In this study, the capacity of methylene blue (MB) uptake from aqueous solution was investigated using acrylic amide-co-3-Allyloxy-2-hydroxy-1-propanesulfonic acid sodium salt/Gum Arabic semi-IPN hydrogel. The semi-IPN hydrogels were prepared by the classical free radical solution technique. The swelling experiments were carried out gravimetrically and the swelling kinetics parameters were calculated. Fourier transform infrared spectroscopy (FTIR) and scanning electron microscope (SEM) confirmed the semi-IPN hydrogel formation. The MB uptake was studied by the batch technique and the impact of different conditions was investigated. It was found that the obtained semi-IPN hydrogels gave adsorption capacity for the dye within a range of 101-187 mg g-1 at the initial dye concentration of 100 mg/L. The adsorption kinetics (pseudo-first-order and pseudo-second-order) and the adsorption isotherms (Langmuir, Freundlich, and Temkin equations) were tested and the adsorption data was well described by the pseudo-second-order and Freundlich models, respectively. The maximum adsorption capacity calculated by the Langmuir model (R2 = 0.8718) was 655.2 mg g-1. The thermodynamic study indicated the spontaneity and the endothermic nature of MB adsorption. Furthermore, the usability study showed that the prepared adsorbents could be employed repeatedly for successive 5 cycles of adsorption and desorption.Reliable separation of peptides, amino acids and proteins as accurate as possible with the maximum conformation and biological activity is crucial and essential for drug discovery. In our previous study, an immuno-stimulatory polysaccharide (RGP-AP-I) was isolated from Korean red ginseng (Panax ginseng C.A. Meyer). The present study aims to elucidate the structural characteristics of RGP-AP-I. Sequential enzyme hydrolysis was performed using four specific glycosylases, and chemical cleavage via β-elimination was carried out to determine the fine structure of RGP-AP-I. The degraded fragments were chemically identified using various chromatographic and spectrometric analyses, including HPLC-UVD, GC-MS, and tandem mass spectrometry. The results indicated that RGP-AP-I comprises a rhamnogalacturonan I (RG-I) backbone with repeating disaccharide units [→2)-Rhap-(1 → 4)-GalAp-(1→] and three side chains substituted at the C(O)4 position of the rhamnose residue in the backbone. The three side chains were identified as a highly branched α-(1 → 5)-arabinan, a branched β-(1 → 4)-galactan, and an arabino-β-3,6-galactan. Our results represent the first findings regarding the fine structure of the immuno-stimulatory polysaccharide RG-AP-I isolated from red ginseng.Chemokines are crucial regulators of cell mobilization for development, homeostasis, and immunity. Chemokines signal through binding to chemokine receptors, a superfamily of seven-transmembrane domain G-coupled receptors. In the present study, seventeen CXC chemokine ligands (SsCXCLs) and nine CXC chemokine receptors (SsCXCRs) were systematically identified from Sebastes schlegelii genome. Phylogeny, synteny, and evolutionary analyses were performed to annotate these genes, indicating that the tandem duplications (CXCL8, CXCL11, CXCL32, CXCR2, and CXCR3), the whole genome duplications (CXCL8, CXCL12, CXCL18, and CXCR4), and the teleost-specific members (CXCL18, CXCL19, and CXCL32) led to the expansion of SsCXCLs and SsCXCRs. In addition, SsCXCLs and SsCXCRs were ubiquitously expressed in nine examined healthy tissues, with high expression levels observed in head kidney, liver, gill and spleen. Moreover, most SsCXCLs and SsCXCRs were significantly differentially expressed in head kidney, liver, and gill after Aeromonas salmonicida infection, and exhibited tissue-specific and time-dependent manner. Finally, protein-protein interaction network (PPI) analysis indicated that SsCXCLs and SsCXCRs interacted with a few immune-related genes such as interleukins, cathepsins, CD genes, and TLRs, etc. These results should be valuable for comparative immunological studies and provide insights for further functional characterization of chemokines and receptors in teleost.
