This study aimed to clarify the regulatory role of Th17-Treg balance in periodontitis and further reveal Treg plasticity.

An experimental periodontitis model was established by ligation and injection of Pg-LPS. Inflammatory factors were measured by ELISA and RT-PCR. Alveolar bone absorption was evaluated by micro-CT and histomorphology. Quantities of Treg and Th17 cell and their related gene expression were examined. Furthermore, after magnetic bead-sorting spleen Treg cells, Treg/Th17 characteristic genes were explored. Immunofluorescence double staining of Foxp3 and IL-17 was conducted to further reveal Treg plasticity.

Inflammatory cytokines in serum and gingival tissue increased significantly in periodontitis, which revealed obvious crestal bone loss. Further analysis showed that the number of Th17 cells and expression of related genes increased more significantly than Treg cells, demonstrating Treg-Th17 imbalance. Flow cytometry showed that the proportions of Treg cells in the blood and spleen were lower in periodontitis group. Emergency Department (ED) visits are known to be anxiety-ridden and stress-provoking experiences especially in the pediatric population. Distraction techniques have been used as a means to reduce anxiety and stress thereby facilitating care in the ED and making the visit less unpleasant. Our study aimed to evaluate the effectiveness of an active and immersive distraction technique, using a mindfulness-based virtual reality (VR) program (Take-Pause), to alleviate anxiety in pediatric ED patients.

A prospective, randomized, single-blinded study, evaluating ED patients aged 13-17 years with a chief complaint of acute pain was conducted. Patients were randomized to either the active distraction intervention (VR group), utilizing the VR headset for 5 minutes; or to the passive distraction intervention (iPad group), playing on an iPad for 5 minutes. The primary outcome was a difference in the change in anxiety scores on the Spielberger State-Trait Anxiety Inventory between the two groups. Secondary outcomes included a difference in pain scores, respiratory rate (RR) and heart rate (HR) between the groups.

However, the functional significance of this pathway during seed development and germination remains to be elucidated. To assess the function of this pathway, we performed a detailed metabolic and transcriptomic analysis of Arabidopsis mutants to test the molecular consequences of a dysfunctional ETF/ETFQO pathway. We demonstrate that the disruption of this pathway compromises seed germination in the absence of an external carbon source and also impacts seed size and yield. Total protein and storage protein content is reduced in dry seeds, whilst sucrose levels remain invariant. Seeds of ETFQO and related mutants were also characterized by an altered fatty acid composition. During seed development, lower levels of fatty acids and proteins accumulated in the etfqo-1 mutant as well as in mutants in the alternative electron donors isovaleryl-CoA dehydrogenase (ivdh-1) and d-2-hydroxyglutarate dehydrogenase (d2hgdh1-2). Furthermore, the content of several amino acids was increased in etfqo-1 mutants during seed development, indicating that these mutants are not using such amino acids as alternative energy source for respiration. Transcriptome analysis revealed alterations in the expression levels of several genes involved in energy and hormonal metabolism. Our findings demonstrated that the alternative pathway of respiration mediated by the ETF/ETFQO complex affects seed germination and development by directly adjusting carbon storage during seed filling. Ion mobility spectrometry (IMS) is a powerful analytical tool extensively applied in numerous domains. However, it still exists the phenomenon of peaks overlapping in analysis of isomers with similar structures due to the limited resolution of IMS. In this paper, a dynamic inertia weight particle swarm optimization algorithm (DIWPSO) combined with second-order differentiation is proposed to separate IMS overlapping peaks efficiently and precisely.

It can identify the component number of overlapping peaks and limit those parameters (ion mobility, intensity and full-width at half maximum of each single peak) of peak model in a small range by second-order differentiation. Based on this, DIWPSO that has been set the best operating parameters, is capable to accurately separate IMS overlapping peaks to identify compound within a short time.

After comparing the performance of DIWPSO with the improved particle swarm optimization (IPSO), it is found that DIWPSO with separation errors less than 2.34% on the whole outperforms IPSO whose maximum error is up to 5.58%. Moreover, the running time of DIWPSO is 30 ~ 80 times less than that of IPSO, and DIWPSO possesses stronger robustness.

The two major DDR cell cycle checkpoints control either the replication process or the G2/M transition.
