FIIN-2 datasheet Here we show that pH-responsive zeolitic imidazolate framework-8 (ZIF-8) offers a novel solution to mutp53 degradation. ZIF-8 facilitated ubiquitination-mediated and glutathionylation-dependent proteasomal degradation of all of the nine mutp53 we tested, including six hot-spot mutp53, but not the wild-type p53 protein. Sustained elevation of intracellular Zn++ level, resulted from decomposition of the internalized ZIF-8 in the acidic endosomes, decreased the intracellular reduced glutathione (GSH) oxidized glutathione (GSSG) ratio and was essential for mutp53 glutathionylation and degradation. ZIF-8 modified with an Z1-RGD peptide, exhibiting enhanced cellular internalization and improved decomposition behavior, preferentially killed mutp53-expressing cancer cells and demonstrated remarkable therapeutic efficacy in a p53 S241F ES-2 ovarian cancer model as well as in a p53 Y220C patient-derived xenograft (PDX) breast cancer model. The ability to induce wide-spectrum mutp53 degradation gives ZIF-8 a clear advantage over other degradation-inducers, and engineered nanomaterials may be promising alternatives to small molecules for the development of mutp53-targeting drugs.We executed a pot experiment to examine the differences of absorption, chemical forms, subcellular distribution, and toxicity of Cd between two cultivars of Chinese flowering cabbage Brassica campestris [Lvbao701 (low-Cd cultivar) and Chicaixin No.4 (high-Cd cultivar)]. Compared to Chicaixin No.4, the presence of Lvbao701 enhanced the proportion of insoluble Cd forms in soil, Lvbao701 roots and leaves had higher proportion of Cd converted into insoluble phosphate precipitates and pectate-or protein-bound forms and lower proportion of inorganic Cd, which result in low accumulation and toxicity of Cd to Lvbao701 and cutworm Spodoptera litura fed on Lvbao701 leaves. Instead of total Cd, Cd transfer and toxicity in B. campestris-S. litura system depend on chemical Cd forms in soil and cabbages and subcellular Cd distributions in cabbages and insects, and the proportions of them were not the highest among all chemical forms and subcellular distributions of Cd. Although exchangeable Cd was major Cd chemical form in cabbage planted soil, Cd bound to iron and manganese oxides and to organic matter were significantly correlated with growth indices and photosynthesis parameters of cabbages. Despite major part of Cd was precipitated in cell wall of roots, Cd in organelle fraction was closely associated with the fitness of cabbages. Metal-rich granules, not cytosolic fraction (the major subcellular Cd distribution), affected the food utilization of S. litura. Therefore, cabbage cultivars significantly affected Cd transfer and toxicity in B. campestris-S. litura system, and the use of Lvbao701 in Cd polluted soil could reduce potential risks for Cd entering food chains.Seasonal variations in hydrogeochemical characteristics of groundwater were assessed from an intensive agricultural region to identify contaminants of concern that are a potential risk to human health. A total of 116 groundwater samples were collected grid-wise from an intensive agricultural region of confined Wanaparthy watershed to evaluate seasonal variations in hydrogeochemical processes of dissolved ions, nitrate health risk assessment and water quality during pre-monsoon (PRM) and post-monsoon (POM) seasons. The major ions concentration found in ascending order as PRM F- 1 with 68.97% of infants and 72.41% of children are in danger for non-carcinogenic ingestion of nitrate contaminated groundwater than in adults.
Hamstring autografts are commonly used for anterior cruciate ligament (ACL) reconstruction. Different folding techniques exist in tripling the semitendinosus. Few anatomical studies exist in evaluating their viscoelastic properties. The purpose of this study was to characterize and compare the viscoelastic and failure properties of two hamstring graft configurations, the "Z" construct and "2" construct.
Ten matched pairs of fresh-frozen cadaveric semitendinosus hamstring grafts were used to create the "2" configuration or "Z" configuration. The biomechanical testing consisted of four phases preconditioning, where graft dimensions (mm) were measured; stress relaxation, where load, displacement and time data were collected and equilibrium relaxation (%) was calculated; dynamic creep, where the total construct elongation was calculated; and ramp-to-failure, where maximum failure load was recorded.
The "2" configuration demonstrated recorded forces (N) significantly greater at each time point when compared gnificance of these findings.
Previous research has shown the effectiveness of simulation training using avatars paired with feedback in improving child sexual abuse interview quality. However, it has room for improvement.
The present study aimed to determine if the combination of two interventions, feedback and modeling, would further improve interview quality compared to either intervention alone.
Thirty-two clinical psychologists were randomly assigned to a feedback, modeling, or the combination of feedback and modeling group.
The participants conducted five simulated child sexual abuse interviews online while receiving the intervention(s) corresponding to their allocated group. Feedback was provided after each interview and consisted of the outcome of the alleged cases and comments on the quality of the questions asked in the interviews. Modeling was provided after the 1st interview and consisted of learning points and videos illustrating good and bad questioning methods.
The proportion of recommended questions improved over the five interviews when considering all groups combined.
