Due to the increasing demand for eco-friendly, cost-effective and safe technologies, biosynthetic metal nanoparticles have attracted worldwide attention. In this study, silver nanoparticles (AgNPs) were extracellularly biosynthesized using the culture supernatants of Aspergillus sydowii. During synthesis, color change was preliminarily judge of the generation of AgNPs, and the UV absorption peak at 420 nm further confirms the production of AgNPs. Transmission electron microscopy and X-ray diffraction were also used to identify the AgNPs. The results shows that AgNPs has crystalline cubic feature and is a polydisperse spherical particle with size between 1 and 24 nm. Three main synthesis factors (temperature, pH and substrate concentration) were optimized, the best synthesis conditions were as follows 50 °C, 8.0 and 1.5 mM. In the biological application of AgNPs, it shows effective antifungal activity against many clinical pathogenic fungi and antiproliferative activity to HeLa cells and MCF-7 cells in vitro. Our research finds a new path to biosynthesis of AgNPs in an eco-friendly manner, and bring opportunity for biomedical applications in clinic. The development of inorganic antibacterial agents that impart antibacterial properties to biomaterials has attracted wide attention. The paper introduced a kind of hybrid nanosphere antibacterial agent composed of wheat gliadin (WG) and zinc oxide (ZnO), with antibacterial efficacy and low toxicity. The ZnO/WG hybrid nanospheres were environment-friendly integrated by molecular self-assembly co-precipitating and freeze-drying transformation, and were characterized using X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), scanning electron microscope (SEM), atomic absorption spectroscopy (AAS), specific surface and pore size analysis, bacteriostasis test, reactive oxygen species (ROS) determination and safety evaluation. It was found that the prepared hybrid nanospheres were composed of two components, WG and ZnO, with a diameter scope of 100-200 nm; the content of ZnO in the hybrid nanospheres can reach 46.9-70.2% (w/w); the bacteriostasis tests proved that the prepared ZnO/WG nanospheres generating ROS, have a significant inhibitory effect on E. coli and S. aureus; furthermore, the ZnO/WG nanospheres are relatively safe and highly biocompatible in cells and mice. Therefore, the prepared novel ZnO/WG hybrid nanospheres were supposed to apply in the preparation of anti-infective wound dressings, tissue engineering skin scaffold materials, food, and cosmetics preservatives, and so on.Trypophobia is induced by viewing multiple clustered objects. To date, several studies have investigated why certain people experience discomfort when looking at clustered patterns. Recently, "involuntary protection against dermatosis" (IPAD) hypothesis was proposed to explain the causes of trypophobia. The IPAD hypothesis suggests that involuntary aversive responses to skin diseases cause discomfort in response to image clusters. However, this idea has not been fully investigated empirically. Therefore, the present study used a modified version of the priming procedure and tested whether the activation of the concept of skin-related diseases affected the evaluation of trypophobic images. In Experiment 1, participants engaged in a lexical decision task in which words related to skin problems, negative events, or neutral events were presented. Then, they evaluated the discomfort of trypophobic, negative, and neutral images. The results indicated that participants evaluated trypophobic images as more discomforting after they were exposed to skin-problem-related words, whereas the exposure to words related to skin-problems did not enhance the discomfort of negative images. These findings demonstrate that the association with skin-related problems increases the discomfort of trypophobic images. In Experiment 2, we further tested the reproducibility of the priming effect observed in Experiment 1 and investigated the effect of priming with words related to COVID-19 in the context of a spreading infection. Contrary to predictions, no priming effect was produced by either skin-related words or COVID-19-related words. Future studies should further explore the causal relationship of the association between skin disease and trypophobia. The innate response of melanocytes to exogenous or endogenous stress stimuli like extreme pH and temperature, metabolite and oxygen deficiency or a high UV dose initiates a cellular stress response. This process activates adaptive processes to minimize the negative impact of the stressor on the pigment cell. Under physiological conditions, a non-cancer cell is directed to apoptosis if the stressor persists. However, malignant melanoma cells will survive persistent stress thanks to distinct "cancerous" signaling pathways (e.g. MEK) and transcription factors that regulate the expression of so-called "survival genes" (e.g. HIF, MITF). In this survival response of cancer cells, MEK pathway directs melanoma cells to deregulate mitochondrial metabolism, to accumulate reduced species (NADH), and to centralize metabolism in the cytosol. The aim of this work was to study the effect of gene silencing in malignant melanoma A375 cells on metabolic processes in cytosol and mitochondria. Gene silencing of HIF-1α, and miR-210 in normoxia and pseudohypoxia, and analysis of its effect on MITF-M, and PDHA1 expression. Detection of cytosolic NADH by Peredox-mCherry Assay. Detection of OCR, and ECAR using Seahorse XF96. Measurement of produced O2•- with MitoTracker Red CMXRos. 1H NMR analysis of metabolites present in cell suspension, and medium. By gene silencing of HIF-1α and miR-210 the expression of PDHA1 was upregulated while that of MITF-M was downregulated, yielding acceleration of mitochondrial respiratory activity and thus elimination of ROS. Hence, we detected a significantly reduced A375 cell viability, an increase in alanine, inositol, nucleotides, and other metabolites that together define apoptosis. Based on the results of measurements of mitochondrial resipiratory activity, ROS production, and changes in the metabolites obtained in cells under the observed conditions, we concluded that silencing of HIF-1α and miR-210 yields apoptosis and, ultimately, apoptotic cell death in A375 melanoma cells.Ziziphus jujuba var. spinosa has been used as a windbreak and for soil conservation and water retention. Previous studies focused on pharmacological effects and extraction of chemical components in this species, and very few explored the breeding system. The present study combined the analysis of floral morphology, behavior of flower visitors, and artificial pollination to reveal reproductive characteristics of the species. Its flowers are characterized by dichogamy, herkogamy, and stamen movement, which are evolutionary adaptations to its breeding system. There were more than 40 species of visiting insects, mainly Hymenoptera and Diptera, and the characteristics of dichogamous and herkogamous flower adapted to the visiting insects. The breeding system is outcrossing, partially self-compatible, and demand for pollinators. The fruit setting rate after natural pollination was 2%. Geitonogamy and xenogamy did not significantly increase the fruit setting rate, indicating that the low fruit setting rate was not due to pollen limitation by likely caused by resource limitation or fruit consumption. The fruit setting rate of zero in emasculated and in naturally and hand self-pollinated individuals suggested the absence of apomixis and spontaneous self-pollination. The above results can be utilized in studies on evolution and cultivation of Z. jujuba var. spinosa.Sensors for structural health monitoring (SHM) need to be permanently integrated on structures and withstand the harsh service environments, which has been a big challenge for the application of SHM in aircrafts. This paper focuses on the durability of flexible eddy current array (FECA) sensors in harsh service environments of aircrafts, including vibration environment and several typical exposed environments. First, a kind of FECA sensor is illustrated and its integration method is proposed. Moreover, in order to study the durability of the sensor in vibration environment, the modal analysis is performed by the finite element method. According to the simulation results, the durability experiment in vibration environment is carried out under the fourth order vibration mode, which makes the sensor suffer the harshest vibration loads. During the vibration experiment, output signals of the sensor keep stable and the sensor is well bonded to the structure, which shows the integrated sensor has high durability in vibration environment. Finally, the durability of integrated sensors is separately tested in three exposed environments, including salt fog corrosion environment, fluid immersion environment, as well as hygrothermal and ultraviolet-radiation environment. After these environmental exposure experiments, all sensors are well bonded to structures and can effectively monitor fatigue cracks, which shows great durability. Therefore, FECA sensors can survive in harsh service environments of aircrafts, which provides important support for the engineering applications of FECA sensors.Site productivity remains a fundamental concern in forestry as a significant driver of resource availability for tree growth. The site index (SI) reflects the overall impact of all environmental factors that determine tree height growth and is the most commonly used indirect proxy for forest site productivity estimated using stand age and height. The SI concept challenges are local variations in climate, soil, and genotype-environmental interactions that lead to variable height growth patterns among ecoregions and cause inappropriate estimation of site productivity. Developing regional models allow us to determine forest growth and SI more appropriately. selleck products This study aimed to develop height growth models for the Scots pine in Poland, considering the natural forest region effect. For height growth modelling, we used the growth trajectory data of 855 sample trees, representing the Scots pine entire range of geographic locations and site conditions in Poland. We compared the development of regional height growth mth interactions with site variables, such as climate, soil properties, and topography, can provide valuable forest management information.DNA N6-methylation (6mA) in Adenine nucleotide is a post replication modification responsible for many biological functions. Automated and accurate computational methods can help to identify 6mA sites in long genomes saving significant time and money. Our study develops a convolutional neural network (CNN) based tool i6mA-CNN capable of identifying 6mA sites in the rice genome. Our model coordinates among multiple types of features such as PseAAC (Pseudo Amino Acid Composition) inspired customized feature vector, multiple one hot representations and dinucleotide physicochemical properties. It achieves auROC (area under Receiver Operating Characteristic curve) score of 0.98 with an overall accuracy of 93.97% using fivefold cross validation on benchmark dataset. Finally, we evaluate our model on three other plant genome 6mA site identification test datasets. Results suggest that our proposed tool is able to generalize its ability of 6mA site identification on plant genomes irrespective of plant species. An algorithm for potential motif extraction and a feature importance analysis procedure are two by products of this research.
