The selected in silico models have provided lower EC50 values and are therefore more conservative. Models from the OECD QSAR ToolBox predicted values for 7 out of 9 and for 4 out of 9 chemicals for Daphnia and P. subcapitata, respectively. No predictive models were identified in such platform for STP microorganism's acute effects. In terms of models's specificity, biocide-specific models, developed from curated datasets integrated by biocidal active substances and implemented in VEGA, perform better in the case of microalgae but for Daphnia an alternative, non biocide-specific has revealed a better performance. For STP microorganisms only biocide-specific models have been identified.Using an ideal parental line to breed hybrid rice with low cadmium (Cd) accumulation in grain is an environmental-friendly approach to reduce the risk of Cd contamination in field. A grain low-Cd rice line YaHui2816 has stably low Cd in grain but strong Cd accumulation in straw, revealing specific pattern of its straw-grain Cd remobilization is beneficial to effectively breed hybrid rice for safe production as well as phytoremediation. In this study, a pot experiment was conducted to investigate Cd partitioning of YaHui2816 at different stages by comparison with a common rice C268A. The Cd from lower nodes and leaves was restricted in nodeⅡ, this Cd retention led to weak Cd transport from straw to ear in YaHui2816. Cd concentration in nodeⅡ of YaHui2816 was 1.56-fold and 7.36-fold higher than C268A at filling and mature stages. Thus, glutathione (GSH) and phytochelatin (PC) concentration, PC synthase (OsPCS1), GSH S-Transferase (OsGST) and Cd-remobilization associated genes were measured at filling stage. OsHMA2 and OsCCX2 were up-regulated in nodeⅡ of YaHui2816, relative expression of them were significantly higher than C268A. PCs participated in Cd remobilization process, remarkable PC increase in nodeⅡ of YaHui2816 was found in response to Cd treatment under regulation of OsPCS1 and OsGST of which PC2 was dominant form. Taken as a whole, the Cd retention in nodeⅡ of YaHui2816 acts as a 'firewall' to restrain Cd transport to grain. #link# This work provides more insight to understand possible function of alleles for reducing Cd concentration in grain as well as strengthening Cd accumulation in straw of YaHui2816.Chlorpyrifos (CPF) is one of the most frequently used pesticide in extensive agriculture around the world and can be incorporated by humans and animals with possible consequences on health. The effects of this pesticide on carcinogenesis are not clear and there is no consensus concerning the risks of this compound. In previous work, we demonstrated that CPF induces proliferation of breast cancer cells both in vivo and in vitro. In this work we investigate whether CPF promotes the epithelial-mesenchymal transition (EMT) in breast cancer cells. link2 Herein, we demonstrate that 50 μM CFP induces invasion in MCF-7 and MDA-MB-231 cells. In addition, 0.05 and 50 μM CPF increases migration in both cell lines. In MCF-7 cells, 0.05 and 50 μM CPF increase the metalloprotease MMP2 expression and decrease E-Cadherin and β-Catenin expression diminishing their membrane location. Furthermore, Selleck Sacituzumab govitecan μM CPF induces Vimentin expression and Slug nuclear translocation in MCF-7 cells. 0.05 and 50 μM CPF increase MMP2 gelatinolytic activity and expression, decrease β-Catenin expression and increase Vimentin expression in MDA-MB-231 cells. Inhibition of the oncoprotein c-Src reverses all the effects induced by CPF in MDA-MB-231 but not in MCF-7 indicating that c-Src is a kinase with a crucial role in the cells which grow in an estrogen-independent way. In MCF-7 cells both c-Src and estrogen receptor alpha must be blocked to completly inhibit the CPF-mediated effects. Our results show for the first time that the exposure to subthreshold concentrations of CPF promotes the modulation of EMT-molecular markers and pathways. These results, together with the ubiquitous distribution of the pesticide CPF, make it of utmost importance to take measures to minimize the risk of exposure to this compound.Reproductive functions may be affected by internal and external factors that are integrated in the central nervous system (CNS). Stressful stimuli induce the neuroendocrine response of the hypothalamic-pituitary-adrenal axis, as well as the synthesis of the neurosteroid allopregnanolone (AL) in the brain. This study tested the hypothesis that centrally administered AL could affect the expression of certain genes involved in reproductive functions at the hypothalamus and pituitary levels, as well as pulsatile gonadotropin secretion in sheep under both natural and stressful conditions. Luteal-phase sheep (n = 24) were subjected to a three-day (day 12-14 of the estrous cycle) series of control or AL (4 × 15 μg/60 μL/30 min, at 30 min intervals) infusions into the third ventricle. Acute stressful stimuli (isolation from other sheep and partial movement restriction) were used in the third day of infusion. Stressful stimuli reduced kisspeptin-1 mRNA levels in both the mediobasal hypothalamus (MBH) and the preoptic ectional effects on hypothalamic centers that regulate reproductive functions and secretory activity of AP gonadotrophs in sheep. However, we indicated the dominant inhibitory effect of AL under natural and stressful conditions.Myotis nigricans is an important species of insectivorous bat from the Vespertilionidae family, which is endemic of the Neotropics. Despite its wide distribution, no detailed study of the reproduction is found for this species. Thus, the aim of the present study was to evaluate and characterize morphological and morphometric variations of the female reproductive organs of M. nigricans during its different reproductive phases. Twenty adult females were collected, divided into four (4) sample groups, according to the reproductive status (nonreproductive, initial and advanced pregnancy and lactating), and submitted to anatomical, histological and morphometric analyses. The results demonstrated that the female reproductive organs of M. nigricans were composed of the ovaries, the uterus, the uterine tubes and the vagina, in addition to the mammary glands. The ovaries present a great number of interstitial glands and a small but persistent corpus luteum. Ovulation is simple, unilateral and non-preferential, possibly alternated in successive ovulations and may be sequential with the occurrence of postpartum estrus. The uterus is bicornuate, implantation is fundic and the placenta is chorioallantoic, discoidal and hemochorial. The vaginal epithelium is weakly keratinized and shows great disorganization during the periods of pregnancy and lactation. This study aimed to evaluate the use of Doppler ultrasound for determining alterations in uterine thickness and uterine arteries, use of chromohysteroscopy in evaluating endometritis with biopsies, and the impact of endometritis on oxidative and hormonal status. Twenty Arabian mares were divided into control and endometritis groups. Each underwent a physical examination, Doppler ultrasound, chromoendoscopy, and uterine biopsy. Blood samples from each animal were collected for hematologic, hormonal, and oxidative analysis. Results revealed a significant increase in estradiol, malondialdehyde (MDA), and nitric oxide metabolites (NOMs) associated with a significant decrease in progesterone and total antioxidant capacity (TAC) in the endometritis group. There was a significant difference in uterine thickness in the endometritis group. Uterine blood flow analyzed by color mode showed a significant increase in the endometritis group, while the spectral mode showed a significant increase in Doppler velocities and blood flow rate accompanied by a significant decrease in Doppler indices. The endometritis group showed ridged, bluish color in the endometrial wall, indicating hyperemia. Biopsy showed mononuclear inflammatory cells and edema in the endometrial interstitial tissue with sloughing of the epithelial lining. Conclusion Endometritis in mares is associated with oxidative processes and presents with an increase in MDA and NOMs and a decrease in TAC. Doppler ultrasound improved endometritis diagnosis because it showed alterations in uterine vascularization. Chromoendoscopy improved the accuracy in taking biopsy samples because it allows the practitioner to select appropriate areas for biopsy to improve histopathological diagnostic outcomes. Therefore, this technique is worthy of further study in the field of veterinary equine reproduction.A man developed vision loss and bilateral subhyaloid, outer plexiform, and subretinal hemorrhages after 2 minutes of chest compressions. Several potential mechanisms, including venous transmission of pressure and elevated intracranial pressure (ICP), could explain these findings.

To investigate whether outer retina hyperreflectivity (ORHR) and outer retina neovascularization (ORNV) are visible in eyes with macular telangiectasia type 2 (MacTel 2) before pigment proliferation is visible.

Retrospective study in a cohort of patients from the MacTel Project.

Thirty-nine MacTel 2 eyes without pigment on color fundus photography (CFP) at presentation were studied over a mean of 36.6 months (range, 13.8-50.4 months; standard deviation, ±10 months).

All patients routinely underwent Snellen best-corrected visual acuity (VA) measurement, CFP, spectral-domain OCT, and OCT angiography (OCTA).

Pigment in the macula on CFP, best-corrected VA, presence of ORHR, presence of ORNV, presence and surface of ellipsoid zone (EZ) loss, and occurrence of ORHR, ORNV, and pigment.

At presentation, 13 eyes showed no EZ loss, and no ORHR nor change was observed during the follow-up. In the 19 eyes with only EZ loss at baseline, ORHR appeared in 6 eyes, and one of them also showed visible pigment on Cassociated with rapid vision loss.

Spectral-domain OCT and OCTA showed that in MacTel 2 eyes, ORHR was associated with ORNV on OCTA and most often did not correspond to pigmentation on CFP. When ORNV developed, hyperreflectivity appeared along the abnormal capillary tracts after a mean follow-up of 36 months. link3 In most cases, the presence of ORHR was not associated with rapid vision loss.

To evaluate the clinical characteristics and investigate the role of surgical antibiotic prophylaxis (SAP) in acute endophthalmitis cases after cataract surgery.

Retrospective, consecutive case series.

A total of 126 patients referred to a tertiary center from 2007 to 2015 for acute endophthalmitis after unilateral cataract surgery.

All patients who underwent a vitreous biopsy were included. Clinical and microbiology data were reviewed, and associations with visual outcome were analyzed using multivariate logistic regression. Data regarding SAP via intracameral injection were also retrieved.

Bacterial culture results and visual acuity outcome.

Bacterial growth was observed in 92 of 126 cases (73%). Among these positive cultures, 49 (53.3%), 29 (31.5%), and 13 (14.1%) were coagulase-negative staphylococci, other gram-positive, and gram-negative bacteria, respectively. Among the 77 gram-positive strains tested, 76 (98.7%) were vancomycin-sensitive; among the 12 gram-negative strains tested, all 12 (100%) were ceftazidime-sensitive.
