of benefits of statin therapy was low and knowledge of recommended statin therapy was low. Increasing patients' awareness of their eligibility through systematic testing and linkage to statin therapy, along with education, may increase statin use among patients recommended for therapy.The first example of solvent-free, organocatalyzed, polymerization of 1,3-dioxolan-4-ones, used as easily accessible monomers for the synthesis of polylactic acid (PLA), is described here. An optimization of reaction conditions was carried out, with p-toluensulfonic acid emerging as the most efficient Brønsted acid catalyst. The reactivity of the monomers in the tested conditions was studied following the monomer conversion by 1H NMR and the molecular weight growth by SEC analysis. The multiple-purpose connectivity map (CMAP) was used to discover potential molecules against HUA and renal fibrosis. We used HUA and unilateral ureteral occlusion (UUO) model mice to verify renoprotective effects of molecules and explore related mechanisms. In vitro experiments were performed in HepG2 and NRK-52E cells induced by UA. Esculetin was the top scoring compound and lowered serum uric acid (SUA) levels with dual functions on UA excretion. Esculetin exerted these effects by inhibiting expression and activity of xanthine oxidase (XO) in liver, and modulating UA transporters in kidney. The mechanism by which esculetin suppressed XO was related to inhibiting the nuclear translocation of hexokinase 2 (HK2). Esculetin was anti-fibrotic in HUA and UUO mice through inhibiting TGF-β1-activated profibrotic signals. The renoprotection effects of esculetin in HUA mice were associated with lower SUA, alleviation of oxidative stress, and inhibition of fibrosis. Esculetin is a candidate urate-lowering drug with renoprotective activity and the ability to inhibit XO, promote excretion of UA, protect oxidative stress injury, and reduce renal fibrosis.Serological diagnosis of Zika virus (ZIKV) infection is challenging because of antigenic cross-reactivity with dengue virus (DENV). This study evaluated the accuracy of the Zika IgM antibody capture enzyme-linked immunosorbent assay (CDC Zika IgM MAC-ELISA) in differentiating between ZIKV and DENV infections. To determine sensitivity, we used acute- and convalescent-phase sera from 21 patients with RT-PCR-confirmed ZIKV infection. To determine specificity, we used acute- and convalescent-phase sera from 60 RT-PCR-confirmed dengue cases and sera from 23 blood donors. During the acute-phase of the illness, the assay presented a sensitivity of 12.5% (2/16) for samples collected 0-4 days post symptoms onset (DPSO), and of 75.0% (3/4) for samples collected 5-9 DPSO. During the convalescent-phase of the illness, the test sensitivity was 90.9% (10/11), 100% (2/2), and 0% (0/2) for samples obtained 12-102, 258-260, and 722-727 DPSO, respectively. Specificity for acute- and convalescent-phase samples from RT-PCR-confirmed dengue cases was 100% and 93.2%, respectively. Specificity for blood donor samples was 100%. This study aimed to investigate the effect of 4-week RRB supplementation on (poly)phenolic metabolism in adults with prediabetes and insulin-resistance (PreDM-IR); and whether adding fructo-oligosaccharides (FOS), prebiotics, would augment the microbial metabolites of RRB (poly)phenols. In a randomized crossover clinical trial, subjects (n = 35 PreDM-IR, n = 25; healthy Reference group, n = 10) consumed 1 cup RRB (fresh weight equivalence) per day and RRB with 8 g FOS per day each for 4 weeks in random order separated by 4-week washout. Plasma and urinary (poly)phenolic metabolites were characterized after (0-24h) consuming a RRB-based test drink (2 cups RRB) at baseline/week 0 and again after 4-week supplementations. A total of 123 (poly)phenolic metabolites were quantified. After 4-week RRB supplementation, several metabolite groups were significantly increased (p less then 0.05), including urolithins, phenyl-γ-valerolactones, and phenolic acids. Supplementing FOS with RRB for 4 weeks enhanced benzoic acid derivatives compared to the baseline (p less then 0.05). Specific effects of supplementation by metabolic status indicated 4-week RRB supplementation significantly increased microbial metabolites that were lower in PreDM-IR group. Our results suggest alterations in the capacity of PreDM-IR group to metabolize and render bioavailable raspberry-derived (poly)phenols when consumed regularly.Metal Additive Manufacturing and Laser Powder Bed Fusion (LPBF), in particular, have come forth in recent years as an outstanding innovative manufacturing approach. The LPBF process is notably characterized by very high solidification and cooling rates, as well as repeated abrupt heating and cooling cycles, which generate the build-up of anisotropic microstructure and residual stresses. Post-processing stress-relieving heat treatments at elevated temperatures are often required in order to release some of these stresses. The effects of 1 h-hold heat treatments at different specific temperatures (solutionizing, annealing, stress-relieve and low-temperature stress-relieve) on residual stress levels together with microstructure characterization were therefore investigated for the popular Alloy 625 produced by LPBF. The build-up of residual stress is accommodated by the formation of dislocations that produce local crystallographic misorientation within grains. Electron backscattered diffraction (EBSD) was used to investigate local misorientation by means of orientation imaging, thereby assessing misorientation or strain levels, in turn representing residual stress levels within the material. The heavily constrained as-built material was found to experience full recrystallization of equiaxed grains after solutionizing at 1150 °C, accompanied by significant drop of residual stress levels due to this grains reconfiguration. Heat treatments at lower temperatures however, even as high as the annealing temperature of 980 °C, were found to be insufficient to promote recrystallization though effective to some extent to release residual stress through apparently dislocations recovery. Average misorientation data obtained by EBSD were found valuable to evaluate qualitatively residual stress levels. The effects of the different heat treatments are discussed and suggest that the peculiar microstructure of alloys produced by LPBF can possibly be transformed to suit specific applications.Acute Myeloid Leukemia (AML) is characterized by recurrent genetic and cytogenetic lesions that are utilized for risk stratification and for making treatment decisions. In recent years, methylation dysregulation has been extensively studied and associated with risk groups and prognosis in adult AML, however, such studies in pediatric AML are limited. Moreover, the mutations in epigenetic genes such as DNMT3A, IDH1 or IDH2 are almost absent or rare in pediatric patients as compared to their abundance in adult AML. In the current study, we evaluated methylation patterns that occur with or independent of the well-defined cytogenetic features in pediatric AML patients enrolled on multi-site AML02 clinical trial (NCT00136084). Our results demonstrate that unlike adult AML, cytosine DNA methylation does not result in significant unique clusters in pediatric AML, however, DNA methylation signatures correlated significantly with the most common and recurrent cytogenetic features. Paired evaluation of DNA methylation and expression identified genes and pathways of biological relevance that hold promise for novel therapeutic strategies. Our results further demonstrate that epigenetic signatures occur complimentary to the well-established chromosomal/mutational landscape, implying that dysregulation of oncogenes or tumor suppressors might be leveraging both genetic and epigenetic mechanisms to impact biological pathways critical for leukemogenesis.The optimization of ecological water supplement scheme in Momoge National Nature Reserve (MNNR), using an interval-parameter two-stage stochastic programming model (IPTSP), still experiences problems with fuzzy uncertainties and the wide scope of the obtained optimization schemes. These two limitations pose a high risk of system failure causing high decision risk for decision-makers and render it difficult to further undertake optimization schemes respectively. Therefore, an interval-parameter fuzzy two-stage stochastic programming (IPFTSP) model derived from an IPTSP model was constructed to address the random variable, the interval uncertainties and the fuzzy uncertainties in the water management system in the present study, to reduce decision risk and narrow down the scope of the optimization schemes. The constructed IPFTSP model was subsequently applied to the optimization of the ecological water supplement scheme of MNNR under different scenarios, to maximize the recovered habitat area and the carrying cscenarios for decision-makers to select and providing decision space to adjust the decision schemes, also shortened the decision range, thereby reducing the decision risk and the difficulty of undertaking decision schemes. In addition, the fuzzy membership obtained from the IPFTSP model, reflecting the relationship among the possibility level, the target value, and the decision risk, assists the decision-makers in planning the ecological water supplement scheme with a preference for target value and decision risk.
