Anthraquinones are a major class of compounds naturally occurring in Asphodelus microcarpus. The pharmacological actions of anthraquinones in cancer cells are known to induce apoptosis or autophagy, and revert multidrug resistance. In this study, five anthraquinone-type analogs were isolated from the methanol extract of A. microcarpus leaves and identified as, emodin, rhein, physcion, aloe-emodin, and emodic acid. Among them, aloe-emodin and emodic-acid strongly inhibited the proliferation, cells-intrinsic NF-κB activity and metastatic ability of breast cancer. Although aloe-emodin inhibited p38 and ERK phosphorylation, emodic-acid more markedly inhibited JNK, in addition to p38 and ERK phosphorylation. Both aloe-emodin and emodic-acid inhibited the secretion of the pro-tumorigenic cytokines IL-1β and IL-6, and VEGF and MMP expression, and subsequently inhibited the invasive and migratory potential of 4T1 cells. Thus, our study demonstrated the effects of aloe-emodin and emodin-acid in controlling the migratory and invasive ability of 4T1 breast cancer cells, in addition to inhibiting NF-κB activity and the expression of its downstream target molecules.In the vertebrate retina, signals generated by cones of different spectral preference and by highly sensitive rod photoreceptors interact at various levels to extract salient visual information. The first opportunity for such interaction is offered by electrical coupling of the photoreceptors themselves, which is mediated by gap junctions located at the contact points of specialised cellular processes synaptic terminals, telodendria and radial fins. Here, we examine the evolutionary pressures for and against interphotoreceptor coupling, which are likely to have shaped how coupling is deployed in different species. The impact of coupling on signal to noise ratio, spatial acuity, contrast sensitivity, absolute and increment threshold, retinal signal flow and colour discrimination is discussed while emphasising available data from a variety of vertebrate models spanning from lampreys to primates. We highlight the many gaps in our knowledge, persisting discrepancies in the literature, as well as some major unanswered questions on the actual extent and physiological role of cone-cone, rod-cone and rod-rod communication. Lastly, we point toward limited but intriguing evidence suggestive of the ancestral form of coupling among ciliary photoreceptors.Vaccine hesitancy is an obstacle to achieving high vaccination rates for COVID-19. Current knowledge on vaccine uptake is mostly based on hypothetical intention to vaccinate surveys. We compared intention to vaccinate and real-world vaccine uptake among 511 soldiers in a military unit during an unrestricted, on-site COVID-19 vaccine rollout. Soldiers were offered group lectures, on-site consultations and primary care office visits, discussing concerns on vaccination with a primary care physician. Overall, 359 (70.3%) soldiers participated in the group lectures, 33 (6.5%) in on-site consultations and 19 (3.7%) attended primary care visits. Overall, 459 (89.8%) of 511 soldiers vaccinated for COVID-19. Of the 90 soldiers initially refusing, 38 (42.2%) had agreed to receive a vaccine. On-site COVID-19 vaccine rollout joined with primary care communication interventions may maximize vaccine uptake within a young-adult community. Future studies should evaluate the effectiveness of these efforts across different populations in a controlled and comparative manner.The COVID-19 pandemic brought forward the challenge of dispersing accurate medical information to the public rapidly. Credible and non-credible sources may impact public reactions to the virus. The purpose of this study is to assess those reactions of women located in or near Kansas. A survey was conducted in July 2020 with questions on knowledge of COVID-19, attitudes and behaviors towards COVID-19, and primary sources of information. 305 survey respondents met criteria for further analysis, and descriptive statistical analyses were applied. Participants were generally knowledgeable of the pandemic, with a mean knowledge score of 11.40 out of 13 (SD 1.3). The attitude statement with the highest rate of agreement was that "social distancing is an effective way of controlling COVID-19 spread" (n = 265, 86.9%) and that with the highest rate of disagreement was, "I am not worried about my friends' and family members health" (n = 253, 83.0%). The most-implemented behaviors as indicated by participants were avoiding contact with sick individuals and washing hands with soap and water often (n = 294, 96.4%), and the least implemented was avoiding meat consumption (n = 257, 84.3%). Finally, most participants indicated that health officials were their primary source of information (n = 215, 70.5%). Participants of this survey had fairly good knowledge of the virus. Attitudes of participants as a whole may be described as cautious without being overly fearful. Reported behaviors also align well with current public health recommendations. These responses may be reflective of where participants are receiving their information, which, for the majority, is from public health officials.Individuals with Autism Spectrum Disorder (ASD) and the Broad Autism Phenotype (BAP) are more likely than individuals with typical development (TD) to report a sexual minority orientation (e.g., Bejerot and Eriksson, PLoS ONE 91-9, 2014; DeWinter et al., Journal of Autism and Developmental Disorders 472927-2934, 2017; Qualls et al., Journal of Autism and Developmental Disorders 483974-3983, 2018). This study operationalized and tested the fit of an existing model of sexual orientation to examine which factors are associated with increased sexual minority orientation (Worthington et al., The Counseling Psychologist 30496-531, 2002) in individuals with TD, BAP, and ASD. The model was found to have adequate fit, χ2 (130) = 374.04, p less then 0.001; RMSEA = 0.07; CFI = 0.95; SRMR = 0.08. Rucaparib Heterosexism was found to be the only predictor of sexual minority orientation and a significant predictor in the BAP and ASD groups, with increased daily heterosexist experiences predicting greater sexual minority orientation in these groups.Transportation and mobility for community participation is difficult for persons with Autism Spectrum Disorders (ASD) under normal circumstances, but the impact of COVID-19 made access even more challenging. Researchers used a single-subject design to examine patterns of change from before and after the COVID-19 pandemic in community mobility and participation as measured by GPS and daily participation questionnaires. Participants were young adults with ASD between the ages of 21 and 27 (4 males, 2 females) who were enrolled in a subsequent study. Community mobility and participation decreased for all participants in both essential and non-essential activities. Additionally, the number of trips for participants decreased substantially in the after COVID-19 periods, as did the variability in modes of transit.Provisioning ecosystem services play a vital role in sustaining human well-being. Agro-ecosystems contribute a significant share of these services, besides food and fodder and also fuel and fibre as well as regulating and cultural ecosystem services. Until now, the indication of provisioning ecosystem services of agro-ecosystems has been based almost only on yield numbers of agricultural products. Such an indication is problematic due to several reasons which include a disregard of the role of significant anthropogenic contributions to ecosystem service co-generation, external environmental effects and strong dependence on site conditions. We argue for an enhanced indication of provisioning ecosystem services that considers multiple aspects of their delivery. The conceptual base for such an indication has been made by prior publications which have been reviewed. Relevant points were taken up in this article and condensed into a conceptual model in order to develop a more holistic and expanded set of indictors, which was then exemplarily applied and tested in three case studies in Germany. The case studies represent different natural conditions, and the indicator set application showed that ecosystem services (ES) flow-in terms of output alone-does not characterise agro-ecosystems sufficiently. The proposed aspects of provisioning ecosystem services can give a fuller picture, for example, by input-output relationships, as it is possible by just using single indicators. Uncertainties as well as pros and cons of such an approach are elaborated. Finally, recommendations for an enhanced indication of provisioning ecosystem services in agro-ecosystems that can help to integrate agricultural principles with ideas of sustainability and site-specific land use are derived.The aim of this study was to evaluate the antibacterial efficacy of copper added chitosan nanoparticles (CU-CNPs) as an irrigation solution with different irrigants in terms of eliminating Enterococcus Faecalis (E. faecalis) from the root canals. Fifty mandibular premolar teeth were prepared and infected with E. faecalis for 21 days. After the incubation period, samples were randomly divided into a control group irrigated with distilled water and 4 experimental groups (n = 10) irrigated with as follows, %6 NaOCl, %6 NaOCl + %9 editronate (HEBP), Chitosan nanoparticles (CNPs), and CU-CNPs. To calculate the proportion of dead E. faecalis cell volume, stained using LIVE/DEAD BacLight Bacterial Viability Kit and were scanned using confocal laser scanning microscope (CLSM). All the irrigation solutions significantly (P less then .05) killed the bacteria in the canal, except for the control group. CU-CNPs solution killed the highest (P less then .05) number of bacteria compared with the other experimental groups. No significant difference was found between CNPs, NaOCl + HEBP, and NaOCl in terms of antibacterial activity. CU-CNPs solution was exhibited higher antibacterial efficacy against E. faecalis.
Available intracystic biomarkers show a limited accuracy for characterizing cystic pancreatic lesions (CPL). Glucose is an attractive alternative due to its availability, low cost and the possibility of on-site quantification by glucometry.
To evaluate the diagnostic accuracy of on-site glucometry from samples obtained by EUS-FNA in the differential diagnosis between mucinous from non-mucinous CPL.
Retrospective, multicentre, cross-sectional study of patients who underwent EUS-FNA of a CPL. A derivation and a validation cohorts were evaluated. Intracystic glucose was quantified by on-site glucometry and colorimetry in the lab. Final diagnosis was based on surgical specimens or global evaluation of clinical and imaging data, cytology and intracystic CEA. Diagnostic accuracy was based on Receiver Operating Curve (ROC) curve analysis. Intraclass correlation coefficient (ICC) between on-site and lab glucose levels was calculated.
Seventy two patients were finally analysed (40 in the derivation cohort and 32 in the validation cohort). Intracystic glucose levels by on-site glucometry was 12.3 ± 28.2 mg/dl for mucinous CPL and 103.3 ± 58.2 mg/dl for non-mucinous CPL, p < 0.001. For an optimal cut-off point of 73 mg/dl, on-site glucose had a sensitivity, specificity, and positive and negative predictive value for the diagnosis of mucinous CPL of 0.89, 0.90, 0.94, 0.82 respectively in the derivation cohort, and 1.0, 0.71, 0.91, 1.0 respectively in the validation cohort. Correlation of on-site and lab glucose quantification was very high (ICC = 0.98).
On-site glucometry is a feasible, accurate and reproducible method for the characterization of CPL after EUS-FNA. It shows an excellent correlation with laboratory glucose values. Registration number 2019/612.
