We have established two human induced pluripotent stem cell clones from a PCD patient carrying a deletion in the NME5 gene (c.415delA (p.Ile139Tyrfs*8)).Here we have generated a human induced pluripotent stem cells (hiPSC) line (MPIi007-A) from skin fibroblasts of a 4-year-old male Metachromatic leukodystrophy (MLD) patient with a heterozygous 1178C > G (Thr393Ser) mutation in arylsulfatase A (ARSA) gene via retroviral expression of OCT4, SOX2, KLF4 and c-MYC. The MPIi007-A iPSC line displayed typical embryonic stem cell-like morphology, carried the ARSA gene mutation, expressed several pluripotent stem cell makers, retained normal karyotype (46, XY) and were capable of forming teratomas containing three germ layers. The MPIi007-A line can be used for the characterization of MLD-associated pathomechanisms and developing new therapeutic options.Low-cost, user-friendly benchtop NMR instruments are often touted as a "one-click" solution for data acquisition, however insufficient peak dispersion in their spectra often reduces the accuracy of quantification and requires user expertise with sophisticated processing tools. Our work aims to facilitate the wide acceptance of benchtop NMR instruments as a viable and effective substitute for cryogenic magnets. We propose an algorithmic approach that completely automates the routine analysis of sets of samples with similar compositions - the problem that often underlies many industrial applications concerned with reaction and process monitoring and quality control. Our solution is rooted in the idea of parametric modelling formulated in terms of Bayesian statistics, which effectively incorporates prior knowledge about the studied system (such as concentration-dependent chemical shift changes) that is usually available in industrial applications. Furthermore, the use of quantum mechanical models for chemical species makes our approach invariant to the spectrometer field strength - a necessary prerequisite for the successful analysis of benchtop data. We demonstrate the performance of our method with two representative sets of samples mixtures of alcohols and acetates, and aqueous mixtures of biologically relevant species. In these examples, our fully automated analysis of benchtop spectra achieves average errors in concentrations of 0.01 mol/mol and 0.02 mol/mol respectively. Our method is competitive with the traditional processing approaches of well resolved high-field data and has the potential to bring the benefits of NMR even to a small chemistry laboratory.

Positive dimension of mental health has gained less attention in previous research on individuals with neurodevelopmenal disorders. However, knowledge on factors associated with mental well-being is crucial for planning effective interventions for this target group.

The aim of the study was to investigate the determinants of positive mental health among young adults with neurodevelopmental disorders.

The study sample consisted of 171 young adults (18-35 years, mean age 25 years, SD 4.35) with a diagnosis of Autism Spectrum Disorder (ASD) or ADHD/ADD. The data were collected with questionnaires.

The mean score of SWEMWBS was 20.98 (n = 168, ranging 14.75-35.00, SD = 3.55). Provisions of social relationships, functional capacity and self-rated state of health associated independently with positive mental health.

Available antileishmanial drugs have several limitations including relatively high toxicity, difficult administration, high production costs and the emergence of resistance in circulating strains. Therefore, the identification of new molecules as potential therapeutics for leishmaniasis is of great relevance. Here, we developed a murine model of L. panamensis infection and evaluated the effect of a new compound in vivo. After treatment of animals with the compound, we observed a significant reduction of inflammation and parasite load at the inoculation site, in a dose-dependent manner. We observed a reduction in IL-10 production by popliteal lymph nodes cells of infected mice. These results pave the way for future evaluation of this compound as a potential antileishmanial drug or as a suitable scaffold for lead optimization strategies.
