Segregation is a common problem in batch-based direct compression (BDC) processes, especially with low-dose tablet products, as is the preparation of a homogenous mixture. The scope of the current work was to explore if a continuous direct compression (CDC) process could serve as a solution for these challenges. Furthermore, the principle of a platform formulation was demonstrated for low dose tablets. The combination of filler excipients and the API in the formulation used was suitable for direct compression, but also prone to induce segregation in BDC process. The CDC process was found to be very promising; it was shown that tablets with the desired quality parameters could be manufactured successfully with both of the APIs studied. Powder analysis indicated that the APIs display some fundamental differences in their physical properties, which was also reflected in powder mixture properties and, hence, eventually in processing. However, process parameters, especially mixer impeller speed, were not found to have any significant influence on end product quality. The present work describes the frequency of IFD in a specialized laboratory in Colombia. A retrospective, descriptive study was implemented between March 2009 and December 2015. Results 13,071 patients with clinical suspicion of IFD were referred during the study period, from which 33,516 biological samples were processed and analyzed using 14 laboratory methods. Diagnosis was confirmed in 1425 patients (11%), distributed according to the mycoses of interest analyzed here histoplasmosis in 641/11,756 patients (6%), aspergillosis in 331/10,985 patients (3%), cryptococcosis in 239/8172 patients (3%), pneumocystosis in 111/1651 patients (7%), paracoccidioidomycosis in 60/10,178 patients (0.6%), and invasive candidiasis in 48/7525 patients (0.6%). From the first year of the study period to the last year, there was a 53% increase in the number of cases of IFD diagnosed. Our laboratory experienced a high frequency of IFD diagnosis, possibly attributable to the availability of a greater range of diagnostic tools. Frequency of IFD in this study was atypical compared with other studies, probably as a result of the single laboratory-site analysis. This demonstrates that implementing educational strategies helps to create a high index of clinical suspicion, while the availability and utilization of appropriate diagnostic assays assure greater reliability in identification of these cases.Parasitic isopod Ceratothoa oestroides (Cymothoidea, Isopoda) is a common and generalist buccal cavity-dweller in marine fish, recognised for its detrimental effect in fingerling and juvenile farmed European sea bass (Dicentrarchus labrax). Although distributed throughout the Mediterranean, the isopod provokes acute outbreaks mainly limited to particular endemic areas in Croatia (Adriatic Sea) and Greece (Aegean Sea). While numerous studies have previously evidenced its gross effect on farmed fish (i.e. decreased condition index, slower growth rate, lethargy and mortality), details on the host-parasite interaction are still lacking. Therefore, using a multimethodological approach, we closely examined the structure and appearance of isopod body parts acting in the attachment and feeding (stereomicroscopy, scanning and transmission electron microscopy), and the extent of host tissues damage (histology, immunohistochemistry, micro-computational tomography) induced by parasitation. Interestingly, while hematophagous nature of the parasite has been previously postulated we found no unambiguous data to support this; we observed host tissues fragmentation and extensive hyperplasia at the parasitation site, and no structures indicative of heme detoxifying mechanisms in the parasite gut, or other traces of a blood meal. The bacterial biofilm covering C. oestroides mouthparts and pereopods suggests that the isopod may play a role in conveying secondary pathogens to the infected host, or alternatively, it serves the parasite in normal interaction with its environment.Alcohol consumed by breastfeeding mothers has been associated with reduced grammatical comprehension and cognition in children. This study examined whether drinking or smoking while breastfeeding was associated with reductions in Australian National Assessment Program-Literacy and Numeracy assessments. Data was sourced from The Growing Up in Australia Study. This is an ongoing longitudinal study of 5107 infants and mothers recruited in 2004 and followed over time every two years. Multivariable linear regression found that maternal alcohol consumption at study entry was associated with reductions in Grade 3 (age 7-10 years) National Assessment Program-Literacy and Numeracy writing (b = -1.56, 95% CI -2.52; -0.60, p = 0.01), spelling (b = -2.06, 95% CI -3.31; -0.81, p less then 0.0001) and grammar and punctuation (b = -2.11, 95% CI -3.59; -0.64, p = 0.01) scores, as well as Grade 5 (age 9-11 years) spelling scores (b = -1.58, 95% CI -2.74; -0.43, p = 0.03) in children who had been breastfed at any time. This was not evident in babies who had never breastfed, or in the smaller group of infants who were actively breastfeeding at study entry. Smoking was not associated with any outcome variable. Drinking alcohol while breastfeeding may result in dose-dependent reductions in children's academic abilities. While reductions are small, they may be of clinical significance if mothers drink large quantities. Despite the availability of attenuated vaccines against CDV, the virus remains responsible for outbreaks of canine distemper (CD) with significant morbidity and mortality in domesticated and wild carnivores worldwide. CDV uses the signaling lymphocytic activation molecule (SLAM, or CD150) and nectin-4 (PVRL4) as entry receptors, well-known tumor-associated markers for several lymphadenomas and adenocarcinomas, which are also responsible for the lysis of tumor cells and apparent tumor regression. Thus, CDV vaccine strains have emerged as a promising platform of oncolytic viruses for use in animal cancer therapy. Recent advances have revealed that use of the CDV reverse genetic system (RGS) has helped increase the understanding of viral pathogenesis and explore the development of recombinant CDV vaccines. In addition, genetic engineering of CDV based on RGS approaches also has the potential of enhancing oncolytic activity and selectively targeting tumors.
