Despite years of research, therapeutics for AD are limited. Overexpression of secretory glutaminyl cyclase (sQC) in AD brain leads to the formation of a highly neurotoxic pyroglutamate variant of amyloid beta, pGlu-Aβ, which acts as a potential seed for the aggregation of full length Aβ. Preventing the formation of pGlu-Aβ through inhibition of sQC has become an attractive disease-modifying therapy in AD. In this current study, through a pharmacophore assisted high throughput virtual screening, we report a novel sQC inhibitor (Cpd-41) with a piperidine-4-carboxamide moiety (IC50 = 34 μM). Systematic molecular docking, MD simulations and X-ray crystallographic analysis provided atomistic details of the binding of Cpd-41 in the active site of sQC. The unique mode of binding and moderate toxicity of Cpd-41 make this molecule an attractive candidate for designing high affinity sQC inhibitors.Bone remodeling is a complex event that maintains bone homeostasis. The epigenetic mechanism of the regulation of bone remodeling has been a major research focus over the past decades. Histone acetylation is an influential post-translational modification in chromatin architecture. Acetylation affects chromatin structure by offering binding signals for reader proteins that harbor acetyl-lysine recognition domains. This review summarizes recent data of histone acetylation in bone remodeling. The crux of this review is the functional role of histone acetyltransferases, the key promoters of histone acetylation. The functional regulation of acetylation via noncoding RNAs in bone remodeling is also discussed. Understanding the principles governing histone acetylation in bone remodeling would lead to the development of better epigenetic therapies for bone diseases.Fatty acids-assisted superparamagnetic maghemite (γ-Fe2O3) NPs was biologically synthesized using extract of polyherbal drug Liv52 (L52E). The NPs were characterized by UV-vis spectroscopy, FT-IR, SEM, TEM, EDX, XRD and VSM. The major biological molecules present in L52E analysed by GC-MS were saturated fatty acids (palmitic acid 21.95%; stearic acid 13.99%; myristic acid 1.14%), monounsaturated fatty acid (oleic acid 18.43%), polyunsaturated fatty acid (linoleic acid 20.45%), and aromatic phenol (cardanol monoene 11.92%) that could imply in bio-fabrication and stabilization of γ-Fe2O3 NPs. The FT-IR spectra revealed involvement of carboxylic group of fatty acids, amide group of proteins and hydroxyl group of phenolic compounds that acts as reducing and capping agents. The synthesized NPs were used to investigate their antimicrobial, antibiofilm activity against P. aeruginosa, MRSA and C. albicans and anticancer activity on colon cancer cells (HCT-116) for biomedical applications. Further, molecular docking study was performed to explore the interaction of Fe2O3 NPs with major cell wall components i.e., peptidoglycan and mannoproteins. The docking studies revealed that Fe2O3 interacted efficiently with peptidoglycan and mannoproteins and Fe2O3 get accommodated into catalytic cleft of mannoprotein. Due to magnetic property, the biological activity of γ-Fe2O3 can be further enhanced by applying external magnetic field alone or in amalgamation with other therapeutics drugs.In this work, the influence of Sodium Acetate Trihydrate (SAT) on the gelling stage of a chitin hydrogel was studied. Characterization techniques, such as FTIR, Raman, solid-state NMR, Dielectric Spectroscopy, Small-angle X-ray scattering (SAXS), Wide-angle X-ray scattering (WAXS), and X-ray diffraction (XRD) were used to study the effect of SAT on the micro and nanostructure of the material in the wet, dry and freeze-dried states. It was demonstrated that the amount of SAT in the gelling solution can induce a variation in the supramolecular interaction among the polysaccharide chains, which leads to a change in the structural characteristics. In addition, it was observed that the polymer-water interactions are also altered by this structural ordering. Also, the affinity interaction with lysozyme was evaluated and an influence on the adsorption capacity was evidenced with the use of SAT. This could be an advance for biotechnological, biomedical, and food applications.Emotional eating is defined as the tendency to increase food consumption in order to modify negative emotional states. Theories on emotional eating attribute its cause to inadequate emotion regulation, specifically an inability to draw awareness to and accept distress. Mindfulness, or the ability to pay attention to and accept internal and external experiences, is thought to attenuate the association between internalizing distress and emotional eating. Nevertheless, there has been little research examining the moderating role of mindfulness in the relationship between psychological distress and emotional eating. The present study used a cross-sectional design to probe the effects of specific facets of mindfulness, as measured by the Five Factor Mindfulness Questionnaire, on the relationship between internalizing distress (i.e., depression and anxiety) and emotional eating in a diverse community sample (N = 248). Results showed that depression significantly interacted with nonjudging of inner experience to predict emotional eating.
