To our knowledge, there is no report of an optimal method for the extraction of acetogenins from the Annonaceae family by ultrasound-assisted extraction (UAE); therefore, the aim was to find the best UEA conditions of acetogenins from A. muricata fruit (peel, pulp, seed, and columella) by using response surface methodology. The effect of amplitude (40%, 70%, and 100%), time (5, 10, and 15 min), and pulse-cycle (0.4, 0.7, and 1 s) of ultrasound at 24 kHz was evaluated on the total acetogenin content (TAC). Optimal extraction conditions of acetogenins (ACGs) with UEA were compared with the extraction of ACGs by maceration. The optimal UEA conditions in the A. muricata pulp and by-products were dependent on each raw material. The highest TAC was found in the seed (13.01 mg/g dry weight (DW)), followed by the peel (1.69 mg/g DW), the pulp (1.67 mg/g DW), and columella (1.52 mg/g DW). The experimental TAC correlated well with the model (Adjusted R2 with values between 0.88 and 0.97). The highest effectiveness in ACG extraction was obtained in seeds and peels using UEA compared to extraction by maceration (993% and 650%, respectively). The nature of the latter molecules (e.g., enzymes, receptors, DNA, proteins, etc.) and the strength of the binding to such chemicals elicit a toxic effect in organisms, which magnitude depends on the doses exposed in a given timeframe. While dose and time of exposure are critical factors determining the toxicity of pesticides, different types of chemicals behave differently. Experimental evidence demonstrates that the toxicity of neonicotinoids increases with exposure time as much as with the dose, and therefore it has been described as time-cumulative toxicity. However, youth runners younger than 19 years of age are also competing in ultra-endurance events, and an increase has been observed in distance-limited events, but no data is available on time-limited ultra-endurance events in this age group. This study investigated participation and performance trends in time-limited ultra-endurance races, including multi-day events, in runners younger than 19 years of age. Between the period 1990 and 2018, the most popular events recorded a total of 214 finishes (from 166 unique finishers (UF)) for 6-hour events, 247 (212 UF) for 12-hour events, and 805 (582 UF) for 24-hour events, respectively. The majority of athletes originated from Europe and North America. Only a minority participated in multi-day events. Overall, speed increased with age, but the overall performance speed decreased across calendar years for 6- and 24-hour events as participation numbers grew. In summary, in youth ultra-endurance runners, differences were observed regarding participation and performance across the different time-limited events, the age of the athletes and their country of origin.(1) Background Applied health services research (AHSR) relies upon coordination across multiple organizational boundaries. Our aim was to understand how competing organizational and professional goals enhance or impede the conduct of high quality AHSR. (2) Methods A qualitative study was conducted in two local health care systems in the UK, linked to a feasibility trial of a clinic-based intervention in secondary care. Data collection involved 24 semi-structured interviews with research managers, clinical research staff, health professionals, and patients. (3) Results This study required a dynamic network of interactions between heterogeneous health and social care stakeholders, each characterized by differing ways of organizing activities which constitute their core functions; cultures of collaboration and interaction and understanding of what research involves and how it contributes to patient care. (4) Conclusions Despite the strategic development of multiple organizations to foster inter-professional collaboration, the competing goals of research and clinical practice can impede the conduct of high quality AHSR. To remedy this requires the alignment and streamlining of organizational goals, so that all agencies involved in AHSR develop a shared understanding and mutual respect for the progress of evidence-based medicine and the complex and often nuanced environments in which it is created and practiced.
