Atypical fetal chromosomal anomalies are more frequent than previously recognized and can affect fetal development. We propose a screening strategy for a genome-wide non-invasive prenatal test (NIPT) to detect these atypical chromosomal anomalies (ACAs). Two sample cohorts were tested. Assay performances were determined using Cohort A, which consisted of 192 biobanked plasma samples-42 with ACAs, and 150 without. The rate of additional invasive diagnostic procedures was determined using Cohort B, which consisted of 3097 pregnant women referred for routine NIPT. Of the 192 samples in Cohort A, there were four initial test failures and six discordant calls; overall sensitivity was 88.1% (37/42; CI 75.00-94.81) and specificity was 99.3% (145/146; CI 96.22-99.88). In Cohort B, there were 90 first-pass failures (2.9%). The rate of positive results indicating an anomaly was 1.2% (36/3007) and 0.57% (17/3007) when limited to significant unbalanced chromosomal anomalies and trisomies 8, 9, 12, 14, 15, 16, and 22. These results show that genome-wide NIPT can screen for ACAs with an acceptable sensitivity and a small increase in invasive testing, particularly for women with increased risk following maternal serum screening and by limiting screening to structural anomalies and the most clinically meaningful trisomies.Urban and peri-urban agricultural (UPA) production systems in West African countries do not only mitigate food and financial insecurity, they may also foster biodiversity of arthropods and partly compensate for structural losses of natural environments. However, management practices in UPA systems like irrigation may also contribute to disturbances in arthropod ecology. To fill knowledge gaps in the relationships between UPA management and arthropod populations, we compared arthropods species across different irrigation sources in Tamale. During a 72-h sampling period, 14,226 arthropods were caught with pitfall traps and pan traps from 36 fields. These specimens comprised 13 orders, 103 families, 264 genera, and 329 taxa (243 identified species, 86 unidentified species) and categorized into five feeding guilds (carnivores, decomposers, herbivores, omnivores, and pollinators). Species richness, species accumulation curves, and diversity functions (richness, evenness, and dispersion) were calculated to characterize the arthropod community. Non-metric multidimensional scaling was applied to examine structural similarity of arthropod communities among sites. To account for the effects of soil-related data, we furthermore applied a redundancy analysis. Arthropods grouped according to the irrigation water source, whereby the dipterans were most dominant under wastewater conditions. Here, particularly the eye gnat, Hippelates pusio, a disease-causing vector for humans, accounted for the dipterans. The occurrence of three alien ant species suggested community shifts through invasive species, while the occurrence of seven ant species (at least one ant species occurred under each water source) that form mutualistic relationships with aphids highlighted future risks of aphid pest outbreak. Future studies on these taxa should specifically target their ecological and economic effects and potential countermeasures.Over the past century, the emergence/reemergence of arthropod-borne zoonotic agents has been a growing public health concern. The present review examines the ecology, epidemiology, disease, past outbreaks, and potential to cause contemporary outbreaks for several alphavirus pathogens.Our aim was to investigate the underlying assumptions of the current gestational weight gain (GWG) paradigm, specifically that-(1) GWG is modifiable through diet and physical activity; (2) optimal GWG and risk of excess GWG, vary by pre-pregnancy body mass index (BMI) category and (3) the association between GWG and adverse pregnancy outcomes is causal. Using data from three large, harmonized randomized controlled trials (RCTs) of interventions to limit GWG and improve pregnancy outcomes and with appropriate regression models, we investigated the link between diet and physical activity and GWG; the relationships between pre-pregnancy BMI, GWG and birth weight z-score; and the evidence for a causal relationship between GWG and pregnancy outcomes. We found little evidence that diet and physical activity in pregnancy affected GWG and that the observed relationships between GWG and adverse pregnancy outcomes are causal in nature. Further, while there is evidence that optimal GWG may be lower for women with higher BMI, target ranges defined by BMI categories do not accurately reflect risk of adverse outcomes. Our findings cast doubt upon current advice regarding GWG, particularly for overweight and obese women and suggest that a change in focus is warranted.

This work aimed to evaluate the efficacy of sonic agitation of a binary mixture of solvents (methyl ethyl ketone/tetrachloroethylene) on filling remnants removal and compare the effects of solvent agitation with the enlargement to the next instrument size.

Twenty-four mandibular incisors were prepared with ProTaper Next (X1, X2) and obturated with the single-cone technique and AH Plus sealer. The teeth were retreated with ProTaper Universal Retreatment and ProTaper Next and divided into two groups (

= 12) according to the final instrument (X3 or X4). All canals were submitted to a supplementary procedure consisting of a mixture of solvents-methyl ethyl ketone/tetrachloroethylene, agitated with EndoActivator. The volume of filling remnants was assessed through micro-computed tomography in the apical 5 mm. Statistical analysis was performed with a significance level of 5%.
