3-89.1 μg) in 100 g. This indicates the prospects for biofortification breeding. Several QTLs associated with carotenoids and folates have been identified in major cereals, and the most promising of these are presented here. Breeding for essential nutrition should be a core objective of next-generation crop breeding. This review synthesizes the available literature on folates, provitamin A, and carotenoids in rice, wheat, maize, and pearl millet, including genetic variation, trait discovery, QTL identification, gene introgressions, and the strategy of genomics-assisted biofortification for these traits. Recent evidence shows that genomics-assisted breeding for grain nutrition in rice, wheat, maize, and pearl millet crops have good potential to aid in the alleviation of micronutrient malnutrition in many developing countries.Circular RNA (circRNA) abnormal expression and regulation are involved in the occurrence and development of a variety of tumors. However, the role of circRNAs still remains unknown in gastrointestinal stromal tumors (GISTs). In the present study, the differential circRNA expression profile of GISTs was screened by human circRNAs chip and verified by qRT-PCR. The circRNA-miRNA-mRNA regulatory network was constructed using the cytoHubba plugin based on the Cytoscape software. Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were performed to explore circRNA functions. Six significantly differential circRNAs were also verified in 20 pairs of GISTs and adjacent tissues by qRT-PCR. The result showed that a total of 543 differentially expressed circRNAs were identified in GISTs, of which 242 were up-regulated and 301 were down-regulated. Additionally, the circRNA-miRNA-mRNA network contained six circRNAs, 30 miRNAs, and 308 mRNAs, and the targeted mRNAs were associated with "regulation of biological process," "intracellular organelle," "protein binding," and enriched in Wnt signaling pathway. Furthermore, qRT-PCR demonstrated that hsa_circRNA_061346, hsa_circRNA_103114, and hsa_circRNA_103870 were significantly up-regulated in GISTs (n = 20), and hsa_ circRNA_405324, hsa_circRNA_406821, and hsa_circRNA_000361 were dramatically down-regulated in GISTs (n = 20). In addition, all of these circRNAs were shown to have high diagnostic values, and most of them were significantly associated with tumor size, mitotic figure, and malignant degrees in GISTs (P less then 0.05). Therefore, we concluded that circRNAs were abnormally expressed in GISTs, and the circRNA-miRNA-mRNA regulatory network plays an important role in the occurrence and development of GISTs. Also, the identified six candidate circRNAs might be critical circRNAs and may present as potential diagnostic biomarkers for GISTs.While our understanding of cellular and molecular processes has grown exponentially, issues related to the cell microenvironment and cellular heterogeneity have sparked a new debate concerning the cell identity. Cell composition (chromatin and nuclear architecture) poses a strong risk for dynamic changes in the diseased condition. Since chromatin accessibility patterns play a major role in human diseases, it is therefore anticipated that a deconvolution tool based on open chromatin data will provide better performance in identifying cell composition. Herein, we have designed the deconvolution tool "DeconPeaker," which can precisely define the uniqueness among subpopulations of cells using open chromatin datasets. read more Using this tool, we simultaneously evaluated chromatin accessibility and gene expression datasets to estimate cell types and their respective proportions in a mixture of samples. In comparison to other known deconvolution methods, we observed the lowest average root-mean-square error (RMSE = 0.042) and the highest average correlation coefficient (r = 0.919) between the prediction and "true" proportion. As a proof-of-concept, we also tested chromatin accessibility data from acute myeloid leukemia (AML) and successfully obtained unique cell types associated with AML progression. Furthermore, we showed that chromatin accessibility represents more essential characteristics in the identification of cell types than gene expression. Taken together, DeconPeaker as a powerful tool has the potential to combine different datasets (primarily, chromatin accessibility and gene expression) and define different cell types in mixtures. The Python package of DeconPeaker is now available at https//github.com/lihuamei/DeconPeaker.RNA interference (RNAi) is a biological process in which double-stranded ribonucleic acid (dsRNA) molecules inhibit protein expression. In recent years, the application of dsRNA has been used in the development of agricultural products for pest control. The 2019 Organisation for Economic Cooperation and Development (OECD) Conference on RNAi Based Pesticides ("the Conference") brought together academic, industry, and government experts in various aspects of RNAi to discuss the current state of knowledge and topics to help in developing considerations for risk assessment. The Conference focused on environment, with some discussion of human health. Along with presentations on the use of dsRNA-based products in agriculture, government regulation, risk assessment, and a background on the Draft OECD Working Paper on "Considerations for the Environmental Risk Assessment of the Application of Sprayed or Externally Applied dsRNA-Based Pesticides" ("OECD Working Paper"), the Conference included panel discussions from presenters at the end of each session and a larger discussion session with Conference participants on the environmental fate of dsRNA, non-target organism (NTO) risk assessment, and human health risk assessment. This paper summarizes input from presenters and Conference participants during these discussions. Key considerations from these discussions have already been incorporated into the OECD Working Paper, that once finalized and published, will facilitate regulators in evaluating externally applied dsRNA-based products for potential environmental risks.Several transcription factors have been involved in the regulation of gene expression during seed development. Nutritional reserves, including iron, are principally accumulated during seed maturation stages. Using the model plant Arabidopsis thaliana, it has been shown that iron is stored during seed development in vacuoles of the endodermis cell layer. During seed germination, these iron reserves are remobilized and used by the seedling during the heterotrophic to autotrophic metabolism switch. To date, no information about how iron distribution is genetically regulated has been reported.Ascorbate (AsA), an antioxidant that cannot be synthesized and stored by the human body, plays an essential role in the proper functioning of both plants and humans. With the goal of increasing the AsA level in lettuce, the effects of different ratios of red (R) to blue (B) light (75R25B, 50R50B, and 25R75B) on AsA pool sizes as well as the transcript levels and activities of key enzymes involved in AsA metabolism were constantly monitored for 12 days under continuous light (200 μmol⋅m-2⋅s-1) from LEDs. The results showed that lettuce biomass was positively correlated with the ratio of red light, while the AsA pool size had a positive correlation with the ratio of blue light during the whole experiment. The 25R75B treatment increased the expression of genes involved in AsA biosynthesis (GMP, GME, GGP, GPP, GLDH) and regeneration (APX, MDHAR, DHAR, and GR) on day 3 but only significantly elevated the activities of enzymes involved in AsA regeneration (APX, MDHAR, DHAR, and GR) subsequently. AsA regeneration enzymes (MDHAR, DHAR and GR) had greater correlations with the AsA level than the AsA synthesis enzyme (GLDH). Thus, it is concluded that a high ratio of blue light elevated the AsA level mainly by promoting AsA regeneration rather than biosynthesis. Taken together, altering the redblue ratio of continuous light from high to low before harvest is recommended for lettuce cultivation to achieve both high yield and high quality.Viticulture is a cropping system in which treatment against fungal diseases (in particular powdery and downy mildews) can be extremely frequent. Accordingly, a reduction in antimicrobial treatments and the application of environmentally-friendly compounds are becoming increasingly important for a more sustainable viticulture. In addition to their effect against pathogens, the impact of these products on the quality of the grapes is very important for the oenological industries, but unfortunately at present few data are available. We evaluated the effect of the application of biocontrol products and resistance inducers in the vineyard on the mechanical properties, microbial ecology, technological and phenolic maturity of Vitis vinifera "Nebbiolo" grapes at harvest. The yield and vigor of vines were not influenced by the treatments, nor were the production of primary and secondary metabolites. However, the active ingredients influenced the mechanical properties of the skin (hardness and thickness). A significant hardening of the skin was detected when laminarin and chito-oligosaccharides were used, and sulfur induced a thickening of the skin with potential consequences for wine quality. Furthermore, the yeast community present on grape berries was influenced by the treatments. The abundance of Aureobasidium pullulans, the dominant species on the grape berry, changed in response to the compounds used. In addition, Alternaria sp. was reduced in some treatments with a potentially positive effect on the quality and the safety of the grapes. This study provides an overview of the effect of biocontrol products and resistance inducers on microbial ecology and "Nebbiolo" grape quality, contributing to the establishment of more sustainable and effective defense strategies in viticulture.Peach (Prunus persica L. Batsch) is a shallow root fruit tree with poor waterlogging tolerance. Hydrogen sulfide (H2S) is a signal molecule which regulates the adaptation of plants to adverse environments. Nevertheless, the effects of exogenous applications of H2S in fruit tree species especially in peach trees under waterlogging stress have been scarcely researched. Thus, the goal of this research was to investigate the alleviating effect of exogenous H2S on peach seedlings under waterlogging stress. In the present study, we found that the effect of exogenous H2S depended on the concentration and 0.2 mM sodium hydrosulfide (NaHS) showed the best remission effect on peach seedlings under waterlogging stress. Waterlogging significantly reduced the stomatal opening, net photosynthetic rate, and Fv/Fm of peach seedlings. The results of histochemical staining and physiological and biochemical tests showed that waterlogging stress increased the number of cell deaths and amounts of reactive oxygen species (ROS) accan insight into the protective role of H2S in waterlogging-stressed peach seedlings and suggest H2S as a potential candidate in reducing waterlogging-induced damage in peach seedlings. The wide band and air-coupled ultrasonic resonant spectroscopy together with a modified Simulated Annealing metaheuristic algorithm and a 1D layered acoustic-model are used to resolve the structure of plant leaves. In particular, this paper focuses on the extraction of the surface density of the different layers of tissue in leaves having a relatively simple structure. There are three main reasons to select the surface density as the focus of this study (i) it is a parameter directly extracted by the proposed technique and it requires no further processing, (ii) it is relevant in order to study the dynamic of the water within the different tissues of the leaves and also to study the differential development of the different tissues, and (iii) unlike other parameters provided by this technique (like resonant frequency, impedance, ultrasonic elastic modulus, or ultrasonic damping), this parameter can be easier to understand as it is a direct measure of mass per unit surface. The selection of leaves with a simple structure is justified by the convenience of avoiding an unnecessary complication of the data extraction step.
