Ribosomopathies constitute a class of inherited disorders characterized by defects in ribosome biogenesis and function. Classically, bone marrow (BM) failure is a clinical symptom shared between these syndromes, including Shwachman-Bodian-Diamond syndrome (SBDS). Eukaryotic translation initiation factor 6 (eIF6) is a critical translation factor that rescues the quasilethal effect of the loss of the SBDS protein.

To determine whether eIF6 activity is necessary for BM development.

We used eIF6(+/-) mice and primary BM megakaryocytes to investigate the involvement of eIF6 in the regulation of hematopoiesis.

We provide evidence that reduced eIF6 expression negatively impacts on megakaryopoiesis. We show that inhibition of eIF6 leads to a reduction in cell size and mean ploidy level of megakaryocytes and a delay in megakaryocyte maturation by blocking the G1 /S transition. Consistent with this phenotype, only few megakaryocyte-forming proplatelets were found in eIF6(+/-) cells. We also discovered that, in eIF6(+/-) cells, the steady-state abundance of mitochondrial respiratory chain complex I-encoding mRNAs is decreased, resulting in decreased reactive oxygen species (ROS) production. Intriguingly, connectivity map analysis showed that eIF6-mediated changes overlap with specific translational inhibitors. eIF6 is a translation factor acting downstream of insulin/phorbol 12-myristate 13-acetate (PMA) stimulation. PMA treatment significantly restored eIF6(+/-) megakaryocyte maturation, indicating that activation of eIF6 is essential for the rescue of the phenotype.

Taken together, our results show a role for eIF6-driven translation in megakaryocyte development, and unveil the novel connection between translational control and ROS production in this cell subset.

Limited research exists regarding the effectiveness of educational and psychological interventions for improving commonly presenting cognitive impairments experienced by children with epilepsy. We evaluated the efficacy of a commercially available, computerized, working memory (WM) program (Cogmed) using a well-defined population of children with epilepsy.

In this controlled trial, 77 children with symptomatic epilepsy (ages 6.5-15.5 years; 100% taking medication) with estimated intellectual ability greater than the 2nd percentile were randomly assigned to an intervention (n = 42) or waitlist-control (n = 35) group. Standardized assessments of attention and WM were administered pre- and posttraining or waitlist interval, 7 weeks apart.

Without intervention, participants displayed significant weaknesses in intelligence, attention, and WM compared to normative samples. After controlling for preintervention scores and intelligence, we found that significant treatment effects for the intervention group wererovement on some untrained tasks immediately postintervention, demonstrating preliminary usefulness of Cogmed as a treatment option.This study explored differences in intimate partner homicides (IPHs) among Asian Americans. Data from newspapers and femicide reports by different state coalitions on 125 intimate partner killings occurring between 2000 and 2005 were analyzed. Men were the perpetrators in nearly 9 out of 10 cases of Asian IPHs. Gender differences were found in ages of victims and perpetrators, types of relationship between partners, and methods of killing. Most homicides occurred among South-east Asians, and East Asians had the highest within-group proportion of suicides. The findings call for culturally competent risk assessment and intervention strategies to prevent IPHs among at-risk Asian Americans.

The aim of this study was to evaluate the associations between pain and physical functional limitation and health status in patients with knee osteoarthritis (OA).

A correlational study.

In a convenience sample of 73 patients with knee OA, pain and physical function were assessed using the Western Ontario and McMaster University Osteoarthritis Index. Health status was evaluated using multiple instruments under the International Classification of Functioning, Disability, and Health framework.

In the study of patients with knee OA, pain and physical functional limitation exhibited mild to strong associations with health status, including body function and structure (r = .26-.71), activities and participation (r = .24-.88), and personal and environmental factors (r = .24-.62).

In patients with knee OA, health status is mildly to strongly associated with pain and physical functional limitation.

Reducing pain and physical functional limitation in patients with knee OA might improve their health status.BACKGROUND To compare risk of carpal tunnel syndrome (CTS) in distal radius fracture (DRF) patients after 7 treatments using bridging external fixation (BrEF), non-bridging external fixation (non-BrEF), plaster fixation, K-wire fixation, dorsal plating fixation, volar plating fixation, and dorsal and volar plating by performing a network meta-analysis. MATERIAL AND METHODS An exhaustive search of electronic databases identified randomized controlled trails (RCTs) closely related to our study topic. The published articles were screened, based on predefined inclusion and exclusion criteria, to select high-quality studies for the present network meta-analysis. Data extracted from the selected studies were analyzed using STATA version 12.0 software. RESULTS The literature search and selection process identified 12 eligible RCTs that contained a total of 1370 DRF patients (394 patients with BrEF, 377 patients with non-BrEF, 89 patients with K-wire fixation, 192 patients with plaster fixation, 42 patients with dorsal plating fixation, 152 patients with volar plating fixation, and 124 patients with dorsal and volar plating fixation). Our network meta-analysis results demonstrated no significant differences in CTS risk among the 7 treatments (P>0.05). The value of surface under the cumulative ranking curve (SUCRA), however, suggested that dorsal plating fixation is the optimal treatment, with the lowest risk of CTS in DRF patients (dorsal plating fixation 89.2%; dorsal and volar plating 57.8%; plaster fixation 50.9%; non-BrEF 50.6%; volar plating fixation 39.6%; BrEF 38.4%; K-wire fixation 23.6%). CONCLUSIONS Our network meta-analysis provides evidence that dorsal plating fixation significantly decreases the risk of CTS and could be the method of choice in DRF patients.Three mononuclear complexes [Co(biq)X2] (biq = 2,2'-biquinoline; X = Cl, Br, I) were prepared by a solvothermal method and characterized by single-crystal X-ray diffraction. In all three complexes the Co(ii) atom is tetrahedrally coordinated by one biq ligand bonded in a chelate manner and two halogenido ligands. Hydrogen bonding interactions (C-HX) along with the π-π interactions contribute to the stability of the formed packing. Magnetic measurements as well as ab initio calculations revealed that the complexes possess a sizable easy-plane magnetic anisotropy (D > 0). They display a superparamagnetic behaviour in an applied external field that culminates between BDC = 0.2-0.3 T. Two relaxation processes are observed; the faster can be analysed in terms of the direct and Orbach processes yielding U/kB = 42.6 K and τ0 = 1.9 × 10(-10) s for X = Cl, U/kB = 39.6 K and τ0 = 1.2 × 10(-10) s for X = Br and U/kB = 57.0 K and τ0 = 3.2 × 10(-13) s for X = I at BDC = 0.2 T.This study investigates the capabilities of low tube voltage computed tomography (CT) and dual-energy CT (DECT) for predicting successful shock wave lithotripsy (SWL) of urinary stones in vitro. A total of 33 urinary calculi (six different chemical compositions; mean size 6 ± 3 mm) were scanned using a dual-source CT machine with single- (120 kVp) and dual-energy settings (80/150, 100/150 Sn kVp) resulting in six different datasets. The attenuation (Hounsfield Units) of calculi was measured on single-energy CT images and the dual-energy indices (DEIs) were calculated from DECT acquisitions. Calculi underwent SWL and the number of shock waves for successful disintegration was recorded. The prediction of required shock waves regarding stone attenuation/DEI was calculated using regression analysis (adjusted for stone size and composition) and the correlation between CT attenuation/DEI and the number of shock waves was assessed for all datasets. The median number of shock waves for successful stone disintegration was 72 (interquartile range 30-361). CT attenuation/DEI of stones was a significant, independent predictor (P less then 0.01) for the number of required shock waves with the best prediction at 80 kVp (β estimate 0.576) (P less then 0.05). Correlation coefficients between attenuation/DEI and the number of required shock waves ranged between ρ = 0.31 and 0.68 showing the best correlation at 80 kVp (P less then 0.001). The lamellar to inverted hexagonal phase transition (Lα-HII) of phospholipids is energetically analogous to the membrane fusion process, therefore understanding the Lα-HII transition with nanoparticulate additives is relevant to how membrane fusion may be affected by these additives, in this case the silica NPs. The overriding observation is that the HII/Lα boundaries in the DOPE p-T phase diagram were shifted by the presence of NPs the hydrophobic NPs enlarged the HII phase region and thus encouraged the inverted hexagonal (HII) phase to occur at lower temperatures, whilst hydrophilic NPs appeared to stabilise the Lα phase region. This effect was also NP-concentration dependent, with a more pronounced effect for higher concentration of the hydrophobic NPs, but the trend was less clear cut for the hydrophilic NPs. There was no evidence that the NPs were intercalated into the mesophases, and as such it was likely that they might have undergone microphase separation and resided at the mesophase domain boundaries. Whilst the loci and exact roles of the NPs invite further investigation, we tentatively discuss these results in terms of both the surface chemistry of the NPs and the effect of their curvature on the elastic bending energy considerations during the mesophase transition.

The global emergence of pathogens of urinary-tract infections resistant to ciprofloxacin or producing extended-spectrum β-lactamases (ESBL) led us to investigate the activity of older antimicrobials such as cefprozil and cefixime against a recent broad collection of urine enterobacteria from 2012 and 2013.

Minimum inhibitory concentrations and minimum bactericidal concentrations of cefprozil, cefixime and ciprofloxacin were determined against 293 Escherichia coli (40 ESBL producers), 54 Klebsiella pneumoniae (10 ESBL producers) and 53 Proteus mirabilis isolates.

Cefprozil was more active than ciprofloxacin against non-ESBL-producing E. coli (93.7% vs 80.2%, p<0.0001); this was not the case for cefixime (85.7% vs 80.2%, p 0.125). Overall, cefprozil and cefixime inhibited 80-90% of ciprofloxacin-resistant isolates of all studied species. However, they were active against less than 20% of ESBL-producing isolates.

Results suggest that cefprozil and cefixime remain a good therapeutic alternative against urine enterobacteria particularly in case of ciprofloxacin-resistant pathogens.
