Here, in the human somatosensory domain, we assessed effects of subthreshold (imperceptible) electrical finger nerve stimulation - either presented as single pulses or as brief (1 s) 7 Hz pulse trains-on mu-alpha rhythm and perceptual performance. In electroencephalography, subthreshold single pulses transiently (~150-350 ms poststimulus) increased mu activity (event-related synchronization), while, interestingly, subthreshold trains led to prolonged (>1 s) mu desynchronization. In psychophysics, detection of near-threshold target stimuli was consistently reduced when presented together with subthreshold trains (at three delays), whereas for targets paired with subthreshold single pulses detection remained unaffected (30 and 180 ms) or was even elevated (60 ms). Though both imperceptible, single pulses and pulse trains exerted opposite effects on neural signaling and perception. We suggest that the common neural basis is preferential activation of cortical inhibitory interneurons. While the inhibitory impact of a subthreshold single pulse (reflected by mu synchronization) is not psychophysically detectable-rather perception may be facilitated-repetition of the same subthreshold pulse shifts the excitation-inhibition balance toward an inhibitory cortical state (reflected by perceptual impediment) accompanied by mu desynchronization. Mucin type O-linked glycosylation is not limited to mucins and occurs on many cell surface glycoproteins including EGFR, where the number of sites can be limited. Upon EGF ligation, EGFR induces a signalling cascade and may also translocate to the nucleus where it directly regulates gene transcription, a process modulated by Galectin-3 and MUC1 in some cancers. Here we show that upon EGF binding, breast cancer cells carrying different O-glycans respond by transcribing different gene expression signatures. MMP10, the principal gene upregulated when cells carrying sialylated core 1 glycans were stimulated with EGF, is also upregulated in ER positive breast carcinoma reported to express high levels of ST3Gal1 and hence mainly core 1 sialylated O-glycans. In contrast, isogenic cells engineered to carry core 2 glycans upregulate CX3CL1 and FGFBP1 and these genes are upregulated in ER negative breast carcinomas, also known to express longer core 2 O-glycans. Under in-vitro modifications, MGO altered the tertiary structure of LDL. TNBS and phenanthrenequinone assays confirmed lysine and arginine residues as main targets of MGO in LDL. HPLC and LCMS studies confirmed the generation of Nϵ-(carboxymethyl) lysine in the modified protein. Comet assay showing increased tail length of DNA in lymphocytes inferred the cytotoxicity of MGO-LDL. The easy penetration of MGO-LDL into the nucleus is possibly a consequence of its reduced size, post-modification, as observed from the studies on hydrodynamic radii studies in DLS experiments. MGO-LDL was found to be more immunogenic, as compared to native LDL, in immunological studies conducted on experimental rabbits. Our results reflect the presence of neo-antigenic determinants on modified LDL.
