Suggestions for improvements of SimpleBox refining POP risk assessment are provided. V.Cadmium (Cd) contamination from mining and smelting operations has led to growing environmental health concerns. In this study, soil, surface water, drinking water, rice, vegetables, and biomarkers (hair and urine) were collected from local residents near an active lead-zinc mine and a copper smelter. The aim was to determine how nonferrous metal mining and smelting activities have affected the health of local residents. It was found that the Cd concentrations in most soil and rice samples exceeded the national tolerance limits of China. Dietary intakes of rice and vegetables were the two major pathways of Cd exposure to local residents, accounting for >97% of the total probable daily intake. The excessive daily intake of Cd resulted in potential non-carcinogenic risks to the local residents, especially to children living around the two areas. The mean hair and urine Cd concentrations were 0.098 ± 0.10 mg kg-1 and 5.7 ± 3.1 μg L-1 in the mining area, and 0.30 ± 0.21 mg kg-1 and 5.5 ± 3.5 μg L-1 in the smelting area, respectively. A significantly positive correlation between hair Cd concentrations and the hazard quotient (HQ) for rice ingestion indicated that rice contamination had the most critical adverse effect on local residents. Due to the high levels of environmental Cd contamination, residents of the smelting area had a much higher Cd exposure than residents of the mining area. The results suggested that nonferrous mining and smelting should not coexist with agricultural activities. Effective contamination mitigation strategies and environmental remediation should be formulated and implemented to improve the health of local residents. Increasing attention has been paid to information intervention in stimulating household energy conservation. However, for group-level energy users who have no financial motivation to save energy, little is known about which types of non-financial information intervention (NFII) strategies can effectively motivate group-level energy conservation. A 14-week controlled field experiment was conducted to test the energy-saving effect of 4 types of NFII strategies. The results show that compared with the control group, the experimental group receiving normative information and group-contrast feedback decreased electricity consumption by 24.23%. Followed by the group receiving normative information and self-contrast feedback and the group receiving environmental education information and group-contrast feedback. The group receiving environmental education information and self-contrast feedback showed no significant energy savings. Analysis of variance further showed that normative information had a better energy-saving effect than environmental education information, and the energy-saving effects produced by group-contrast feedback were better than that of self-contrast feedback. These conclusions provide insights and recommendations on how to use NFII to motivate group-level energy conservation. Immune checkpoint inhibitors (ICIs) targeting against programmed cell death-1(PD-1) and cytotoxic T-lymphocyte antigen-4 (CTLA-4) have shown efficacy in cancer treatment. However, a spectrum of immune-related adverse events (irAEs) have raised concerns about their clinical application. IrAEs are distinct from traditional chemo- and radiotherapy-induced toxicities, as they are related in particular to the dysregulation of immune system and autoimmunity. The underlying pathogenesis of irAEs remains elusive. Understanding of the potential underlying mechanism is of great importance for the management of irAEs and the development of new ICIs with insignificant irAEs. In this review, we summarize the current evidence to provide insights into the biological basis of irAEs and provide a potential explanation for their pathogenesis, with focus on the relationship between checkpoint molecules and immune cell regulation. Inflammation is a colorectal cancer (CRC) hallmark. Inflammasome-dependent cytokines IL-1ß and IL-18 can play a beneficial or detrimental role in tumorigenesis depending on cancer type. Variants in inflammasome genes were associated with tumor development and/or outcome, and have been proposed as potential biomarkers for population screening. In this study, 215 CRC patients followed-up for 10 years were examined for 9 polymorphisms in selected inflammasome genes. Multivariate association analysis and survival analysis were performed to evaluate the association between the polymorphisms and CRC prognosis. Variants associated with lower levels of IL-18 (rs1834481, rs5744256), or with increased activation of inflammasome receptors NLRP1 (rs12150220) and NLRP3 (rs35829419) resulted detrimental to CRC prognosis and may be used as prognostic markers. The genetic causes of these diseases can be identified using whole exome sequencing (WES). Here, DNA samples from 106 patients with a clinical suspicion of PID were subjected to WES in order to test the diagnostic yield of this test in a highly consanguineous community. A likely genetic diagnosis was achieved in 70% of patients. Several factors were considered to possibly influence the diagnostic rate of WES among our cohort including early age, presence of consanguinity, family history suggestive of PID, the number of family members who underwent WES and the clinical phenotype of the patient. The highest diagnostic rate was in patients with combined immunodeficiency or with a syndrome. Notably, WES findings altered the clinical management in 39% (41/106) of patients in our cohort. Our findings support the use of WES as an important diagnostic tool in patients with suspected PID, especially in highly consanguineous communities.
