Most corals, except P. damicornis, were capable of adjusting to a turbidity treatment involving a mean light level of 2.3 mol photons m-2 d-1 in conjunction with a SSC of 10 mg L-1 over the 7 week period.Phasic alerting cues temporarily increase the brain's arousal state. In younger and older participants, visual processing speed in a whole report task, estimated based on the theory of visual attention, is higher in cue than no-cue conditions. The present study assessed whether older participants' ability to profit from warning cues is related to intrinsic functional connectivity (iFC) in the cingulo-opercular and/or right fronto-parietal network. We acquired resting-state functional magnetic resonance imaging data from 31 older participants. By combining an independent component analysis and dual regression, we investigated iFC in both networks. A voxel-wise multiple regression in older participants yielded that higher phasic alerting effects on visual processing speed were significantly related to lower right fronto-parietal network iFC. This result supports a particular role of the right fronto-parietal network in maintaining phasic alerting capabilities in aging. We then compared healthy older participants to a previously reported sample of healthy younger participants to assess whether behaviour-iFC relationships are age group specific. The comparison revealed that the association between phasic alerting and cingulo-opercular network iFC is significantly lower in older than in younger adults.The aim of the present study was to explore whether kinematic indicators could improve the detection of subjects demonstrating faking-good behaviour when responding to personality questionnaires. One hundred and twenty volunteers were randomly assigned to one of four experimental groups (honest unspeeded, faking-good unspeeded, honest speeded, and faking-good speeded). Participants were asked to respond to the MMPI-2 underreporting scales (L, K, S) and the PPI-R Virtuous Responding (VR) scale using a computer mouse. The collected data included T-point scores on the L, K, S, and VR scales; response times on these scales; and several temporal and spatial mouse parameters. These data were used to investigate the presence of significant differences between the two manipulated variables (honest vs. faking-good; speeded vs. unspeeded). The results demonstrated that T-scores were significantly higher in the faking-good condition relative to the honest condition; however, faking-good and honest respondents showed no statistically significant differences between the speeded and unspeeded conditions. Concerning temporal and spatial kinematic parameters, we observed mixed results for different scales and further investigations are required. The most consistent finding, albeit with small observed effects, regards the L scale, in which faking-good respondents took longer to respond to stimuli and outlined wider mouse trajectories to arrive at the given response.Resonators with multi-frequency generations at the device-level are highly desired in the future multi-band, reconfigurable, and compact wireless communications. In this work, a switchable radio frequency micro-electro-mechanical system (RF-MEMS) resonator with multiple electrodes is presented. The resonator is designed to operate at the whispering gallery modes (WGMs). Simultaneous excitations of the second to seventh modes with high Q values are implemented within a single device using a pair of electrodes for driving and sensing. For effective multi-frequency excitations, the electrode span angle is optimized. BAY 2416964 chemical structure The frequencies of the 37 μm and 18 μm-radius resonators range from 53 MHz to 176 MHz and 112 MHz to 366 MHz, respectively. The Q values in each mode are over 104. Moreover, with the multi-electrode configurations, the specific mode can be enhanced with other modes suppressed. A more than 6 dB improvement of the spectrum peak is realized and the high Q values are maintained. A comprehensive theory is built up to clarify the driving/sensing principles under different electrode configurations. Furthermore, the air damping is found to have a significant effect on Q values for resonator with high stiffness vibrating in all the WGMs. The Q values in vacuum have at least two times improvement. The high-performance switchable resonator could dramatically reduce the power consumption, simplify the processing circuits, and occupy less footprint, which has great potential applications in future advanced RF front end systems.Abnormal keratinocyte differentiation is fundamental to pathologies such as skin cancer and mucosal inflammatory diseases. The ability to grow keratinocytes in vitro allows the study of differentiation however any translational value is limited if keratinocytes get altered by the culture method. Although serum lipids (SLPs) and phenol red (PR) are ubiquitous components of culture media their effect on differentiation is largely unknown. We show for the first time that PR and SLP themselves suppress expression of differentiation-specific keratins K1, K10 and K2 in normal human epidermal keratinocytes (NHEK) and two important cell lines, HaCaT and N/TERT-1. Removal of SLP increased expression of K1, K10 and K2 in 2D and 3D cultures, which was further enhanced in the absence of PR. The effect was reversed for K1 and K10 by adding all-trans retinoic acid (ATRA) but increased for K2 in the absence of PR. Furthermore, retinoid regulation of differentiation-specific keratins involves post-transcriptional mechanisms as we show KRT2 mRNA is stabilised whilst KRT1 and KRT10 mRNAs are destabilised in the presence of ATRA. Taken together, our results indicate that the presence of PR and SLP in cell culture media may significantly impact in vitro studies of keratinocyte differentiation.Quantum dot (QD) coupling in nanophotonics has been widely studied for various potential applications in quantum technologies. Micro-machining has also attracted substantial research interest due to its capacity to use miniature robotic tools to make precise controlled movements. In this work, we combine fluorescent QDs and magnetic nanoparticles (NPs) to realize multifunctional microrobotic structures and demonstrate the manipulation of a coupled single-photon source (SPS) in 3D space via an external magnetic field. By employing the low one photon absorption (LOPA) direct laser writing (DLW) technique, the fabrication of 2D and 3D magneto-photonic devices containing a single QD is performed on a hybrid material consisting of colloidal CdSe/CdS QDs, magnetite Fe3O4 NPs, and SU-8 photoresist. Two types of devices, contact-free and in-contact structures, are investigated to demonstrate their magnetic and photoradiative responses. The coupled SPS in the devices is driven by the external magnetic field to perform different movements in a 3D fluidic environment. The optical properties of the single QD in the devices are characterized.Wolfram syndrome (WS) is a recessive multisystem disorder defined by the association of diabetes mellitus and optic atrophy, reminiscent of mitochondrial diseases. The role played by mitochondria remains elusive, with contradictory results on the occurrence of mitochondrial dysfunction. We evaluated 13 recessive WS patients by deep clinical phenotyping, including optical coherence tomography (OCT), serum lactic acid at rest and after standardized exercise, brain Magnetic Resonance Imaging, and brain and muscle Magnetic Resonance Spectroscopy (MRS). Finally, we investigated mitochondrial bioenergetics, network morphology, and calcium handling in patient-derived fibroblasts. Our results do not support a primary mitochondrial dysfunction in WS patients, as suggested by MRS studies, OCT pattern of retinal nerve fiber layer loss, and, in fibroblasts, by mitochondrial bioenergetics and network morphology results. However, we clearly found calcium mishandling between endoplasmic reticulum (ER) and mitochondria, which, under specific metabolic conditions of increased energy requirements and in selected tissue or cell types, may turn into a secondary mitochondrial dysfunction. Critically, we showed that Wolframin (WFS1) protein is enriched at mitochondrial-associated ER membranes and that in patient-derived fibroblasts WFS1 protein is completely absent. These findings support a loss-of-function pathogenic mechanism for missense mutations in WFS1, ultimately leading to defective calcium influx within mitochondria.Highly fluorescent blue and green-emitting carbon dots have been designed to be integrated into sol-gel processing of hybrid organic-inorganic materials through surface modification with an organosilane, 3-(aminopropyl)triethoxysilane (APTES). The carbon dots have been synthesised using citric acid and urea as precursors; the intense fluorescence exhibited by the nanoparticles, among the highest reported in the scientific literature, has been stabilised against quenching by APTES. When the modification is carried out in an aqueous solution, it leads to the formation of silica around the C-dots and an increase of luminescence, but also to the formation of large clusters which do not allow the deposition of optically transparent films. On the contrary, when the C-dots are modified in ethanol, the APTES improves the stability in the precursor sol even if any passivating thin silica shell does not form. Hybrid films containing APTES-functionalized C-dots are transparent with no traces of C-dots aggregation and show an intense luminescence in the blue and green range.The increasing incidence of non-healing wounds constitutes a pivotal socio-economic burden. 60-80% of chronic wounds are colonized by pathogenic microorganisms within a protective extracellular polymeric substance, bearing a great challenge in wound management. Human plasma was used to prepare the biofilm model (hpBIOM), adding pathogens to the plasma and forming Coagula-like discs with integrated pathogens were produced. The antiseptics Octenisept and Lavasorb were tested regarding their antibacterial properties on clinically relevant biofilm-growing bacteria (MRSA, P. aeruginosa) in the hpBIOM. Biofilm-typical glycocalyx-formation was confirmed using immunohistochemical staining. Treatment of a 12 h-maturated biofilm with Octenisept resulted in complete eradication of P. aeruginosa and MRSA after 48 h. Lavasorb proved less effective than Octenisept in this setting. In more mature biofilms (24 h), both antiseptics showed a delayed, partially decreased efficacy. Summarized, the hpBIOM provides essential factors for a translational research approach to be used for detailed human biofilm analyses and evaluation of antimicrobial/-biofilm properties of established and novel therapeutic strategies and products. Octenisept and Lavasorb showed an attenuated efficacy in the hpBIOM compared to planktonic conditions and previously published biofilm-studies, prompting the question for the necessity of introducing new international standards and pre-admission requirements on a translational base.Kv7 channels are enriched at the axonal plasma membrane where their voltage-dependent potassium currents suppress neuronal excitability. Mutations in Kv7.2 and Kv7.3 subunits cause epileptic encephalopathy (EE), yet the underlying pathogenetic mechanism is unclear. Here, we used novel statistical algorithms and structural modeling to identify EE mutation hotspots in key functional domains of Kv7.2 including voltage sensing S4, the pore loop and S6 in the pore domain, and intracellular calmodulin-binding helix B and helix B-C linker. Characterization of selected EE mutations from these hotspots revealed that L203P at S4 induces a large depolarizing shift in voltage dependence of Kv7.2 channels and L268F at the pore decreases their current densities. While L268F severely reduces expression of heteromeric channels in hippocampal neurons without affecting internalization, K552T and R553L mutations at distal helix B decrease calmodulin-binding and axonal enrichment. Importantly, L268F, K552T, and R553L mutations disrupt current potentiation by increasing phosphatidylinositol 4,5-bisphosphate (PIP2), and our molecular dynamics simulation suggests PIP2 interaction with these residues.
