Furthermore, the association statistics of the machine learning algorithm were also consistent, which suggested that depletion of Ca below 8.245 mg/dl was shown to elevate PTH levels greater than 167 pg/ml when P > 4.66. Subnormal depletion in 25-OHD (9.3-16.2 ng/ml) is associated with subnormal elevation in PTH (47-73.6 pg/ml). To conclude, ANFIS and machine learning algorithm are in agreement with each other in stating that 25-OHD deficiency triggers lower calcium levels, lower calcium and higher phosphorus trigger PTH production. © Association of Clinical Biochemists of India 2018.A genetic variant may alter a gene expression level and as a result be associated with pathological characteristics in breast cancer. In this research, the frequency and association of the ErbB4 3'-untranslated region (3'-UTR) variant, rs12471583 (c.*3622A>G) was studied in an Iranian breast cancer patients. In silico assessment was performed to predict the function of the rs12471583 variant located on the 3'-UTR of ErbB4. Furthermore, as a case-control study, this polymorphism was genotyped in 243 breast cancer patients and non-cancerous controls using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique. The Armitage's trend test and regular association tests were performed to analyze a possible association between the rs12471583 and risk of breast cancer and its relevant pathological traits. The bioinformatics analysis predicted that the rs12471583 SNP is located on the four miRNA binding sites, including miR-511-5p, miR-4659a-5p, miR-4659b-5p, and miR-6830-3p. Novobiocin research buy According to logistic regression tests, the G allele is negatively associated with ER- (OR = 0.20, 95% C.I. = 0.04-0.93, p = 0.026), PR- (OR = 0.31, 95% C.I. = 0.10-0.98, p = 0.039), ER-/PR- (OR = 0.20, 95% C.I. = 0.04-0.93, p = 0.026), and advanced breast cancer (OR = 0.40, 95% C.I. = 0.18-0.85, p = 0.016). It has been found that ErbB4 expression may be linked to unfavorable outcomes in breast cancer. Likewise, our results suggest that the G allele may strengthen miRNA-ErbB4 binding efficiency and as a result reduce expression of ErbB4. This is a possible explanation for the observed association. © Association of Clinical Biochemists of India 2018.Empagliflozin, a SGLT-2 inhibitor, improves diabetic nephropathy through its pleiotropic anti-inflammatory effects. The present study aims to evaluate empagliflozin effects on renal and urinary levels of tubular epithelial cell injury markers in streptozotocin-induced diabetic rats. Empagliflozin at 10 mg/kg (p.o.) was administered for 4 weeks, beginning 8 weeks after induction of diabetes. Renal function as well as markers of renal tubular epithelial cell injury were assessed in kidney tissue homogenates and urine. Empagliflozin was able to ameliorate diabetes induced elevations in serum cystatin C levels. It also alleviated renal KIM-1/NGAL levels and urinary albumin, α-GST, and RBP excretions. In addition to decreasing urinary levels of cell cycle arrest indices i.e. TIMP-2 and IGFBP7, empagliflozin mitigated acetylated NF-κB levels in renal tissues of diabetic rats. As a whole, these findings reveal empagliflozin capability in improving diabetic nephropathy via ameliorating indices of renal inflammation, injury, and cell cycle arrest on streptozotocin-induced diabetic rats. © Association of Clinical Biochemists of India 2018.Altered blood viscosity (BV) may affect blood pressure (BP) and develops further complications in diabetes. A case-control study was performed to examine the relationship of erythrocyte sedimentation rate (ESR), hematocrit, fibrinogen, and BV with glycemic markers and BP in middle-aged normotensive and hypertensive type 2 diabetic patients and healthy controls. A total of 145 participants between age group 30-50 years divided into three groups; controls (n = 60), type 2 diabetes mellitus (T2DM, n = 55), and T2DM with hypertension (T2DM + HTN, n = 30). ESR and hematocrit were determined by Wintrobe's method. Plasma fibrinogen was measured using Lempert method and BV calculated using Merill's formula. T2DM and T2DM + HTN patients had higher fasting plasma glucose (FPG), glycated hemoglobin (HbA1c), systolic BP (SBP), diastolic BP (DBP), mean arterial pressure (MAP), ESR, and fibrinogencompared to controls. In both male and female SBP, DBP, MAP, FPG, and HbA1c were significantly higher in T2DM and T2DM + HTN groups, compared to controls. Further, linear regression analysis revealed a positive association of ESR and fibrinogen with SBP, DBP, MAP, FPG, HbA1c, and positive diabetic status in all participants. Also, in the same analysis, BV showed a positive association with SBP, DBP, and MAP. The association of ESR and fibrinogenwith glycemic markers and BP in diabetes supporting the value of emerging marker's for early prediction of T2DM and hypertension. © Association of Clinical Biochemists of India 2018.Vitamin D deficiency in known to be high in obese and overweight adolescents. Few studies in other countries have shown lower parathyroid hormone (PTH) levels in obese adolescents despite high vitamin D deficiency. The aim of the study is to assess vitamin D and PTH levels and their relationship in overweight and obese adolescents in Telangana. This cross-sectional study was carried out in 108 overweight and obese boys and girls aged 12-18 years. A pretested Questionnaire was administered to collect information on demographics, sun exposure, medical and nutritional history. Anthropometry and blood sample was collected for estimating 25 hydroxy vitamin D, and PTH. Body mass index Z scores (WHO growth standards) were used for assessing overweight and obesity in adolescents. 54.6% of the subjects were boys. Mean (SD) levels of vitamin D and PTH were 18.25 (9.26) ng/mL and 45.39 (28.23) ng/L respectively. The prevalence of vitamin D deficiency and insufficiency in the current study was 54.6% and 25.9% respectively. However, PTH levels were high only in 24.2% of subjects (> 65 ng/L). There was an inverse but non-significant correlation between vitamin D and PTH. The association was same after adjusting for age, gender and weight. Our study highlights the high prevalence of vitamin D deficiency among overweight and obese adolescents in Telangana. Despite high vitamin D deficiency, very few had high PTH levels and the vitamin D-PTH axis may be altered in overweight and obese adolescents. © Association of Clinical Biochemists of India 2018.
