In this study, different non-lethal high temperatures and treatment durations were used to stress the third-instar larvae of B. odoriphaga, and the effects of this treatment on their growth and offspring were recorded and analyzed. The results showed that the average larval mortality increased with increased temperature and prolonged exposure times. After stress treatment at 40 °C for 2 h, 100% of larvae died within 5 days, which was not significantly different from the 5-day average larval mortality (90.66%) after stress at 37 °C for 4 h, but significantly higher than the 5-day average larval mortality (72.00%) after stress at 40 °C for 1 h. After 5 days, all still-living larvae could pupate, and there was no significant difference in average pupal period after pupation. However, the eclosion rate of subsequent pupae decreased with increased temperature and prolonged exposure times, and were only 43.00% and 42.73% after larvae were stressed at 37 °C for 4 h and 40 °C for 1 h, respectively. After eclosion into adults, there was no significant difference in the lifespan of unmated female adults, while the lifespan of unmated male adults was significantly reduced to 1.67 d and 2 d after larvae were stressed at 37 °C for 4 h and 40 °C for 1 h, respectively. However, there was no significant difference in male and female adult longevity after mating. There was no significant difference in oviposition or egg hatchability. This indicates that non-lethal high temperature at 37 °C for 4 h can hinder development and allow control of B. odoriphaga. There is great potential for non-lethal high temperature to be applied in the field to control agricultural pests.Illusory visual phenomena, such as palinopsia, polyopsia or allesthesia, are rare manifestations of posterior cortical damage. Symptoms are characterized by illusory perceptions, ranging from isolated stationary objects to scenes and moving persons. Such illusions may appear while the original object is still in view, or become manifest with a delay and last for minutes, hours or even days. Some authors have suggested a disinhibited cortical response underlying visual illusions, but experimental studies supporting this hypothesis are lacking. Here, we examined a rare patient who after focal right parietal injury consistently reported a second stimulus on the left when briefly shown a target in his right hemifield. The patient perceived the illusory stimulus as less intense, and therefore concluded that it must have a different shape than the original stimulus. A masking experiment revealed that the frequency of the illusion was inversely related to the visibility of the original stimulus, suggesting that it depended on early, feedforward visual processing. Currently, I. tinctoria is utilized more often as medicinal remedy and also as a cosmetic ingredient. In 2011, I. tinctoria root was accepted in the official European phytotherapy by introducing its monograph in the European Pharmacopoeia. The biological properties of raw material have been known from Traditional Chinese Medicine (TCM). Over recent decades, I. tinctoria has been investigated both from a phytochemical and a biological point of view. The modern in vitro and in vivo scientific studies proved anti-inflammatory, anti-tumour, antimicrobial, antiviral, analgesic, and antioxidant activities. The phytochemical composition of I. tinctoria has been thoroughly investigated and the plant was proven to contain many valuable biologically active compounds, including several alkaloids, among which tryptanthrin, indirubin, indolinone, phenolic compounds, and polysaccharides as well as glucosinolates, carotenoids, volatile constituents, and fatty acids. This article provides a general botanical and ethnobotanical overview that summarizes the up-to-date knowledge on the phytochemistry and biological properties of this valuable plant in order to support its therapeutic potential. They provide input for future research prospects.BACKGROUND The approvals of immune checkpoint inhibitors for several cancer types and the rapidly growing recognition that T cell-based immunotherapy significantly improves outcomes for cancer patients led to a re-emergence of cancer vaccines, including dendritic cell (DC)-based immunotherapy. Blood and tissue biomarkers to identify responders and long-term survivors and to optimize cost and cost-effectiveness of treatment are greatly needed. We wanted to investigate whether blood eosinophilia is a predictive biomarker for patients with solid tumors receiving vaccinations with DCs loaded with autologous tumor-RNA. METHODS In total, 67 patients with metastatic solid tumors, who we treated with autologous monocyte-derived DCs transfected with total tumor mRNA, were serially analyzed for eosinophil counts and survival over the course of up to 14 years. Eosinophilic counts were performed on peripheral blood smears. RESULTS Up to 87% of the patients treated with DC-based immunotherapy experienced at least once an ed DC, specifically autologous tumor mRNA-transfected DC vaccines, and it correlates with long-term patient outcome. Although primate studies report the pulvinar is engaged under attentional demands, there are open questions about the detailed role of the pulvinar in visuospatial attention. The pulvinar provides the primary thalamic input to the posterior parietal cortex (PPC). Both the pulvinar and the PPC are known to be important for visuospatial attention. Our previous work showed that neuronal activity in the PPC correlated with multiple phases of a visuospatial attention (VSA) task, including onset of the visual stimuli, decision-making, task-relevant locations, and behavioral outcomes. Here, we hypothesized that the pulvinar, as the major thalamic input to the PPC, is involved in visuospatial attention as well as in other cognitive functions related to the processing of visual information. We recorded the neuronal activity of the pulvinar in rats during their performance on the VSA task. The task was designed to engage goal-directed, top-down attention as well as stimulus-driven, bottom-up attention. Rats monitored three possible locations for the brief appearance of a target stimulus. An approach to the correct target location was followed by a liquid reward. For analysis, each trial was divided into behavioral epochs demarcated by stimulus onset, selection behavior, and approach to reward. We found that neurons in the pulvinar signaled stimulus onset and selection behavior consistent with the interpretation that the pulvinar is engaged in both bottom-up and top-down visuospatial attention. Our results also suggested that pulvinar cells responded to allocentric and egocentric task-relevant locations.The sterilization processes of nanoparticles (NP) by autoclaving and filtration are two of the most utilized methods in the pharmaceutical industry but are not always a viable option. For this reason, the search for alternative options such as UV and gamma radiation is of interest. In this work, we evaluated both types of sterilization on two types of NP in solid state widely employed in the literature for biomedical applications, poly-(ε-caprolactone) and poly(D, L-lactide-co-glycolide) acid NP stabilized with polyvinyl alcohol. Physicochemical properties and cell viability were studied pre- and post-sterilization. The efficiency of irradiation sterilization was performed by a test of sterility using 1 × 108 CFU/mL of Escherichia coli, Staphylococcus aureus, and Candida albicans. Microbiological monitoring revealed that both methods were sufficient for sterilization. After the UV irradiation sterilization (100 µJ/cm2), no substantial changes were observed in the physicochemical properties of the NP or in the interaction or morphology of human glial cells, though 5 and 10 kGy of gamma irradiation showed slight changes of NP size as well as a decrease in cell viability (from 100 µg/mL of NP). At 5 kGy of radiation doses, the presence of trehalose as cryoprotectant reduces the cell damage with high concentrations of NP, but this did not occur at 10 kGy. Therefore, these methods could be highly effective and low-processing-time options for sterilizing NP for medical purposes. However, we suggest validating each NP system because these generally are of different polymer-composition systems.A gateless lateral p-n junction with reconfigurability is demonstrated on graphene by ion-locking using solid polymer electrolytes. Ions in the electrolytes are used to configure electric-double-layers (EDLs) that induce p- and n-type regions in graphene. These EDLs are locked in place by two different electrolytes with distinct mechanisms (1) a polyethylene oxide (PEO)-based electrolyte, PEOCsClO4, is locked by thermal quenching (i.e., operating temperature less then Tg (glass transition temperature)), and (2) a custom-synthesized, doubly-polymerizable ionic liquid (DPIL) is locked by thermally triggered polymerization that enables room temperature operation. Both approaches are gateless because only the source/drain terminals are required to create the junction, and both show two current minima in the backgated transfer measurements, which is a signature of a graphene p-n junction. The PEOCsClO4 gated p-n junction is reconfigured to n-p by resetting the device at room temperature, reprogramming, and cooling to T less then Tg. In this study, gene encoding Haemonchus contortus tropomyosin (Hc-TpMy), was successfully cloned and expressed, and the recombinant protein after host cell surface attachment was evaluated for immune functional analysis with goat peripheral blood mononuclear cells (PBMCs) in vitro. The isopropyl-β-D-thiogalactopyranoside (IPTG)-induced recombinant protein was successfully recognized by the sera of rat experimentally infected with rHc-TpMy. The immunofluorescence assay detected attachment of rHc-TpMy on the surface of host PBMCs. Furthermore, immunoregulatory roles of rHc-TpMy on cytokines expression, PBMC proliferation, migration, nitric oxide (NO) production, apoptosis and monocytes phagocytosis were observed. The results showed that expression of IL-4 and IFN-γ cytokines, cell proliferation, NO production and PBMC migration were significantly suppressed by goat PBMCs after co-incubation with rHc-TpMy protein.
