To report electroretinographic (ERG) findings in advanced glaucoma treated with a single intravitreal injection of bone marrow-derived mesenchymal stem cells (MSCs).

Intravitreal injection of autologous MSCs (1 × 10

cells) was performed in 2 eyes from 2 patients with open-angle glaucoma in advanced stage of optic neuropathy (ClinicalTrials.gov, NCT02330978, 01.05.2015) cup/disk ratio worse than 0.9, visual field mean deviation index lower than -15dB, visual acuity of light perception, but controlled intraocular pressure. ERG tests were recorded at baseline and week 1, 4 and 48 after injection, using DTL electrodes following the ISCEV standard After dark adaptation, ERG was elicited using white flashes of 0.01cd.s/m

and 3.0cd.s/m

, followed by 10-min light adaptation (30cd/m

) and stimuli of 3.0cd.s/m

and 30Hz flicker.

Patients did not show improvement on visual acuity or visual field after treatment. At baseline, ERG responses showed typical findings for advanced glaucoma, with a- and b-wave amplitude and latency within normative range, but reduced photopic negative responses. No noteworthy changes were observed on ERG responses for both cases up to 1week after treatment, but at day 15, one patient showed retinal detachment with proliferative vitreoretinopathy and was removed from the trial. The other patient kept ERG responses stable throughout study period.

Although no ERG response changes were observed after MSCs injection in one case, the complication observed on the second one, along with the lack of visual function improvement, warrants further studies involving modified MSCs to treat ocular disorders, including glaucoma.

POLG mutations are a major cause of inherited mitochondrial diseases. They present with varied phenotypes, age of onset, and severity. Reports on POLG mutations from India are limited. Hence, this study aimed to describe the clinico-pathological and molecular observations of POLG mutations. A total of 446 patients with clinical diagnosis of mitochondrial disorders were sequenced for all exons and intron-exon boundaries of POLG. S28463 Of these, 19 (4.26%) patients (MF 109) had POLG mutations. The age of onset ranged from 5 to 55 years with an overlapping phenotypic spectrum. Ptosis, peripheral neuropathy, seizures, and ataxia were the common neurological features observed. The most common clinical phenotype was chronic progressive external ophthalmoplegia (CPEO) and CPEO plus (n = 14). Muscle biopsy showed characteristic features of mitochondrial myopathy in fourteen patients (14/19) and respiratory chain enzyme deficiency in eleven patients (11/19). Multiple mtDNA deletions were seen in 47.36% (9/19) patients. Eight pathogenic POLG variations including two novel variations (p.G132R and p.V1106A) were identified. The common pathogenic mutation identified was p.L304R, being present in eight patients (42.1%) predominantly in the younger age group followed by p.W748S in four patients (21%). To the best of our knowledge, this is the first extensive study from India, highlights the clinico-pathological and molecular spectrum of POLG mutations.Perspective-taking ability is crucial for supporting social interactions. It has been widely suggested that the calculation of an individual's perspective is spontaneous. Nevertheless, people typically engage with more than one individual, and computing what individuals in a crowd see is important. The current study explored whether people spontaneously compute the perspectives of individuals displayed in a crowd. The classic visual perspective-taking task was adopted, but the picture of the room was presented with four human avatars facing two walls. The results showed that if the crowd of individuals was treated as a high entitative group, when none of the perspectives of the individuals contained the same number of discs as that from the perspective of the participant, the judgment of the participant's perspective was slower than when a proportion of the perspectives of the individuals displayed in the crowd were consistent with the participant's perspective, even if the perspectives of the multiple individuals in a crowd were not explicitly noticed. This altercentric intrusion effect was not present when the crowd had low entitativity. These findings were replicated by using different methods to operationalize group entitativity. Hence, this study demonstrates that spontaneously tracking the perspectives of individuals displayed in a crowd has a boundary condition and that people can spontaneously compute what individuals in high entitative groups see.Visual processing near the hands is altered compared with stimuli far from the hands. Here, we aimed to test whether this alteration can be found in auditory processing. Participants were required to perform an auditory Simon task either with their hands close to the loudspeakers or far from the loudspeakers. Two experiments consistently showed that the auditory Simon effect was enhanced when the hands were close to the speakers compared with far from the speakers. This is consistent with previous findings of an enhanced visual Simon effect near the hands. Furthermore, the hand-proximity effects in auditory and visual Simon tasks (an enhanced Simon effect near hands compared with far from hands) were comparable, indicating hand-proximity effect is reliable across visual and auditory modalities. Thus, the present study extended the hand-proximity effect from vision to audition by showing that the auditory Simon effect was enhanced near the hands compared with far from the hands.

The aim of this study was to evaluate the use of a Bayesian penalized likelihood reconstruction algorithm (Q.Clear) for

Zr-immunoPET image reconstruction and its potential to improve image quality and reduce the administered activity of

Zr-immunoPET tracers.

Eight

Zr-immunoPET whole-body PET/CT scans from three

Zr-immunoPET clinical trials were selected for analysis. On average, patients were imaged 6.3 days (range 5.0-8.0 days) after administration of 69 MBq (range 65-76 MBq) of [

Zr]Zr-DFO-daratumumab, [

Zr]Zr-DFO-pertuzumab, or [

Zr]Zr-DFO-trastuzumab. List-mode PET data was retrospectively reconstructed using Q.Clear with incremental β-values from 150 to 7200, as well as standard ordered-subset expectation maximization (OSEM) reconstruction (2-iterations, 16-subsets, a 6.4-mm Gaussian transaxial filter, "heavy" z-axis filtering and all manufacturers' corrections active). Reduced activities were simulated by discarding 50% and 75% of original counts in each list mode stream. All reconstructed PET images were scored for image quality and lesion detectability using a 5-point scale.
